Three-dimensional plotted scaffolds with controlled pore size gradients : effect of scaffold geometry on mechanical performance and cell seeding efficiency
Scaffolds produced by rapid prototyping (RP) techniques have proved their value for tissue engineering
applications, due to their ability to produce predetermined forms and structures featuring fully interconnected
pore architectures. Nevertheless, low cell seeding efficiency and non-uniform distribution of cells
remain major limitations when using such types of scaffold. This can be mainly attributed to the inadequate
pore architecture of scaffolds produced by RP and the limited efficiency of cell seeding techniques
normally adopted. In this study we aimed at producing scaffolds with pore size gradients to enhance cell
seeding efficiency and control the spatial organization of cells within the scaffold. Scaffolds based on
blends of starch with poly(e-caprolactone) featuring both homogeneously spaced pores (based on pore
sizes of 0.75 and 0.1 mm) and pore size gradients (based on pore sizes of 0.1–0.75–0.1 and 0.75–0.1–
0.75 mm) were designed and produced by three-dimensional plotting. The mechanical performance of
the scaffolds was characterized using dynamic mechanical analysis (DMA) and conventional compression
testing under wet conditions and subsequently characterized using scanning electron microscopy and
micro-computed tomography. Osteoblast-like cells were seeded onto such scaffolds to investigate cell
seeding efficiency and the ability to control the zonal distribution of cells upon seeding. Scaffolds featuring
continuous pore size gradients were originally produced. These scaffolds were shown to have intermediate
mechanical and morphological properties compared with homogenous pore size scaffolds. The
pore size gradient scaffolds improved seeding efficiency from !35% in homogeneous scaffolds to !70%
under static culture conditions. Fluorescence images of cross-sections of the scaffolds revealed that scaffolds
with pore size gradients induce a more homogeneous distribution of cells within the scaffold.This work was supported by the European NoE EXPERTISSUES (NMP3-CT-2004-500283)

Moisture sorption isotherms and shelf life evaluation of pinhão (Araucaria angustifolia) flour.
In this work moisture sorption isotherms and shelf life of pinhão flour were evaluated at 20 ±1 °C using static gravimetric methods in an aw range of 0.011 to 1.0. Pinhão endosperm, after convective drying at 40 oC, 50 oC and 60 °C, was milled to a finely ground powder (dp<1mm) and packed in metallized PET, vacuum sealed and stored in a low atmospheric relative humidity at 25 °C. The sorption data, fitted by GAB model, regardeless of drying temperature showed the characteristics of type III isotherm. Evaluation of self-absorption of manganese emission lines in Laser Induced Breakdown Spectroscopy measurements
This paper is part of a more general study aimed to the determination of the best experimental procedures for reliable quantitative measurements of Fe-Mn alloys by LIBS. In this work, attention is pointed on the self-absorption processes, whose effect deeply influences the LIBS measurements, reflecting in non-linear calibration curves. The effect of self-absorption on the line intensity can be quantified by defining a self absorption coefficient, that measures the deviation of the line intensity from the linear extrapolation of the curve of growth in the optically thin regime. The authors demonstrated in a previous paper that self absorption coefficients could be calculated once the electron density of the plasma is known and the Stark coefficients of the lines are available. However, when the Stark coefficients of the lines of interest are not known, a different approach is needed. In this work a new method for evaluation of self absorption coefficients in LIBS measurements is presented, which does not require the knowledge of Stark coefficients. In order to understand the basic principles and setting out the theoretical tools that will be used for the analysis o f the alloys, a preliminary study was done on pure Mn; LIBS spectra were acquired in different experimental conditions, at different laser energies and different delays after the laser irradiation o f the sample. Moreover, collinear double pulse measurements were also performed. Analytical relations were derived and experimental procedures devised for evaluation o f the self absorption coefficients of several Mn lines, which are important for characterization and control of the experimental conditions in which the analysis is performed

Forage cactus as an additive in corn without the cob silages of feedlot sheep diets.
This study aimed to assess the impact of adding forage cactus as an additive to the production of corn silage without the cob on the performance of feedlot sheep and subsequent silage losses. The experimental design was completely randomized, consisting of three treatments: corn silage without cob; 0% = 100% corn plant without the cob; 10% = 90% corn plant without cob + 10% forage cactus; 20% = 80% corn plant without cob + 20% forage cactus. Significant effects were observed for dry matter intake (P = 0.0201), organic matter (P = 0.0152), ether extract (P = 0.0001), non-fiber carbohydrates (P = 0.0007). Notably, nutrient digestibility showed significant differences in organic matter (P = 0.0187), ether extract (P = 0.0095), neutral detergent fiber (P = 0.0005), non-fiber carbohydrates (P = 0.0001), and metabolizable energy (P = 0.0001). Performance variables, including total weight gain (P = 0.0148), average daily weight gain (P = 0.0148), feeding efficiency, and rumination efficiency of dry matter (P = 0.0113), also exhibited significant effects. Consequently, it is recommended to include 20% forage cactus in corn silage, which, based on natural matter, helps meet animals’ water needs through feed. This inclusion is especially vital in semi-arid regions and aids in reducing silage losses during post-opening silo disposal

Publisher Copyright: Copyright © 2024 Ersoy et al.Methicillin-resistant Staphylococcus aureus (MRSA) strains are a major challenge for clinicians due, in part, to their resistance to most β-lactams, the first-line treatment for methicillin-susceptible S. aureus. A phenotype termed “NaHCO3-responsiveness” has been identified, wherein many clinical MRSA isolates are rendered susceptible to standard-of-care β-lactams in the presence of physiologically relevant concentrations of NaHCO3, in vitro and ex vivo; moreover, such “NaHCO3-responsive” isolates can be effectively cleared by β-lactams from target tissues in experimental infective endocarditis (IE). One mechanistic impact of NaHCO3 exposure on NaHCO3responsive MRSA is to repress WTA synthesis. This NaHCO3 effect mimics the phenotype of tarO-deficient MRSA, including sensitization to the PBP2-targeting β-lactam, cefuroxime (CFX). Herein, we further investigated the impacts of NaHCO3 exposure on CFX susceptibility in the presence and absence of a WTA synthesis inhibitor, ticlopidine (TCP), in a collection of clinical MRSA isolates from skin and soft tissue infections (SSTI) and bloodstream infections (BSI). NaHCO3 and/or TCP enhanced susceptibility to CFX in vitro, by both minimum inhibitor concentration (MIC) and time-kill assays, as well as in an ex vivo simulated endocarditis vegetations (SEV) model, in NaHCO3-responsive MRSA. Furthermore, in experimental IE (presumably in the presence of endogenous NaHCO3), pre-exposure to TCP prior to infection sensitized the NaHCO3-responsive MRSA strain (but not the non-responsive strain) to enhanced clearances by CFX in target tissues. These data support the notion that NaHCO3 is acting similarly to WTA synthesis inhibitors, and that such inhibitors have potential translational applications in the treatment of certain MRSA strains in conjunction with specific β-lactam agents.publishersversionpublishe
