    Reliability and Validity of Ultrasound Cross Sectional Area Measurements for Long-Duration Spaceflight

    Limb muscle atrophy and the accompanying decline in function can adversely affect the performance of astronauts during mission-related activities and upon re-ambulation in a gravitational environment. Previous characterization of space flight-induced muscle atrophy has been performed using pre and post flight magnetic resonance imaging (MRI). In addition to being costly and time consuming, MRI is an impractical methodology for assessing in-flight changes in muscle size. Given the mobility of ultrasound (US) equipment, it may be more feasible to evaluate changes in muscle size using this technique. PURPOSE: To examine the reliability and validity of using a customized template to acquire panoramic ultrasound (US) images for determining quadriceps and gastrocnemius anatomical cross sectional area (CSA). METHODS: Vastus lateralis (VL), rectus femoris (RF), medial gastrocnemius (MG), and lateral gastrocnemius (LG) CSA were assessed in 10 healthy individuals (36+/-2 yrs) using US and MRI. Panoramic US images were acquired by 2 sonographers using a customized template placed on the thigh and calf and analyzed by the same 2 sonographers (CX50 Philips). MRI images of the leg were acquired while subjects were supine in a 1.5T scanner (Signa Horizon LX, General Electric) and were analyzed by 3 trained investigators. The average of the 2 US and 3 MRI values were used for validity analysis. RESULTS: High inter-experimenter reliability was found for both the US template and MRI analysis as coefficients of variation across muscles ranged from 2.4 to 4.1% and 2.8 to 3.8%, respectively. Significant correlations were found between US and MRI CSA measures (VL, r = 0.85; RF, r = 0.60; MG, r = 0.86; LG, r = 0.73; p < 0.05). Furthermore, the standard error of measurement between US and MRI ranged from 0.91 to 2.09 sq cm with high limits of agreement analyzed by Bland-Altman plots. However, there were significant differences between absolute values of MRI and US for all muscles. CONCLUSION: The present results indicate that utilizing a customized US template provides reliable measures of leg muscle CSA, and thus could be used to characterize changes in muscle CSA both in flight and on the ground

    The Alliance for System Safety of UAS through Research Excellence (ASSURE) conducted research focused on minimum pilot procedures and operational practices used by unmanned aircraft systems (UAS) operators today for the purpose of developing recommendations. This research recommends four pilot and 46 operational minimum procedures to operate a civil single-engine, fixed-wing, single-pilot-configured UAS flying in beyond visual line-of-sight (BVLOS) conditions. These recommendations are anticipated to support potential future aircrew procedure requirements for UAS larger than 55 lbs. operating in the National Airspace System (NAS). These procedures were validated using representative Control Stations in simulated environments

    Opposing brain differences in 16p11.2 deletion and duplication carriers

    Deletions and duplications of the recurrent ∼600 kb chromosomal BP4–BP5 region of 16p11.2 are associated with a broad variety of neurodevelopmental outcomes including autism spectrum disorder. A clue to the pathogenesis of the copy number variant (CNV)'s effect on the brain is that the deletion is associated with a head size increase, whereas the duplication is associated with a decrease. Here we analyzed brain structure in a clinically ascertained group of human deletion (N = 25) and duplication (N = 17) carriers from the Simons Variation in Individuals Project compared with age-matched controls (N = 29 and 33, respectively). Multiple brain measures showed increased size in deletion carriers and reduced size in duplication carriers. The effects spanned global measures of intracranial volume, brain size, compartmental measures of gray matter and white matter, subcortical structures, and the cerebellum. Quantitatively, the largest effect was on the thalamus, but the collective results suggest a pervasive rather than a selective effect on the brain. Detailed analysis of cortical gray matter revealed that cortical surface area displays a strong dose-dependent effect of CNV (deletion > control > duplication), whereas average cortical thickness is less affected. These results suggest that the CNV may exert its opposing influences through mechanisms that influence early stages of embryonic brain development

    ABSTRACT A dry-low-NOx, high-aimow-capacity fuel injection system for a leanpremixed combustor has been developed for a moderate pressure ratio (20:l) aeroderivative gas turbine engine. Engine requirements for cornbustor pressure drop, emissions, and operability have been met. Combustion performance was evaluated at high power conditions in a high-pressure, single-nozzle test facility which operates at full baseload conditions. Single digit NOx levels and high combustion efficiency were achieved A wide operability range with no signs of flashback, autoignition. or thermal problems was demonsuated. NOx sensitivities 10 pressure and residence time were found to be small at flame temperatures below 1850 K (2870 D. Above 1850 K some NOx sensitivity to pressure and residence Lime was observed and was associated with the increased role of the thermal NOx production mechanism at elevated flame temperatures

    Physical drivers facilitating a toxigenic cyanobacterial bloom in a major Great Lakes tributary

    The Maumee River is the primary source for nutrients fueling seasonal Microcystis-dominated blooms in western Lake Erie\u27s open waters though such blooms in the river are infrequent. The river also serves as source water for multiple public water systems and a large food services facility in northwest Ohio. On 20 September 2017, an unprecedented bloom was reported in the Maumee River estuary within the Toledo metropolitan area, which triggered a recreational water advisory. Here we (1) explore physical drivers likely contributing to the bloom\u27s occurrence, and (2) describe the toxin concentration and bacterioplankton taxonomic composition. A historical analysis using 10-years of seasonal river discharge, water level, and local wind data identified two instances when high-retention conditions occurred over ≥ 10 d in the Maumee River estuary: in 2016 and during the 2017 bloom. Observation by remote sensing imagery supported the advection of cyanobacterial cells into the estuary from the lake during 2017 and the lack of an estuary bloom in 2016 due to a weak cyanobacterial bloom in the lake. A rapid-response survey during the 2017 bloom determined levels of the cyanotoxins, specifically microcystins, in excess of recreational contact limits at sites within the lower 20 km of the river while amplicon sequencing found these sites were dominated by Microcystis. These results highlight the need to broaden our understanding of physical drivers of cyanobacterial blooms within the interface between riverine and lacustrine systems, particularly as such blooms are expected to become more prominent in response to a changing climate
