    Předmětem této práce je seznámení se s technikami zpracovávání obrazu, pro účel počítačového vidění. Jsou zde použity funkce z knihovny OpenCv zajišťující vyhledávání rohových bodů. Dále jsou zde navržené algoritmy pro vyhledávání shodných bodů na po sobě jdoucích snímcích videosekvence. V práci jsou navržené dvě techniky pro řešení tohoto porovnávání bodů a v závěru práce jsou shrnuté jejich výsledky. Rovněž jsou zde naznačena možná řešení některých nežádoucích vlastností těchto navržených algoritmů.The goal of this thesis is to get familiar with techniques of image processing for computer vision. Functions from OpenCv library were used for corner point location. Furthermore, algorithms designed for identical point location were also used for location of points in sequence of images represented by a video sequence. In the thesis, two techniques were designed for solving this point comparison. Results of these methods are summarized in the end of thesis. Possible solutions of negative properties of these methods are presented.

V úvodu práce je charakterizována geologická struktura ložiska a následují výpočty náloží pro jednotlivé etáže, možnosti použití různých druhů trhavin a využití moderních technologií a jejich zavedení do procesu prací souvisejících s přípravou a projektováním trhacích prací. Dále je zde obsažen návrh technologického postupu všech činností, kterých je zapotřebí k bezpečnému provedení trhacích prací. Práce je řešena tak, aby byly dodrženy veškeré zákony a vyhlášky, které se týkají trhacích prací.This thesis deals with the updating and modernization of the current general design of the large-scale blasting work at the quarry Skoupý. In the introduction the work presents the characteristics of the geological structure of the deposit, follows with calculations for charges for the individual benches, the possibility for the use of different types of explosives, and the use of modern technologies and their implementation in the process of work connected to the preparation and design of blasting work. The work also contains a plan for the technical process of all activities which are necessary for the safe execution of blasting work.     The theory of functionalism is based on the notion that the cumulative effect of "functional" activities will tend to influence the political aims of governments towards more cooperation and fewer conflicts. This has not been the case in ICAO and it remains to be seen whether technical cooperation can transcend the vested national interests of states in the other aspects of civil aviation. Anything that is non-technical is likely to be political in nature, even when such issues come up before the ICAO Council, under the guise of technical problems. Any expectation that political and functional activities in aviation could be separated were dashed as soon as ICAO joined the United Nations System. As a consequence, it is bound to follow political positions which do not necessarily accord with its mandate and with the requirements of air transport

    Many public transport authorities are looking to how they can enable better mobility for their citizens, in\ua0congested urban settings as well as in poorly connected peripheral communities and rural areas. Mobility as a\ua0Service (MaaS) is increasingly being presented as a possible solution in both contexts, although the path forward\ua0has proven far more difficult than hoped. Part of this may lie in underestimating the complexities of achieving\ua0integrated mobility services, which is often linked to a lack of practical experience in implementing and running\ua0MaaS services. To quote one project partner in a MaaS pilot project, who represented a public organisation\ua0new to MaaS but with long experience within one transport silo, and who shall remain nameless: “This was a\ua0lot harder than we expected”. So, what should one expect? Why has the MaaS vision not naturally coalesced?\ua0The purpose of this first discussion paper in the series is to try to understand the bigger picture of MaaS\ua0primarily from the user perspective but in an interplay with the service perspective, as the service offer and\ua0design inherently affect the use of the service. Questions include: What is MaaS? and What could it be?; Who\ua0are the (potential) users? and What do they stand to gain?; and How can related research and tools be utilised\ua0by public authorities and transport organisations to more proactively engage in MaaS developments so as to\ua0improve sustainability outcomes (ecological, economic and social) for users, public and private organisations,\ua0and society

    Student Perspectives on the Presence and Usefulness of Navigational Course Elements in Distance Education Courses

    The Quality Matters Standards for Course Design (2018) rubric is a course development rubric for online courses in higher education. General Standard 1 provides a list of specific review standards that are integral in the development of courses, including course expectations, course structure, learning guidance, communication expectations, technology skill requirements, criteria for student assessment, and overall course organization. Student feedback is necessary in order to determine whether students view elements of the course overview and introduction as present in their courses, and whether those elements are useful for the successful completion of their online course. This research utilized a mixed-methods approach, with a survey being sent to all students enrolled in an online degree program at a midsized regional university during the Spring 2021 term. The results of this study indicated students found all elements of Quality Matters Standard 1 to be somewhat to mostly always present in their courses. Study findings indicate students perceive all elements to be somewhat to mostly useful in their successful completion of an online course. These findings indicate students find General Standard 1 rubric elements as useful in the successful completion of an online course

    Allostery and Applications of the Lac Repressor

    The lac repressor has been extensively studied for nearly half a century; this long and complicated experimental history leaves many subtle connections unexplored. This thesis sought to forge those connections from isolated and purified components up to functioning lac genetic switches in cells and even organisms. We first connected the genetics and structure of the lac repressor to in vivo gene regulation in Escherichia coli. We found that point mutations of amino acids that structurally make specific contacts with DNA can alter repressor-operator DNA affinity and even the conformational equilibrium of the repressor. We then found that point mutations of amino acids that structurally make specific contacts with effector molecules can alter repressor-effector affinity and the conformational equilibrium. All results are well explained by a Monod, Wyman, and Changeux model of allostery. We next connected purified in vitro components with in vivo gene regulation in E. coli. We used an in vitro transcription assay to measure repressor-operator DNA binding affinity, repressor-effector binding affinity, and conformational equilibrium. Only the repressor-operator DNA binding affinity disagreed with literature values from other in vitro experiments, however it did agree with a published value which should hold under in vivo conditions. We were able to use our in vitro thermodynamic parameters to accurately predict the in vivo gene regulation when cell crowding was considered. Finally we developed an autogenously regulated lac repressor for AAV-mediated gene therapy. We were able to improve the gene regulation of the autogenous switch by using multiple operator DNA sites, a tetrameric lac repressor, and point mutations to the lac repressor. The autogenous switch was shown to function in various cell types and was capable of reversible regulation of luciferase in living mice
