    Erratic Male Meiosis Resulting in 2n Pollen Grain Formation in a 4x Cytotype (2n = 28) of Ranunculus laetus Wall. ex Royle

    Two accessions were studied for male meiosis in Ranunculus laetus from the cold regions of Northwest Himalayas. One accession showed the presence of 14 bivalents at diakinesis and regular segregation of bivalents at anaphase I which lead to normal tetrad formation with four n microspores and consequently n pollen grains and 100% pollen fertility. Second accession from the same locality revealed the erratic meiosis characterized by the presence of all the 28 chromosomes as univalents in meiocytes at metaphase I. Univalent chromosomes failed to segregate during anaphases and produced restitution nuclei at meiosis I and II. These restitution nuclei resulted into dyads and triads which subsequently produced two types of apparently fertile pollen grains. On the basis of size, the two types of pollen grains were categorized as n (normal reduced) and 2n (unreduced, 1.5-times larger than the n pollen grains). The estimated frequency of 2n pollen grains from dyads and triads (61.59%) was almost the same as that of the observed one (59.90%), which indicated that 2n pollen grains in R. laetus were the result of dyads and triads. The present paper herein may provide an insight into the mechanisms of the formation of various intraspecific polyploids through sexual polyploidization in R. laetus

    USE OF SOME ETHNOMEDICINAL HERBS BY THE NATIVES OF SOLANG VALLEY, KULLU DISTRICT, HIMACHAL PRADESH

    Objective: The objective of the study was to enlist the ethnobotanical uses of wild plants of Solang Valley, Kullu District, Himachal Pardesh. Due to the specific geographical location of the valley, it possesses a high amount of endemic plant diversity. The plants are being used medically by local people against many ailments such as rheumatism, gastric disorders, muscular pain, asthma, dysentery, diabetes, constipation, cold, cough, fever, etc.Methods: For documentation of ethnomedicinal information, a questionnaire containing vernacular name, plant part/s used, medicinal uses, method of preparation and amount of dose taken was prepared. To gather such information, personal interviews/interactions were conducted with medicine men (vaids and hakims), local healers, village elders, tribals and shepherds.Results: Presently, 42 plant species belonging to 41 genera and 24 families are included for ethnobotanical study with respect to their scientific names, vernacular names, plant parts used, ethnomedicinal purposes, mode of preparation, and dose/amount taken. As many as 23 species are used for treating more than one ailment. By consulting the previous literature, it was found that there are 19 species which have not been reported earlier and are used by the natives for different medicinal purposes.Conclusion: The unsustainable harvesting such as uprooting of whole plant of medicinal use from the wild is resulting into a serious decline in plant populations. Therefore various cultivation techniques should be designed, and implemented especially for the highly medicinally important and endangered plant species. Grazing should be restricted in high altitudinal zones possessing high endemic plant diversity. Further, information gathered on ethnobotanical aspects of plants of medicinal use will be helpful in the selection of elite genotypes/chemotypes which could provide a base for future plantation programmes which will be helpful for sustainable development of the valley

    Objective: The objective of the present study was to enlist the ethnobotanical uses and chromosomal status of wild plants of Parvati Valley, Himachal Pradesh. Plants are locally used in curing ulcers, sores, insect bites, muscular and joint pains, pneumonia, jaundice, diarrhoea, piles, swellings, allergies and skin diseases.Methods: For documentation of ethnomedicinal information, a questionnaire containing the vernacular name, plant part/s used, medicinal uses, mode of preparation and amount of dose taken was prepared. Personnel interviews/interactions were conducted with medicine men (vaids and hakims), local healers, village elders, tribals and shepherds. The interviews were cross-validated to ascertain the facts about the local use of each plant species. Chromosomal status was determined through male meiosis by using standard acetocarmine technique.Results: Present paper contains information on ethnomedicinal uses, chromosome counts, male meiosis and pollen fertility on 62 medicinal herbs from Parvati Valley. Plants are used as a decoction, paste or powder in cooked form or as raw. Preparations are taken orally in pure form or as a mixture or with a little amount of salt, milk, honey or butter.Conclusion: Due to indiscriminate forest clearing, grazing and collection by traders, most of the species became rare and restricted to specific pockets or are now almost extinct. To preserve herbal diversity, grazing should be restricted in alpine and sub-alpine zones. Database on ethnic knowledge, chromosomal diversity, natural regeneration and distribution pattern should be prepared for designing future plans for sustainable development

    Preparation and <em>in vitro</em> Characterisation of Solid Dispersion Floating Tablet by Effervescent Control Release Technique with Improved Floating Capabilities

    In this research, an effort has been done for the development of effervescent controlled release floating tablet (ECRFT) from solid dispersions (SDs) of diclofenac sodium (DS) for upsurge the solubility and dissolution rate. ECRFT of DS was prepared by using SDs of DS and its SDs prepared with PEG as carrier using thermal method (simple fusion). SDs of DS was formulated in many ratios (1:1, 1:2, 1:3 and 1:4). Prepared SDs were optimised for its solubility, % drug content and % dissolution studies. Tablets were formulated by using optimised SDs products and all formulation was evaluated for various parameters. A clear rise in dissolution rate was detected with entirely SD, amid that the optimised SD (SD4) was considered for ECRFT. Among all the tablet formulations, its F3 formulation was better in all the terms of pre-compression and post-compression parameters. It had all the qualities of a good ECRFT, based on this F3 formulation was selected as the best formulation. Data of in vitro release were fitted in several kinetics models to explain release mechanism. The F3 formulation shows zero order release. From this study, we can conclude that ECRFT containing SDs of DS can be successfully used for achieving better therapeutic objective

    Chromosome count, meiotic abnormalities and pollen sterility in Lahaul sweetvetch (Hedysarum astragaloides Benth. ex Baker, Fabaceae), an endemic and threatened species from India

    Male meiotic studies were carried out on eight different accessions of Hedysarum astragaloides Benth. ex Baker (Fabaceae), an endemic and threatened species of northwest Himalaya, India. Although genetic factors such as meiosis, chromosome number, and ploidy level may be causative for the evolution, endemism, rare distribution or even extinction of the species, no detailed information exists. Keeping this in mind H. astragaloides has been studied cytologically. Male meiotic investigations revealed diploid level (2n=2x=14) for species and normal meiotic course in the accessions from the Manali Hills resulting in nearly 100% pollen fertility. However, the accessions scored from the Manimahesh Hills and Pangi Valley depicted inter-pollen mother cell transfer of chromatin material and structural heterozygosity for reciprocal translocations. Consequent upon these meiotic anomalies, some pollen sterility (21%) resulted. On account of this sweeping genetic outcome, the incidence of anomalies such as this in an endemic and threatened species warrants grave consideration. It is sensible to conclude that conservation measures should include the collection of germplasm from the localities where plants are meiotically stable with high gametic fertility, to ensure good germination and healthy plants for future use. Seeds from meiotically normal individuals should be given priority for inclusion in seed banks

    Potential Role of Plant Growth Regulators in Administering Crucial Processes Against Abiotic Stresses

    Plant growth regulators are naturally biosynthesized chemicals in plants that influence physiological processes. Their synthetic analogous trigger numerous biochemical and physiological processes involved in the growth and development of plants. Nowadays, due to changing climatic scenario, numerous biotic and abiotic stresses hamper seed germination, seedling growth, and plant development leading to a decline in biological and economic yields. However, plant growth regulators (PGRs) can potentially play a fundamental role in regulating plant responses to various abiotic stresses and hence, contribute to plant adaptation under adverse environments. The major effects of abiotic stresses are growth and yield disturbance, and both these effects are directly overseen by the PGRs. Different types of PGRs such as abscisic acid (ABA), salicylic acid (SA), ethylene (ET), and jasmonates (JAs) are connected to boosting the response of plants to multiple stresses. In contrast, PGRs including cytokinins (CKs), gibberellins (GAs), auxin, and relatively novel PGRs such as strigolactones (SLs), and brassinosteroids (BRs) are involved in plant growth and development under normal and stressful environmental conditions. Besides, polyamines and nitric oxide (NO), although not considered as phytohormones, have been included in the current review due to their involvement in the regulation of several plant processes and stress responses. These PGRs are crucial for regulating stress adaptation through the modulates physiological, biochemical, and molecular processes and activation of the defense system, upregulating of transcript levels, transcription factors, metabolism genes, and stress proteins at cellular levels. The current review presents an acumen of the recent progress made on different PGRs to improve plant tolerance to abiotic stress such as heat, drought, salinity, and flood.     Science with the Daksha High Energy Transients Mission

    We present the science case for the proposed Daksha high energy transients mission. Daksha will comprise of two satellites covering the entire sky from 1~keV to >1>1~MeV. The primary objectives of the mission are to discover and characterize electromagnetic counterparts to gravitational wave source; and to study Gamma Ray Bursts (GRBs). Daksha is a versatile all-sky monitor that can address a wide variety of science cases. With its broadband spectral response, high sensitivity, and continuous all-sky coverage, it will discover fainter and rarer sources than any other existing or proposed mission. Daksha can make key strides in GRB research with polarization studies, prompt soft spectroscopy, and fine time-resolved spectral studies. Daksha will provide continuous monitoring of X-ray pulsars. It will detect magnetar outbursts and high energy counterparts to Fast Radio Bursts. Using Earth occultation to measure source fluxes, the two satellites together will obtain daily flux measurements of bright hard X-ray sources including active galactic nuclei, X-ray binaries, and slow transients like Novae. Correlation studies between the two satellites can be used to probe primordial black holes through lensing. Daksha will have a set of detectors continuously pointing towards the Sun, providing excellent hard X-ray monitoring data. Closer to home, the high sensitivity and time resolution of Daksha can be leveraged for the characterization of Terrestrial Gamma-ray Flashes.Comment: 19 pages, 7 figures. Submitted to ApJ.     Initial Commitment to Pre-Exposure Prophylaxis and Circumcision for HIV Prevention amongst Indian Truck Drivers

    Studies of HIV prevention interventions such as pre-exposure prophylaxis (PREP) and circumcision in India are limited. The present study sought to investigate Indian truck-drivers initial commitment to PREP and circumcision utilizing the AIDS Risk Reduction Model. Ninety truck-drivers completed an in-depth qualitative interview and provided a blood sample for HIV and HSV-2 testing. Truck-drivers exhibited low levels of initial commitment towards PREP and even lower for circumcision. However, potential leverage points for increasing commitment were realized in fear of infecting family rather than self, self-perceptions of risk, and for PREP focusing on cultural beliefs towards medication and physicians. Cost was a major barrier to both HIV prevention interventions. Despite these barriers, our findings suggest that the ARRM may be useful in identifying several leverage points that may be used by peers, health care providers and public health field workers to enhance initial commitment to novel HIV prevention interventions in India

    Antiinflammatory Therapy with Canakinumab for Atherosclerotic Disease

    Background: Experimental and clinical data suggest that reducing inflammation without affecting lipid levels may reduce the risk of cardiovascular disease. Yet, the inflammatory hypothesis of atherothrombosis has remained unproved. Methods: We conducted a randomized, double-blind trial of canakinumab, a therapeutic monoclonal antibody targeting interleukin-1β, involving 10,061 patients with previous myocardial infarction and a high-sensitivity C-reactive protein level of 2 mg or more per liter. The trial compared three doses of canakinumab (50 mg, 150 mg, and 300 mg, administered subcutaneously every 3 months) with placebo. The primary efficacy end point was nonfatal myocardial infarction, nonfatal stroke, or cardiovascular death. RESULTS: At 48 months, the median reduction from baseline in the high-sensitivity C-reactive protein level was 26 percentage points greater in the group that received the 50-mg dose of canakinumab, 37 percentage points greater in the 150-mg group, and 41 percentage points greater in the 300-mg group than in the placebo group. Canakinumab did not reduce lipid levels from baseline. At a median follow-up of 3.7 years, the incidence rate for the primary end point was 4.50 events per 100 person-years in the placebo group, 4.11 events per 100 person-years in the 50-mg group, 3.86 events per 100 person-years in the 150-mg group, and 3.90 events per 100 person-years in the 300-mg group. The hazard ratios as compared with placebo were as follows: in the 50-mg group, 0.93 (95% confidence interval [CI], 0.80 to 1.07; P = 0.30); in the 150-mg group, 0.85 (95% CI, 0.74 to 0.98; P = 0.021); and in the 300-mg group, 0.86 (95% CI, 0.75 to 0.99; P = 0.031). The 150-mg dose, but not the other doses, met the prespecified multiplicity-adjusted threshold for statistical significance for the primary end point and the secondary end point that additionally included hospitalization for unstable angina that led to urgent revascularization (hazard ratio vs. placebo, 0.83; 95% CI, 0.73 to 0.95; P = 0.005). Canakinumab was associated with a higher incidence of fatal infection than was placebo. There was no significant difference in all-cause mortality (hazard ratio for all canakinumab doses vs. placebo, 0.94; 95% CI, 0.83 to 1.06; P = 0.31). Conclusions: Antiinflammatory therapy targeting the interleukin-1β innate immunity pathway with canakinumab at a dose of 150 mg every 3 months led to a significantly lower rate of recurrent cardiovascular events than placebo, independent of lipid-level lowering.
