    Inter-annual and inter-seasonal variability of the Orkney wave power resource

    The waters surrounding the Orkney archipelago in the north of Scotland are one of the key regions in the world suitable for exploitation of both wave and tidal energy resources. Accordingly, Orkney waters are currently host to 1.08 GW of UK Crown Estate leased wave and tidal energy projects, with a further 0.5 GW leased in the southern part of the adjacent Pentland Firth. Although several wave resource models exist of the region, most of these models are commercial, and hence the results not publicly available, or have insufficient spatial/temporal resolution to accurately quantify the wave power resource of the region. In particular, no study has satisfactorily resolved the inter-annual and inter-seasonal variability of the wave resource around Orkney. Here, the SWAN wave model was run at high resolution on a high performance computing system, quantifying the Orkney wave power resource over a ten year period (2003–2012), a decade which witnessed considerable inter-annual variability in the wave climate. The results of the validated wave model demonstrate that there is considerable variability of the wave resource surrounding Orkney, with an extended winter (December–January–February–March, DJFM) mean wave power ranging from 10 to 25 kW/m over the decade of our study. Further, the results demonstrate that there is considerably less uncertainty (30%) in the high energy region to the west of Orkney during winter months, in contrast to much greater uncertainty (60%) in the lower energy region to the east of Orkney. The DJFM wave resource to the west of Orkney correlated well with the DJFM North Atlantic Oscillation (NAO). Although a longer simulated time period would be required to fully resolve inter-decadal variability, these preliminary results demonstrate that due to considerable inter-annual variability in the NAO, it is important to carefully consider the time period used to quantify the wave power resource of Orkney, or regions with similar exposure to the North Atlantic. Finally, our study reveals that there is significantly less variability in the practical wave power resource, since much of the variability in the theoretical resource is contained within relatively few extreme events, when a wave device enters survival mode

    EEG Signature of Out-of-Body Experiences Induced by Virtual Reality: A Novel Methodological Approach.

    Out-of-body experiences (OBEs) are subjective experiences of seeing one's own body and the environment from a location outside the physical body. They can arise spontaneously or in specific conditions, such as during the intake of dissociative drug. Given its unpredictable occurrence, one way to empirically study it is to induce subjective experiences resembling an OBE using technology such as virtual reality. We employed a complex multisensory method of virtual embodiment in a virtual reality scenario with 7 healthy participants to induce virtual OBE-like experiences. Participants performed 2 conditions in a randomly determined order. For both conditions, the participant's viewpoint was lifted out of the virtual body toward the ceiling of the virtual room, and real body movements were (visuo-tactile ON condition) or were not (visuo-tactile OFF condition) translated into movements on the virtual body below-the latter aiming to maintain a feeling of connection with the virtual body. A continuous 128-electrode EEG was recorded. Participants reported subjective experiences of floating in the air and of feeling high up in the virtual room at a strong intensity, but a weak to moderate feeling of being "out of their body" in both conditions. The EEG analysis revealed that this subjective experience was associated with a power shift that manifested in an increase of delta and a decrease of alpha relative power. A reduction of theta complexity and an increase of beta-2 connectivity were also found. This supports the growing body of evidence revealing a prominent role of delta activity during particular conscious states

    Labor-associated gene expression in the human uterine fundus, lower segment, and cervix

    Background Preterm labor, failure to progress, and postpartum hemorrhage are the common causes of maternal and neonatal mortality or morbidity. All result from defects in the complex mechanisms controlling labor, which coordinate changes in the uterine fundus, lower segment, and cervix. We aimed to assess labor-associated gene expression profiles in these functionally distinct areas of the human uterus by using microarrays. Methods and Findings Samples of uterine fundus, lower segment, and cervix were obtained from patients at term (mean +/- 6 SD = 39.1 +/- 0.5 wk) prior to the onset of labor (n = 6), or in active phase of labor with spontaneous onset (n = 7). Expression of 12,626 genes was evaluated using microarrays ( Human Genome U95A; Affymetrix) and compared between labor and non-labor samples. Genes with the largest labor-associated change and the lowest variability in expression are likely to be fundamental for parturition, so gene expression was ranked accordingly. From 500 genes with the highest rank we identified genes with similar expression profiles using two independent clustering techniques. Sets of genes with a probability of chance grouping by both techniques less than 0.01 represented 71.2%, 81.8%, and 79.8% of the 500 genes in the fundus, lower segment, and cervix, respectively. We identified 14, 14, and 12 those sets of genes in the fundus, lower segment, and cervix, respectively. This enabled networks of coregulated and co-expressed genes to be discovered. Many genes within the same cluster shared similar functions or had functions pertinent to the process of labor. Conclusions Our results provide support for many of the established processes of parturition and also describe novel-to-labor genes not previously associated with this process. The elucidation of these mechanisms likely to be fundamental for controlling labor is an important prerequisite to the development of effective treatments for major obstetric problems - including prematurity, with its long-term consequences to the health of mother and offspring

    Optimised insert design for improved single-molecule imaging and quantification through CRISPR-Cas9 mediated knock-in

The use of CRISPR-Cas9 genome editing to introduce endogenously expressed tags has the potential to address a number of the classical limitations of single molecule localisation microscopy. In this work we present the first systematic comparison of inserts introduced through CRISPR-knock in, with the aim of optimising this approach for single molecule imaging. We show that more highly monomeric and codon optimised variants of mEos result in improved expression at the TubA1B locus, despite the use of identical guides, homology templates, and selection strategies. We apply this approach to target the G protein-coupled receptor (GPCR) CXCR4 and show a further insert dependent effect on expression and protein function. Finally, we show that compared to over-expressed CXCR4, endogenously labelled samples allow for accurate single molecule quantification on ligand treatment. This suggests that despite the complications evident in CRISPR mediated labelling, the development of CRISPR-PALM has substantial quantitative benefits

    Optical mapping as a routine tool for bacterial genome sequence finishing

    Background: In sequencing the genomes of two Xenorhabdus species, we encountered a large number of sequence repeats and assembly anomalies that stalled finishing efforts. This included a stretch of about 12 Kb that is over 99.9% identical between the plasmid and chromosome of X. nematophila. Results: Whole genome restriction maps of the sequenced strains were produced through optical mapping technology. These maps allowed rapid resolution of sequence assembly problems, permitted closing of the genome, and allowed correction of a large inversion in a genome assembly that we had considered finished. Conclusion: Our experience suggests that routine use of optical mapping in bacterial genome sequence finishing is warranted. When combined with data produced through 454 sequencing, an optical map can rapidly and inexpensively generate an ordered and oriented set of contigs to produce a nearly complete genome sequence assembly
