    Cellular proteostasis during influenza A virus infection: friend or foe?

    In order to efficiently replicate, viruses require precise interactions with host components and often hijack the host cellular machinery for their own benefit. Several mechanisms involved in protein synthesis and processing are strongly affected and manipulated by viral infections. A better understanding of the interplay between viruses and their host-cell machinery will likely contribute to the development of novel antiviral strategies. Here, we discuss the current knowledge on the interactions between influenza A virus (IAV), the causative agent for most of the annual respiratory epidemics in humans, and the host cellular proteostasis machinery during infection. We focus on the manipulative capacity of this virus to usurp the cellular protein processing mechanisms and further review the protein quality control mechanisms in the cytosol and in the endoplasmic reticulum that are affected by this virus.publishe

    As potencialidades do pensamento visual no ensino de marketing de serviços: um estudo de caso sobre a experiência de aprendizagem dos estudantes

    O pensamento visual é uma metodologia que utiliza um conjunto de ferramentas para externalizar os processos internos de pensamento, tornando-os mais claros, explícitos e acionáveis. Em termos do potencial das estratégias de pensamento visual para o ensino e educação, a literatura demonstra que podem ajudar os estudantes a compreender informações complexas, melhorar as competências de pensamento crítico, desenvolver a literacia visual e proporcionar uma experiência de aprendizagem muito mais memorável. O racional deste trabalho é a adequação potencial e natural entre pensamento visual e Marketing de Serviços. Os serviços são dominados por elementos intangíveis e, como resultado, não podem ser facilmente visualizados e compreendidos. Ao mesmo tempo, essa intangibilidade levanta vários desafios para os profissionais de marketing, como a abstração, a generalidade, a não-pesquisabilidade e a impalpabilidade mental. Apesar do potencial do pensamento visual no contexto da educação e ensino de marketing de serviços, a literatura académica sobre essa interseção é surpreendentemente escassa. Através de uma metodologia de estudo de caso, e com o foco na experiência de aprendizagem dos estudantes, este trabalho demonstra os benefícios e oportunidades do pensamento visual no contexto do ensino de marketing de serviços.publishe

    The nuclear levels of thioredoxin reductase 1, gamma-H2AX, and yap are modulated by primary cilia in response to high glucose levels

H&TRC authors gratefully acknowledge the FCT/MCTES national support through the UIDB/05608/2020 and UIDP/05608/2020.Diabetes is a condition characterized by impaired regulation of blood glucose levels, leading to various complications such as hypertension, cardiovascular disease, and retinopathy. Diabetic retinopathy (DR), caused by a disrupted retinal blood barrier, is associated with oxidative stress resulting from dysregulated glucose levels in the retina. The primary cilium, an organelle involved in energy balance and glucose homeostasis, has been implicated in the development of various diseases known as ciliopathies, which include overlapping phenotypes such as obesity, diabetes, and retinopathy. This study aims to investigate the impact of high glucose levels on primary cilia assembly in retinal pigment epithelium (RPE-1) cell cultures and explore the role of cilia in the cellular response to high glucose levels. RPE-1 cells were grown in media supplemented with different glucose concentrations (5 mM, 25 mM, and 5 mM glucose + 20 mM mannitol), and cilia assembly was induced before or after glucose supplementation. The results revealed that glucose supplementation did not affect the number of ciliated cells, but cells supplemented with 25 mM glucose exhibited shorter cilia. To understand the role of cilia in response to high glucose levels, the nuclear levels of thioredoxin reductase 1 (TRXR1), a key enzyme involved in combating oxidative stress triggered by hyperglycemia, were evaluated. Additionally, γH2AX, a marker of DNA breaks and cellular senescence, and YAP, a Hippo pathway effector, were examined. It was observed that glucose supplementation, particularly at high levels (25 mM), influenced the nuclear levels of TRXR1, γH2AX, and YAP. Notably, the presence of cilia modulated the cellular response to high glucose levels, modulating the levels of these proteins. These preliminary findings indicate that primary cilia significantly influence the cellular response to high glucose concentrations, which are known to induce oxidative stress and potentially contribute to the development of DR.info:eu-repo/semantics/publishedVersio

    O trabalho objetiva analisar possibilidades de soluções metodológicas e conceituais comuns e particulares da Arquivologia, Biblioteconomia e Museologia para o estudo e as práticas da preservação de documentos. Compreende a preservação como um campo científico a partir de Bourdieu (2004), no qual se inserem e interagem algumas disciplinas científicas, como as supracitadas. A partir desta perspectiva, observaremos as relações de integração, interseção e as suas respectivas particularidades disciplinares para com a preservação em arquivos, bibliotecas e museus. Tem como objeto parte da literatura produzida no campo da preservação, especialmente algumas obras de referência sobre conservação preventiva e gestão de riscos, a saber: Michalski (2004), Castro (2008), Froner e Rosado (2008), Hollós e Pedersoli Júnior (2009), Pedersoli Júnior (2010), Spinelli e Pedersoli Júnior (2010), Ribeiro (2012), Tavares (2014), Zúñiga (2015), IBERMUSEUS e ICCROM (2017), Bojanoski (2018) e Fundação Oswaldo Cruz (2020).     The role of temperature on the impact of remediated water towards marine organisms

    Marine organisms are frequently exposed to pollutants, including trace metals, derived from natural and anthropogenic activities. In order to prevent environmental pollution, di erent approaches have been applied to remove pollutants from waste water and avoid their discharge into aquatic systems. However, organisms in their natural aquatic environments are also exposed to physico-chemical changes derived from climate change-related factors, including temperature increase. According to recent studies, warming has a negative impact on marine wildlife, with known e ects on organisms physiological and biochemical performance. Recently, a material based on graphene oxide (GO) functionalized with polyethyleneimine (PEI) proved to be e ective in the remediation of mercury (Hg) contaminated water. Nevertheless, no information is available on the toxic impacts of such remediated water towards aquatic systems, neither under actual nor predicted temperature conditions. For this, the present study assessed the toxicity of seawater, previously contaminated with Hg and remediated by GO-PEI, using the clam species Ruditapes philippinarum exposed to actual and a predicted temperature conditions. The results obtained demonstrated that seawater contaminated with Hg and/or Hg+GO-PEI induced higher toxicity in clams exposed to 17 and 22 C compared to organisms exposed to remediated seawater at the same temperatures. Moreover, similar histological and biochemical results were observed between organisms exposed to control and remediated seawater, independently of the temperatures (17 and 21 C), highlighting the potential use of GO-PEI to remediate Hg from seawater without significant toxicity issues to the selected marine species.publishe

    Oxidative stress, metabolic and histopathological alterations in mussels exposed to remediated seawater by GO-PEI after contamination with mercury

    The modern technology brought new engineering materials (e.g. nanostructured materials) with advantageous characteristics such as a high capacity to decontaminate water from pollutants (for example metal(loid)s). Among those innovative materials the synthesis of nanostructured materials (NSMs) based on graphene as graphene oxide (GO) functionalized with polyethyleneimine (GO-PEI) had a great success due to their metal removal capacity from water. However, research dedicated to environmental risks related to the application of these materials is still non-existent. To evaluate the impacts of such potential stressors, benthic species can be a good model as they are affected by several environmental constraints. Particularly, the mussel Mytilus galloprovincialis has been identified by several authors as a bioindicator that responds quickly to environmental disturbances, with a wide spatial distribution and economic relevance. Thus, the present study aimed to evaluate the impacts caused in M. galloprovincialis by seawater previously contaminated by Hg and decontaminated using GO-PEI. For this, histopathological and biochemical alterations were examined. This study demonstrated that mussels exposed to the contaminant (Hg), the decontaminant (GO-PEI) and the combination of both (Hg + GO-PEI) presented an increment of histopathological, oxidative stress and metabolic alterations if compared to organisms under remediated seawater and control conditions The present findings highlight the possibility to remediate seawater with nanoparticles for environmental safety purposes.publishe

    Rubinstein – Taybi in Syndrome a Peruvian patient with mild mental retardation: A case report / Síndrome de Rubinstein - Taybi em um paciente Peruano com leve retardo mental: Um relato de caso

    Rubisntein-Taybi syndrome is of genetic origin, caused by a mutation in the CREBBP gene, which undergoes classic phenotypic changes and changes in neuropsychomotor development. In this sense, this article aims to report the case of a Peruvian patient with the clinical and genotypic diagnosis of the disease. During the prenatal period, the fetus did not have alterations that would suggest the syndrome, but at birth, it had a hemangioma, but without clinical repercussion. In the first three months of life, its development was normal, but from the fourth month on, delays were noticed. At 6 months of age, he was not holding his head, so the parents looked for a geneticist who first made the clinical diagnosis and then requested the karyotype, which confirmed the diagnosis. With this, he already started with multidisciplinary treatment and was still placed to study in regular schools in order to improve his development. Currently, at the age of 15, the patient is in treatment with a multidisciplinary approach and in a school to include children with some neurological weakness in the social environment, in order to avoid bullying from other colleagues. He presents phenotypic alterations characteristic of the syndrome, but the cognitive delay is mild when compared to other adolescents with the same pathology. Therefore, this work shows the need for diagnosis and a multidisciplinary approach in treatment. Even so, more studies are needed to learn more about this syndrome and thus provide a better safeguard of the lives of the patients, as occurred in the case of this patient

    Rett Syndrome in a Peruvian patient: A case report / Síndrome de Rett em um paciente peruano: Um relato de caso

    Rett syndrome is of genetic origin, caused by a mutation of dominant characteristic in the MeCP2 gene, which studies with psychomotor alterations in children. In this sense, this article aims to report the case of a Peruvian patient with the late diagnosis of the disease. The patient started the clinical characteristics of the disease at one year and two months, but the diagnosis, with a genetic study that identified the mutation, was only made at 3 years. During this period her diagnoses were: absence epilepsy and microcephaly. These misconceptions favored the accentuation of biopsychomotor delays. Currently, at the age of 4, the patient is being treated with a multidisciplinary approach, but as there are serious sequelae, there are delays in development and results take time to appear. Therefore, this work shows the difficulty of diagnosis, the need for a multidisciplinary approach in treatment and that there is a need for more studies to learn more about this syndrome and thus be able to provide a better safeguard of the lives of the patients
