    Who is more likely to feel ostracized? A latent class analysis of personality traits

    Perceived ostracism (e.g., feeling ignored or excluded) is a painful and distressing experience. However, little empirical research has investigated the types (profiles) of people more likely to perceive ostracism. The present study (N = 395) used latent class analysis to (a) identify potential classes based on the big five personality traits (i.e., openness, agreeableness, negative emotionality, extroversion, and conscientiousness) and (b) examine whether such classes could reliably differentiate levels of self-reported perceived ostracism. We extracted three classes: (a) Moderate Traits (MT), (b) the Quiet Over-Reacting Procrastinators (QORP), and (c) the Active and Adaptable Thinkers (AAT). Those in the QORP class reported the highest levels of perceived ostracism, whereas those in the AAT class reported the lowest levels of perceived ostracism compared to the MT class. This study provides new insight into the profiles of individuals who may be more likely to perceive ostracism. However, further research is needed to explore the association between personality and ostracism (e.g., as ostracism may lead to changes in personality), so that potential risk markers to trigger early psychological interventions of such ostracized individuals can be identified

    Distinct sulfur saturation histories within the Palaeogene Magilligan Sill, Northern Ireland: Implications for Ni-Cu-PGE mineralisation in the North Atlantic Igneous Province

The final version is available from NRC Research Press via the DOI in this record.The ~60 m thick Magilligan Sill is part of the British Palaeogene Igneous Province in the North Atlantic. The sill comprises layers of dolerite and olivine gabbro, and it intrudes a thick sequence of Mesozoic mudstones and marls, which are locally baked at the sill margins. Since 2014, the sill has been an exploration target for orthomagmatic Ni-Cu-PGE sulfide mineralisation analogous to the Noril’sk-Talnakh intrusion in Russia. We present new petrological, geochemical and S-isotope data to assess the prospectivity of the sill and the underlying magmatic plumbing system. Most sulfides in the dolerite portions of the sill are < 50 μm in size and comprise only pyrite with PGE abundances below detection limit. In the olivine gabbros, > 150 μm size pentlandite, chalcopyrite and pyrrhotite grains contain < 4 ppm total PGE, 1 460 ppm Co and 88 ppm Ag. Pyrite from the dolerites have δ34S ranging from -10.0 to +3.4 ‰ and olivine gabbro sulfides range from -2.5 to -1.1 ‰, suggesting widespread crustal contamination. The S/Se ratios of sulfides in the dolerites and olivine gabbros range from 3 500 to 19 500 and from 1 970 to 3 710, respectively, indicating that the latter may have come from upstream in the magma plumbing system. The Magilligan Sill records multiple injections of mafic magma into an inflating sill package, each with distinct mechanisms towards S-saturation. Whilst the sulfide minerals in the sill do not constitute significant mineralisation themselves, detailed in-situ studies highlight a divergence in Ssaturation histories, and suggest that a larger volume of olivine gabbro sulfides at depth may be prospective

    On the Emergence of Tremor in Prodromal Parkinson's Disease

    Clinical, neuropathological and neuroimaging research suggests that pathological changes in Parkinson’s disease (PD) start many years before the emergence of motor signs. Since disease-modifying treatments are likely to be most effective when initiated early in the disease process, there has been significant interest in characterizing prodromal PD. Some people with PD describe autonomic symptoms at the time of diagnosis suggesting that autonomic dysfunction is a common feature of prodromal PD. Furthermore, subtle motor signs may be present and emerge prior to the time of diagnosis. We present a series of patients who, in the prodromal phase of PD, experienced the emergence of tremor initially only while yawning or straining at stool and discuss how early involvement of autonomic brainstem nuclei could lead to these previously unreported phenomena. The hypothalamic paraventricular nucleus (PVN) plays a central role in autonomic control including bowel/bladder function, cardiovascular homeostasis and yawning and innervates multiple brainstem nuclei involved in autonomic functions (including brainstem reticular formation, locus ceruleus, dorsal raphe nucleus and motor nucleus of the vagus). The PVN is affected in PD and evidence from related phenomena suggest that the PVN could increase tremor either by increasing downstream cholinergic activity on brainstem nuclei such as the reticular formation or by stimulating the locus ceruleus to activate the cerebellothalamocortical network via the ventrolateral nucleus of the thalamus. Aberrant cholinergic/noradrenergic transmission between these brainstem nuclei early in PD couldlead to tremor before the emergence of other parkinsonian signs, representing an early clinical clue to prodromal PD

    Detection of Gene Expression in an Individual Cell Type within a Cell Mixture Using Microarray Analysis

    BACKGROUND: A central issue in the design of microarray-based analysis of global gene expression is the choice between using cells of single type and a mixture of cells. This study quantified the proportion of lipopolysaccharide (LPS) induced differentially expressed monocyte genes that could be measured in peripheral blood mononuclear cells (PBMC), and determined the extent to which gene expression in the non-monocyte cell fraction diluted or obscured fold changes that could be detected in the cell mixture. METHODOLOGY/PRINCIPAL FINDINGS: Human PBMC were stimulated with LPS, and monocytes were then isolated by positive (Mono+) or negative (Mono-) selection. The non-monocyte cell fraction (MonoD) remaining after positive selection of monocytes was used to determine the effect of non-monocyte cells on overall expression. RNA from LPS-stimulated PBMC, Mono+, Mono- and MonoD samples was co-hybridised with unstimulated RNA for each cell type on oligonucleotide microarrays. There was a positive correlation in gene expression between PBMC and both Mono+ (0.77) and Mono- (0.61-0.67) samples. Analysis of individual genes that were differentially expressed in Mono+ and Mono- samples showed that the ability to detect expression of some genes was similar when analysing PBMC, but for others, differential expression was either not detected or changed in the opposite direction. As a result of the dilutional or obscuring effect of gene expression in non-monocyte cells, overall about half of the statistically significant LPS-induced changes in gene expression in monocytes were not detected in PBMC. However, 97% of genes with a four fold or greater change in expression in monocytes after LPS stimulation, and almost all (96-100%) of the top 100 most differentially expressed monocyte genes were detected in PBMC. CONCLUSIONS/SIGNIFICANCE: The effect of non-responding cells in a mixture dilutes or obscures the detection of subtle changes in gene expression in an individual cell type. However, for studies in which only the most highly differentially expressed genes are of interest, separating and analysing individual cell types may be unnecessary

    Technical Variability Is Greater than Biological Variability in a Microarray Experiment but Both Are Outweighed by Changes Induced by Stimulation

    INTRODUCTION: A central issue in the design of microarray-based analysis of global gene expression is that variability resulting from experimental processes may obscure changes resulting from the effect being investigated. This study quantified the variability in gene expression at each level of a typical in vitro stimulation experiment using human peripheral blood mononuclear cells (PBMC). The primary objective was to determine the magnitude of biological and technical variability relative to the effect being investigated, namely gene expression changes resulting from stimulation with lipopolysaccharide (LPS). METHODS AND RESULTS: Human PBMC were stimulated in vitro with LPS, with replication at 5 levels: 5 subjects each on 2 separate days with technical replication of LPS stimulation, amplification and hybridisation. RNA from samples stimulated with LPS and unstimulated samples were hybridised against common reference RNA on oligonucleotide microarrays. There was a closer correlation in gene expression between replicate hybridisations (0.86-0.93) than between different subjects (0.66-0.78). Deconstruction of the variability at each level of the experimental process showed that technical variability (standard deviation (SD) 0.16) was greater than biological variability (SD 0.06), although both were low (SD<0.1 for all individual components). There was variability in gene expression both at baseline and after stimulation with LPS and proportion of cell subsets in PBMC was likely partly responsible for this. However, gene expression changes after stimulation with LPS were much greater than the variability from any source, either individually or combined. CONCLUSIONS: Variability in gene expression was very low and likely to improve further as technical advances are made. The finding that stimulation with LPS has a markedly greater effect on gene expression than the degree of variability provides confidence that microarray-based studies can be used to detect changes in gene expression of biological interest in infectious diseases
