Cellular O-Glycome Reporter/Amplification to explore O-glycans of living cells
Protein O-glycosylation has key roles in many biological processes, but the repertoire of O-glycans synthesized by cells is difficult to determine. Here we describe an approach termed Cellular O-Glycome Reporter/Amplification (CORA), a sensitive method used to amplify and profile mucin-type O-glycans synthesized by living cells. Cells convert added peracetylated benzyl-α-N-acetylgalactosamine to a large variety of modified O-glycan derivatives that are secreted from cells, allowing for easy purification for analysis by HPLC and mass spectrometry (MS). Relative to conventional O-glycan analyses, CORA resulted in an âŒ100-1,000-fold increase in sensitivity and identified a more complex repertoire of O-glycans in more than a dozen cell types from Homo sapiens and Mus musculus. Furthermore, when coupled with computational modeling, CORA can be used for predictions about the diversity of the human O-glycome and offers new opportunities to identify novel glycan biomarkers for human diseases
The Spectrum of Strings on Warped AdS_3 x S^3
String theory on NS-NS AdS_3 x S^3 admits an exactly marginal deformation
which breaks the SL(2,R)_R x SL(2,R)_L isometry of AdS_3 down to SL(2,R)_R x
U(1)_L. The holographic dual is an exotic and only partially understood type of
two-dimensional CFT with a reduced unbroken global conformal symmetry group. In
this paper we study the deformed theory on the string worldsheet. It is found
to be related by a spectral flow which is nonlocal in spacetime to the
undeformed worldsheet theory. An exact formula for the spectrum of massive
strings is presented.Comment: 26 pages, no figure
Contribution of the cyclic nucleotide gated channel subunit, CNG-3, to olfactory plasticity in Caenorhabditis elegans.
In Caenorhabditis elegans, the AWC neurons are thought to deploy a cGMP signaling cascade in the detection of and response to AWC sensed odors. Prolonged exposure to an AWC sensed odor in the absence of food leads to reversible decreases in the animal's attraction to that odor. This adaptation exhibits two stages referred to as short-term and long-term adaptation. Previously, the protein kinase G (PKG), EGL-4/PKG-1, was shown necessary for both stages of adaptation and phosphorylation of its target, the beta-type cyclic nucleotide gated (CNG) channel subunit, TAX-2, was implicated in the short term stage. Here we uncover a novel role for the CNG channel subunit, CNG-3, in short term adaptation. We demonstrate that CNG-3 is required in the AWC for adaptation to short (thirty minute) exposures of odor, and contains a candidate PKG phosphorylation site required to tune odor sensitivity. We also provide in vivo data suggesting that CNG-3 forms a complex with both TAX-2 and TAX-4 CNG channel subunits in AWC. Finally, we examine the physiology of different CNG channel subunit combinations
Network meta-analysis-highly attractive but more methodological research is needed
Network meta-analysis, in the context of a systematic review, is a meta-analysis in which multiple treatments (that is, three or more) are being compared using both direct comparisons of interventions within randomized controlled trials and indirect comparisons across trials based on a common comparator. To ensure validity of findings from network meta-analyses, the systematic review must be designed rigorously and conducted carefully. Aspects of designing and conducting a systematic review for network meta-analysis include defining the review question, specifying eligibility criteria, searching for and selecting studies, assessing risk of bias and quality of evidence, conducting a network meta-analysis, interpreting and reporting findings. This commentary summarizes the methodologic challenges and research opportunities for network meta-analysis relevant to each aspect of the systematic review process based on discussions at a network meta-analysis methodology meeting we hosted in May 2010 at the Johns Hopkins Bloomberg School of Public Health. Since this commentary reflects the discussion at that meeting, it is not intended to provide an overview of the field
Sorghum cobalt analysis on not determined wave length with atomic absorption spectrophotometer on background correction mode
This study was to know the better wave length on measuring cobalt content in forage sorghum hybrid (Sorghum bicolor) with an atomic absorption spectrophotometer. The analysis was on background correction mode with three wave lengths; 240.8, 240.7 (determined wave length or recommended wave length) and 240.6 nm, respectively. The larger absorbance value on the 240.7 nm, apparently, it might be considered as a good wave length but the smaller background value was a more important factor for the analysis as was shown on 240.6 nm. Correlation coefficients between the values on 240.7 nm: 240.6 nm and between them (240.8 nm: 240.6 nm) were higher and this common 240.6 nm was considered the better wave length.Key words: Atomic absorption spectrophotometer; background correction mode, cobalt analysis, forage sorghum, not determined wave lengths
Digit-only sauropod pes trackways from China - evidence of swimming or a preservational phenomenon?
For more than 70 years unusual sauropod trackways have played a pivotal role in debates about the swimming ability of sauropods. Most claims that sauropods could swim have been based on manus-only or manus-dominated trackways. However none of these incomplete trackways has been entirely convincing, and most have proved to be taphonomic artifacts, either undertracks or the result of differential depth of penetration of manus and pes tracks, but otherwise showed the typical pattern of normal walking trackways. Here we report an assemblage of unusual sauropod tracks from the Lower Cretaceous Hekou Group of Gansu Province, northern China, characterized by the preservation of only the pes claw traces, that we interpret as having been left by walking, not buoyant or swimming, individuals. They are interpreted as the result of animals moving on a soft mud-silt substrate, projecting their claws deeply to register their traces on an underlying sand layer where they gained more grip during progression. Other sauropod walking trackways on the same surface with both pes and manus traces preserved, were probably left earlier on relatively firm substrates that predated the deposition of soft mud and silt . Presently, there is no convincing evidence of swimming sauropods from their trackways, which is not to say that sauropods did not swim at all

Experimental delayed-choice entanglement swapping
Motivated by the question, which kind of physical interactions and processes
are needed for the production of quantum entanglement, Peres has put forward
the radical idea of delayed-choice entanglement swapping. There, entanglement
can be "produced a posteriori, after the entangled particles have been measured
and may no longer exist". In this work we report the first realization of
Peres' gedanken experiment. Using four photons, we can actively delay the
choice of measurement-implemented via a high-speed tunable bipartite state
analyzer and a quantum random number generator-on two of the photons into the
time-like future of the registration of the other two photons. This effectively
projects the two already registered photons onto one definite of two mutually
exclusive quantum states in which either the photons are entangled (quantum
correlations) or separable (classical correlations). This can also be viewed as
"quantum steering into the past"
Late diagnosis of medial condyle fracture of the humerus with rotational displacement in a child
For displaced medial condyle fractures in children, open reduction with internal fixation seems to be most popular treatment method. The major complication of this method is failure to make the proper early diagnosis. Corrective supracondylar humeral osteotomy has been preferred to open reduction and internal fixation for managing malunited fragments. We report a case of a child with nonunion of the medial condyle of the humerus who was subsequently treated successfully with open reduction and internal fixation
On the classical equivalence of monodromy matrices in squashed sigma model
We proceed to study the hybrid integrable structure in two-dimensional
non-linear sigma models with target space three-dimensional squashed spheres. A
quantum affine algebra and a pair of Yangian algebras are realized in the sigma
models and, according to them, there are two descriptions to describe the
classical dynamics 1) the trigonometric description and 2) the rational
description, respectively. For every description, a Lax pair is constructed and
the associated monodromy matrix is also constructed. In this paper we show the
gauge-equivalence of the monodromy matrices in the trigonometric and rational
description under a certain relation between spectral parameters and the
rescalings of sl(2) generators.Comment: 32pages, 3figures, references added, introduction and discussion
