    Response of micro-algae in the Kromme Estuary to managed freshwater inputs

    The Kromme is a permanently open estuary that receives little freshwater input because the capacity of the dams is equivalent to the mean annual runoff from the catchment. The estuary is marine dominated and phytoplankton chlorophyll a is low because of the low freshwater pulses that introduce nutrient-rich freshwater. Water released (2 x 106 m3) from the Mpofu Dam in 1998 produced little micro-algal response in the estuary. The study reported here addresses further runoff scenarios to see which might be beneficial in stimulating microalgal production. Recent surveys together with past research were used to describe the present state and reference condition of the estuary. Average intertidal chlorophyll a was 12.9 ± 2.5 μg·g-1 of sediment and 4.9 ± 0.4 μg·g-1 of sediment during November 2003 and July 2004. These concentrations are relatively low but comparable to those found in intertidal sediments in other South African estuaries and might indicate that intertidal microalgal biomass is not severely limited by low freshwater inputs. Average water column chlorophyll a concentrations have ranged from 0.6 ± 0.1 to 5.6 ± 0.3 μg·ℓ-1. Present state conditions can thus be described as those where water column chlorophyll a seldom exceeds 5 μg·ℓ-1 and small flagellates (3.5 μm x 2.8 μm) dominate the phytoplankton. The diatoms introduced via freshwater have been lost. Under reference conditions before the Mpofu Dam was built, baseflow would have been greater than 1 m3·s-1 for approximately 8 months of the year. The flocculation of fine particles associated with the mixing of fresh and saline waters would have resulted in phytoplankton peaks (chl a >10 μg·ℓ-1) in the middle reaches of the estuary. A more suitable habitat might also have been present for the epipelic (mud associated) benthic microalgae. An assessment of the future runoff scenarios indicated that the most beneficial for the microalgae would be a flow release from the Mpofu Dam of 5 x 106 m3 in October and then again in January. This would stimulate a 25 to 33% increase in phytoplankton chlorophyll a and a doubling in intertidal benthic chlorophyll a for a period of two months following the releases.     The response of microalgal biomass and community composition to environmental factors in the Sundays Estuary

    The Sundays Estuary is permanently open to the sea and experiences regular freshwater inflow in the form of agricultural return flows with large supplies of nutrients. The objectives of this study were to measure microalgal biomass and community composition and relate these to freshwater inflow, water quality and other environmental variables. These data can then be used in setting the ecological water requirements of the estuary. Surveys in August 2006, March 2007, February, June and August 2008 showed that salinity less than 10, expressed in practical salinity units, mostly occurred from 12.5 km from the mouth in the middle reaches of the estuary, which was also where the highest water column chlorophyll a (>20 ƒÊgE.-1) was found. The study showed that different groups of microalgae formed phytoplankton blooms during individual samplingsessions. These included blooms of green algae (August 2006), flagellates (March 2007), dinoflagellates (June 2008) and diatom species (February and August 2008). The estuary was then sampled over 5 consecutive weeks from March to April 2009 to identify environmental factors that support different microalgal bloom species. Phytoplankton blooms werefound during Weeks 1, 4 and 5 from the middle to the upper reaches of the estuary. It was shown that diatoms occurred in blooms during warm, calm conditions whereas wind-mixing and reduced temperature, as a result of a cold front during 17 to 19 March 2009, promoted the dominance of flagellates throughout the estuary although they were present at all times.Dominant diatom species (Cylindrotheca closterium, Cyclotella atomus and Cyclostephanus dubius) indicated brackish, nutrient-rich water. Nanoplankton (2.7 - 20 ƒÊm) was dominant during each week sampled and contributed 55 - 79% to the phytoplankton biomass. Maximum benthic chlorophyll a was found 12.5 km from the mouth. This study is the first to show successive chlorophyll a blooms consisting of different phytoplankton groups in an estuary, an indication of the eutrophicstate of the system

    The decomposition of estuarine macrophytes under different temperature regimes

    The Great Brak Estuary is a temporarily open/closed system situated along the warm temperate coast of the Western Cape, South Africa. The estuary is subject to a variety of anthropogenic impacts (e.g. freshwater abstraction and sewage discharge) that increases its susceptibility to prolonged  periods of mouth closure, eutrophication, and ultimately the formation of macroalgal blooms. The aim of this study was to determine the  decomposition characteristics of the most dominant submerged  macrophyte and macroalgal species in the Great Brak Estuary. Laboratory experiments were conducted to determine the effect of different  temperature regimes on the rate of decomposition of 3 macrophyte  species and the extent of inorganic nutrients released. The results  demonstrated that anaerobic decomposition of Zostera capensis, Ruppia  cirrhosa, and Cladophora glomerata resulted in high levels of inorganic  nutrient release over the 28-day study period. Ammonium (NH4+) was the dominant form of dissolved inorganic nitrogen (DIN) released during the decomposition process for all three species. The low levels of total  oxidised nitrogen (nitrate and nitrite) released during decomposition were attributed to the inhibition of nitrification by heterotrophic bacteria under anoxic conditions. The relative levels of dissolved inorganic phosphorus (DIP) released were lower than that observed for DIN, and peaked early on in the experimental period (ca. 7 days), thereafter stabilising or  decreasing. The DIP levels were explained, in part, by the varying nutrient requirements and limitations of each species (e.g. nitrogen-limited). The release of inorganic nutrients was greatest at higher temperatures (i.e. 25°C and 30°C), due to the reduced bacterial activity experienced at lower temperatures (i.e. 15°C). Ultimately, estuarine health deteriorates during macroalgal blooms, and therefore it is important to implement mitigation measures, such as artificial mouth breaching and plant harvesting, in order to minimise or reverse the effects of eutrophication.Keywords: Great Brak Estuary, decomposition, temperature, inorganic nutrient release, Cladophora glomerata, Zostera capensis, Ruppia cirrhos

    Interactions and potential implications of Plasmodium falciparum-hookworm coinfection in different age groups in south-central Côte d'Ivoire

    BACKGROUND: Given the widespread distribution of Plasmodium and helminth infections, and similarities of ecological requirements for disease transmission, coinfection is a common phenomenon in sub-Saharan Africa and elsewhere in the tropics. Interactions of Plasmodium falciparum and soil-transmitted helminths, including immunological responses and clinical outcomes of the host, need further scientific inquiry. Understanding the complex interactions between these parasitic infections is of public health relevance considering that control measures targeting malaria and helminthiases are going to scale.METHODOLOGY: A cross-sectional survey was carried out in April 2010 in infants, young school-aged children, and young non-pregnant women in south-central Côte d'Ivoire. Stool, urine, and blood samples were collected and subjected to standardized, quality-controlled methods. Soil-transmitted helminth infections were identified and quantified in stool. Finger-prick blood samples were used to determine Plasmodium spp. infection, parasitemia, and hemoglobin concentrations. Iron, vitamin A, riboflavin, and inflammation status were measured in venous blood samples.PRINCIPAL FINDINGS: Multivariate regression analysis revealed specific association between infection and demographic, socioeconomic, host inflammatory and nutritional factors. Non-pregnant women infected with P. falciparum had significantly lower odds of hookworm infection, whilst a significant positive association was found between both parasitic infections in 6- to 8-year-old children. Coinfected children had lower odds of anemia and iron deficiency than their counterparts infected with P. falciparum alone.CONCLUSIONS/SIGNIFICANCE: Our findings suggest that interaction between P. falciparum and light-intensity hookworm infections vary with age and, in school-aged children, may benefit the host through preventing iron deficiency anemia. This observation warrants additional investigation to elucidate the mechanisms and consequences of coinfections, as this information could have important implications when implementing integrated control measures against malaria and helminthiases

    Observation of Coherent Elastic Neutrino-Nucleus Scattering

    The coherent elastic scattering of neutrinos off nuclei has eluded detection for four decades, even though its predicted cross-section is the largest by far of all low-energy neutrino couplings. This mode of interaction provides new opportunities to study neutrino properties, and leads to a miniaturization of detector size, with potential technological applications. We observe this process at a 6.7-sigma confidence level, using a low-background, 14.6-kg CsI[Na] scintillator exposed to the neutrino emissions from the Spallation Neutron Source (SNS) at Oak Ridge National Laboratory. Characteristic signatures in energy and time, predicted by the Standard Model for this process, are observed in high signal-to-background conditions. Improved constraints on non-standard neutrino interactions with quarks are derived from this initial dataset

    West Nile virus vector Culex modestus established in southern England

    Background: The risk posed to the United Kingdom by West Nile virus (WNV) has previously been considered low, due to the absence or scarcity of the main Culex sp. bridge vectors. The mosquito Culex modestus is widespread in southern Europe, where it acts as the principle bridge vector of WNV. This species was not previously thought to be present in the United Kingdom. Findings: Mosquito larval surveys carried out in 2010 identified substantial populations of Cx. modestus at two sites in marshland in southeast England. Host-seeking-adult traps placed at a third site indicate that the relative seasonal abundance of Cx. modestus peaks in early August. DNA barcoding of these specimens from the United Kingdom and material from southern France confirmed the morphological identification. Conclusions: Cx. modestus appears to be established in the North Kent Marshes, possibly as the result of a recent introduction. The addition of this species to the United Kingdom’s mosquito fauna may increase the risk posed to the United Kingdom by WNV

    Fanconi anaemia with bilateral diffuse pulmonary arterio venous fistulae: a case report

    <p>Abstract</p> <p>Background</p> <p>We report a patient with cytogenetically confirmed Fanconi anaemia with associated diffuse bilateral pulmonary arterio-venous fistulae. This is only the second reported case of diffuse pulmonary arterio-venous fistulae with Fanconi anaemia.</p> <p>Case Presentation</p> <p>A 16 year old Sri Lankan boy, with a cytogenetically confirmed Fanconi anaemia was admitted to University Medical Unit, National Hospital of Sri Lanka for further assessment and treatment. Both central and peripheral cyanosis plus clubbing were noted on examination. The peripheral saturation was persistently low on room air and did not improve with supplementary Oxygen. Contrast echocardiography failed to demonstrate an intra cardiac shunt but showed early crossover of contrast, suggesting the possibility of pulmonary arterio-venous fistulae. Computed tomography pulmonary angiogram was inconclusive. Subsequent right heart catheterisation revealed bilateral diffuse arterio-venous fistulae not amenable for device closure or surgical intervention.</p> <p>Conclusion</p> <p>To our knowledge, this is the second reported patient with diffuse pulmonary arterio-venous fistulae associated with Fanconi anaemia. We report this case to create awareness among clinicians regarding this elusive association. We recommend screening patients with Fanconi anaemia using contrast echocardiography at the time of assessment with transthoracic echocardiogram. Though universal screening may be impossible given the cost constraints, such screening should at least be performed in patients with clinical evidence of desaturation or when a therapeutic option such as haematopoietic stem cell transplantation is considered. Treatment of pulmonary arteriovenous fistulae would improve patient outcome as desaturation by shunting worsens the anaemic symptoms by reducing the oxygen carrying capacity of blood.</p

    In Situ Photodegradation of Incorporated Polyanion Does Not Alter Prion Infectivity

    Single-stranded polyanions ≥40 bases in length facilitate the formation of hamster scrapie prions in vitro, and polyanions co-localize with PrPSc aggregates in vivo [1], [2]. To test the hypothesis that intact polyanionic molecules might serve as a structural backbone essential for maintaining the infectious conformation(s) of PrPSc, we produced synthetic prions using a photocleavable, 100-base oligonucleotide (PC-oligo). In serial Protein Misfolding Cyclic Amplification (sPMCA) reactions using purified PrPC substrate, PC-oligo was incorporated into physical complexes with PrPSc molecules that were resistant to benzonase digestion. Exposure of these nuclease-resistant prion complexes to long wave ultraviolet light (315 nm) induced degradation of PC-oligo into 5 base fragments. Light-induced photolysis of incorporated PC-oligo did not alter the infectivity of in vitro-generated prions, as determined by bioassay in hamsters and brain homogenate sPMCA assays. Neuropathological analysis also revealed no significant differences in the neurotropism of prions containing intact versus degraded PC-oligo. These results show that polyanions >5 bases in length are not required for maintaining the infectious properties of in vitro-generated scrapie prions, and indicate that such properties are maintained either by short polyanion remnants, other co-purified cofactors, or by PrPSc molecules alone

    Familial Linkage between Neuropsychiatric Disorders and Intellectual Interests

    From personality to neuropsychiatric disorders, individual differences in brain function are known to have a strong heritable component. Here we report that between close relatives, a variety of neuropsychiatric disorders covary strongly with intellectual interests. We surveyed an entire class of high-functioning young adults at an elite university for prospective major, familial incidence of neuropsychiatric disorders, and demographic and attitudinal questions. Students aspiring to technical majors (science/mathematics/engineering) were more likely than other students to report a sibling with an autism spectrum disorder (p = 0.037). Conversely, students interested in the humanities were more likely to report a family member with major depressive disorder (p = 8.8×10−4), bipolar disorder (p = 0.027), or substance abuse problems (p = 1.9×10−6). A combined PREdisposition for Subject MattEr (PRESUME) score based on these disorders was strongly predictive of subject matter interests (p = 9.6×10−8). Our results suggest that shared genetic (and perhaps environmental) factors may both predispose for heritable neuropsychiatric disorders and influence the development of intellectual interests
