Hydrocarbon and halocarbon measurements as photochemical and dynamical indicators of atmospheric hydroxyl, atomic chlorine, and vertical mixing obtained during Lagrangian flights
Nonmethane hydrocarbons and halocarbons were measured during two Lagrangian experiments conducted in the lower troposphere of the North Atlantic as part of the June 1992, Atlantic Stratosphere Transition Experiment/Marine Aerosol and Gas Exchange (ASTEX/MAGE) expedition. The first experiment was performed in very clean marine air. Meteorological observations indicate that the height of the marine boundary layer rose rapidly, entraining free tropospheric air. However, the free tropospheric and marine boundary layer halocarbon concentrations were too similar to allow this entrainment to be quantified by these measurements. The second Lagrangian experiment took place along the concentration gradient of an aged continental air mass advecting from Europe. The trace gas measurements confirm that the National Center for Atmospheric Research (NCAR) Electra aircraft successfully intercepted the same air mass on consecutive days. Two layers, a surface layer and a mixed layer with chemically distinct compositions, were present within the marine boundary layer. The composition of the free troposphere was very different from that of the mixed layer, making entrainment from the free troposphere evident Concentrations of the nonmethane hydrocarbons in the Lagrangian surface layer were observed to become depleted relative to the longer-lived tetrachloroethene. A best fit to the observations was calculated using various combinations of the three parameters, loss by reaction with hydroxyl, loss by reaction with chlorine, and/or dilution from the mixed layer. These calculations provided estimated average concentrations in the surface layer for a 5-hour period from dawn to 11 UT of 0.3±0.5 ×106 molecules cm-3 for HO, and 3.3±;1.1 ×104 molecules cm-3 for Cl. Noontime concentration estimates were 2.6±0.7 ×106 molecules cm-3 for HO and 6.5±1.4 ×104 molecules cm-3 for Cl. Copyright 1996 by the American Geophysical Union
MILCS: A mutual information learning classifier system
This paper introduces a new variety of learning classifier system (LCS), called MILCS, which utilizes mutual information as fitness feedback. Unlike most LCSs, MILCS is specifically designed for supervised learning. MILCS's design draws on an analogy to the structural learning approach of cascade correlation networks. We present preliminary results, and contrast them to results from XCS. We discuss the explanatory power of the resulting rule sets, and introduce a new technique for visualizing explanatory power. Final comments include future directions for this research, including investigations in neural networks and other systems. Copyright 2007 ACM
Embryonic Development and a Quantitative Model of Programmed DNA Elimination in Mesocyclops Edax (S. A. ) (Copepoda: Cyclopoida)
The highly programmed fragmentation of chromosomes and elimination of large amounts of nuclear DNA from the presomatic cell lineages (i.e., chromatin diminution), occurs in the embryos of the freshwater zooplankton Mesocyclops edax (S. A. Forbes, 1891) (Crustacea: Copepoda). The somatic genome is reorganized and reduced to a size five times smaller even though the germline genome remains intact. We present the first comprehensive, quantitative model of DNA content throughout embryogenesis in a copepod that possesses embryonic DNA elimination. We used densitometric image analysis to measure the DNA content of polar bodies, germline and somatic nuclei, and excised DNA “droplets.” We report: 1) variable DNA contents of polar bodies, some of which do not contain the amount corresponding to the haploid germline genome size; 2) presence of pronuclei in newly laid embryo sacs; 3) gonomeric chromosomes in the second to fourth cleavage divisions and in the primordial germ cell and primordial endoderm cell during the fifth cleavage division; 4) timing of early embryonic cell stages, elimination of DNA, and divisions of the primordial germ cell and primordial endoderm cell at 22∘C role= presentation \u3e22∘C; and 5) persistence of a portion of the excised DNA “droplets” throughout embryogenesis. DNA elimination is a trait that spans multiple embryonic stages and a knowledge of the timing and variability of the associated cytological events with DNA elimination will promote the study of the molecular mechanisms involved in this trait. We propose the “genome yolk hypothesis” as a functional explanation for the persistence of the eliminated DNA that might serve as a resource during postdiminution cleavage divisions
Ribosomal S6K1 in POMC and AgRP Neurons Regulates Glucose Homeostasis but Not Feeding Behavior in Mice.
SummaryHypothalamic ribosomal S6K1 has been suggested as a point of convergence for hormonal and nutrient signals in the regulation of feeding behavior, bodyweight, and glucose metabolism. However, the long-term effects of manipulating hypothalamic S6K1 signaling on energy homeostasis and the cellular mechanisms underlying these roles are unclear. We therefore inactivated S6K1 in pro-opiomelanocortin (POMC) and agouti-related protein (AgRP) neurons, key regulators of energy homeostasis, but in contrast to the current view, we found no evidence that S6K1 regulates food intake and bodyweight. In contrast, S6K1 signaling in POMC neurons regulated hepatic glucose production and peripheral lipid metabolism and modulated neuronal excitability. S6K1 signaling in AgRP neurons regulated skeletal muscle insulin sensitivity and was required for glucose sensing by these neurons. Our findings suggest that S6K1 signaling is not a general integrator of energy homeostasis in the mediobasal hypothalamus but has distinct roles in the regulation of glucose homeostasis by POMC and AgRP neurons
Ribbed tiled vaulting: Innovation through two design-build workshops
Traditional tile vaults are typically constructed springing off from walls or straight arches built from support element to support element on falsework. From these, the vault's surface can be built in space with minimal or no guidework. Built on previous research and focusing on continuous surface expression and fully representing three-dimensional equilibrium surfaces in compression, this research explores the design potential of three-dimensional networks of structural ribs, made possible by new funicular form-finding approaches. This new structural typology for tile vaults was investigated and tested through two intensive, design-build workshops in Australia, the first at the University of Technology, Sydney (UTS) in October 2012, and the second at Monash Art Design & Architecture (MADA), in May 2013
Trends in the lifetime risk of developing cancer in Great Britain: comparison of risk for those born from 1930 to 1960
BACKGROUND: Typically, lifetime risk is calculated by the period method using current risks at different ages. Here, we estimate the probability of being diagnosed with cancer for individuals born in a given year, by estimating future risks as the cohort ages. METHODS: We estimated the lifetime risk of cancer in Britain separately for men and women born in each year from 1930 to 1960. We projected rates of all cancers (excluding non-melanoma skin cancer) and of all cancer deaths forwards using a flexible age-period-cohort model and backwards using age-specific extrapolation. The sensitivity of the estimated lifetime risk to the method of projection was explored. RESULTS: The lifetime risk of cancer increased from 38.5% for men born in 1930 to 53.5% for men born in 1960. For women it increased from 36.7 to 47.5%. Results are robust to different models for projections of cancer rates. CONCLUSIONS: The lifetime risk of cancer for people born since 1960 is >50%. Over half of people who are currently adults under the age of 65 years will be diagnosed with cancer at some point in their lifetime
The impact of HIV infection on tuberculosis transmission in a country with low tuberculosis incidence: a national retrospective study using molecular epidemiology
Background: HIV is known to increase the likelihood of reactivation of latent tuberculosis to active TB disease;
however, its impact on tuberculosis infectiousness and consequent transmission is unclear, particularly in lowincidence settings.
Methods: National surveillance data from England, Wales and Northern Ireland on tuberculosis cases in adults from
2010 to 2014, strain typed using 24-locus mycobacterial-interspersed-repetitive-units–variable-number-tandem-repeats
was used retrospectively to identify clusters of tuberculosis cases, subdivided into ‘first’ and ‘subsequent’ cases.
Firstly, we used zero-inflated Poisson regression models to examine the association between HIV status and the
number of subsequent clustered cases (a surrogate for tuberculosis infectiousness) in a strain type cluster. Secondly, we
used logistic regression to examine the association between HIV status and the likelihood of being a subsequent case
in a cluster (a surrogate for recent acquisition of tuberculosis infection) compared to the first case or a non-clustered
case (a surrogate for reactivation of latent infection).
Results: We included 18,864 strain-typed cases, 2238 were the first cases of clusters and 8471 were subsequent cases.
Seven hundred and fifty-nine (4%) were HIV-positive.
Outcome 1: HIV-positive pulmonary tuberculosis cases who were the first in a cluster had fewer subsequent cases
associated with them (mean 0.6, multivariable incidence rate ratio [IRR] 0.75 [0.65–0.86]) than those HIV-negative
(mean 1.1).
Extra-pulmonary tuberculosis (EPTB) cases with HIV were less likely to be the first case in a cluster compared to HIVnegative EPTB cases. EPTB cases who were the first case had a higher mean number of subsequent cases (mean 2.5,
IRR (3.62 [3.12–4.19]) than those HIV-negative (mean 0.6).
Outcome 2: tuberculosis cases with HIV co-infection were less likely to be a subsequent case in a cluster (odds ratio 0.82 [0.69–0.98]), compared to being the first or a non-clustered case.
Conclusions: Outcome 1: pulmonary tuberculosis-HIV patients were less infectious than those without HIV. EPTB
patients with HIV who were the first case in a cluster had a higher number of subsequent cases and thus may be
markers of other undetected cases, discoverable by contact investigations.
Outcome 2: tuberculosis in HIV-positive individuals was more likely due to reactivation than recent infection, compared
to those who were HIV-negative
