    Migration Control in Cloud Computing to Reduce the SLA Violation

    The requisition of cloud based services are more eminent because of the enormous benefits of cloud such as pay-as-you-use flexibility,scalability and low upfront cost. Day-by-day due to growing number of cloud consumers the load on the datacenters is also increasing. Various load distribution and dynamic load balancing approaches are being followed in the datacenters to optimize the resource utilization so that the performance may be maintained during the increased load. Virtual machine (VM) migration is primarily used to implement dynamic load balancing in the datacenters. But, the poorly designed dynamic VM migration policies may negate its benefits. The VM migration overheads result in the violations of service level agreement (SLA) in the cloud environment.In this paper,an extended VM migration control model is proposedto minimize the SLA violations while controlling the energy consumption of the datacenter during VM migration. The parameters of execution boundary threshold is used to extend an existing VM migration control model. The proposed model is tested through extensive simulations using CloudSim toolkit by executing real world workload. Results are obtained in terms of number of SLA violations while controlling the energy consumption in the datacenter. Results show that the proposed modelachieves better performance in comparison to the existing model

    Effects of customer trust and online experiences in building hospitality brands

    Customer trust embodies customer beliefs of actually receiving a promised service and manifestations of consumer’s confidences in an exchange parties reliability and integrity. The study is based on the fact as to how trusts criteria affect online purchase especially in regard to booking and buying the accommodations and also that accommodation providers assume that are very essential for consumers to make the online purchase. In total 150 consumers and 80 hotels owners/operators in India were examined. There are enormous discrepancies between consumers and accommodation providers were searched. Like formal guarantee of providers, security concern, refund of price paid delivery time and information about confirmation and they will switch from one brand to other due to promise breakage, less service quality, high price charged. However, these trust criteria were viewed inconsequential by the accommodation providers. It concluded with vast number of suggestions and recommendations for the accommodation providers need to include in their websites and build reputation and strong brands in the hospitality market

    Efficacy of checkpoint inhibition after CAR-T failure in aggressive B-cell lymphomas: Outcomes from 15 US institutions

    Checkpoint inhibitor (CPI) therapy with anti-PD-1 antibodies has been associated with mixed outcomes in small cohorts of patients with relapsed aggressive B-cell lymphomas after CAR-T failure. To define CPI therapy efficacy more definitively in this population, we retrospectively evaluated clinical outcomes in a large cohort of 96 patients with aggressive B-cell lymphomas receiving CPI therapy after CAR-T failure across 15 US academic centers. Most patients (53%) had diffuse large B-cell lymphoma, were treated with axicabtagene ciloleucel (53%), relapsed early (≤180 days) after CAR-T (83%), and received pembrolizumab (49%) or nivolumab (43%). CPI therapy was associated with an overall response rate of 19% and a complete response rate of 10%. Median duration of response was 221 days. Median progression-free survival (PFS) and overall survival (OS) were 54 and 159 days, respectively. Outcomes to CPI therapy were significantly improved in patients with primary mediastinal B-cell lymphoma. PFS (128 vs 51 days) and OS (387 vs 131 days) were significantly longer in patients with late (\u3e180 days) vs early (≤180 days) relapse after CAR-T. Grade ≥3 adverse events occurred in 19% of patients treated with CPI. Most patients (83%) died, commonly because of progressive disease. Only 5% had durable responses to CPI therapy. In the largest cohort of patients with aggressive B-cell lymphoma treated with CPI therapy after CAR-T relapse, our results reveal poor outcomes, particularly among those relapsing early after CAR-T. In conclusion, CPI therapy is not an effective salvage strategy for most patients after CAR-T, where alternative approaches are needed to improve post-CAR-T outcomes

    Determination of proper gamma radiation dose for creating variation in Indian garlic varieties

    Garlic (Allium sativum L.) an important neutraceutical crop. It is used as traditional medicine to aid digestion, respiration, parasitic infestation and to treat leprosy and various other diseases. But biologically garlic is sterile in nature hence for creating variation; traditional breeding methods are not applicable. Further clonal propagation method leads garlic more prone for various seed and soil borne diseases including virus infection. And natural genetic variation in morphological as well as biochemical traits is limited in garlic. Considering this is in view, among various methods of creating variation, treating bulbs with radiation is simple and easy technique. Main activity of mutation breeding using gamma radiation is to determine optimum dose (LD-50). Hence in present experiment, two garlic varieties were subjected to gamma radiation treatments (0, 1, 5, 10, 25, 50, 75 and 100 gray). Aim of this experiment is to identify LD-50 dose and then generate variation in garlic like increase in bulb size, bulb shape, high neutraceutical compounds like allicin etc. In results, both treated varieties recorded 50 percent germination at dose 10 grays as LD-50. Further on basis of survival of plantlets and final matured plants count, optimum gamma radiation dose of 5 gray for Bhima Omkar and in between 5 to 10 gray for Bhima Purple is identified as LD-50 respectively. In case of Bhima Purple, there is further need to screen the population for exact dose value of LD-50. All other morphological traits in both varieties recorded decreasing trend with increase in radiation doses. However no significant alterations observed visually in any treatment. This optimized radiation dose will be helpful for subjecting other genotypes for getting mutants and generating diversity in garlic which may have high neutracutical compounds and would suitable for processing and export

    Characterization of <em>Allium</em> germplasms for conservation and sustainable management using SSR markers

    193-199Allium species are very important due to their medicinal values. Quercetin and allicin are medicinally important compound of onion and garlic, respectively which are proved useful to treat various diseases. However, highly heterozygous nature, self-incompatibility and long gestation period limits genetic improvement of Allium species. Further, the existing germplasms in Indian subcontinents are largely cultivated ones with poor genetic characterization, which limits the germplasm conservation and future management. A total of thirty polymorphic Simple Sequence Repeats (SSRs) were utilized for characterisation of popular onion germplasms and their cross-transferability revealed relatedness with fifteen garlic and wild relatives. Average number of alleles per SSR locus, PIC and heterozygosity was found to be 3.9, 0.51 and 0.57,respectively.Overall genetic diversity recorded was higher in wild relative compared to cultivated A. cepa, possibly because most of the A. cepa variety is derived by domestication but wild relatives are open pollinated and undergoes extensive gene pool shuffling leading to higher heterogeneity. In this study, SSR markers were successfully utilized to assess genetic variations in popular Indian A. cepa, A. sativum and establish genetic relationships with wild Allium species. These markers can be harnessed for molecular breeding, varietal identification and planning germplasm conservation strategies in future

    Loss of seed viability in onion (Allium cepa L.) in relation to degradation of lipids during storage

    Onion seeds have one of the lowest longevity among the cultivated crops as it loses viability within a year. The unmarketable seeds have no reported food value or feed value. Onion seeds contain considerable oil content which has potential industrial utility. The present study reports the changes in the quality of seed oil extracted from seeds stored for one year (2019-2020), after which the seeds were unmarketable due to low germination. The physico-chemical properties of seed oil were studied in four onion (Allium cepa L.) varieties (Bhima Safed, Bhima Super, Bhima Dark Red and Bhima Shakti) by standard analytical test. The per cent oil content varied from 12.94% - 16.02% and storage resulted in 0.64% loss in seed oil content. The mean seed germination in freshly harvested seeds was 80.50%, which declined to 52.33% after one year of ambient storage. The electrical conductivity, which measures the membrane integrity increased by 108.60% in one-year-old seeds. The physico-chemical properties of the seed oil deteriorated with the age of the seed. In comparison to the control, the acid values and peroxide values of the seed oil increased by 51.11% and 12.90% respectively, while saponification value decreased by 36.18%. Among the varieties, Bhima Dark Red had the highest degradation in the seed storability and vigour, which also had the highest deterioration in the lipid quality. The present study reports for the first time the degradation of seed lipids during ageing in onion which might be contributing to the poor storability of onion seeds.

Capsicum, commonly called chilli, is one such crop with its fruits extensively used as vegetable across the world. Knowledge on various traits is important for genetic improvement of such species. Here, we assessed the genetic diversity among 10 genotypes of six Capsicum species, namely Capsicum annuum, C. chinense, C. chacoense, C. frutescens, C. tovarii and C. galapagoense. C. annuum MS-12 is a genetic male sterile line. We used morphological descriptors and simple-sequence repeat (SSR) molecular markers for this study. Out of 60 SSR screened, 22 markers (36.66%) showed polymorphism. Alleles number per locus varied from 3 to 7. Average PIC value for 22 polymorphic markers was 0.69, and ranged from 0.54 for the primer Hpms 1-139 to 0.85 for the primer CAMS-072. Ten genotypes of Capsicum species were grouped into three major clusters such that genotypes in a single cluster had less dissimilarity matrix values among themselves than which belongs to other clusters. Range of fruit weight and pericarp thickness varied from 0.1 g (‘PAU-621’) to 2.3 g (‘MS-12’), and from 0.29 mm (‘PAU-621’) to1.09 mm (‘MS12’), respectively. These two genotypes can be used in hybridization or in recombinant breeding program for obtaining higher heterotic effects/ heterosis or for transgressive segregants in chilli pepper

    The Effect of Psyllium Husk on Intestinal Microbiota in Constipated Patients and Healthy Controls

    Psyllium is a widely used treatment for constipation. It traps water in the intestine increasing stool water, easing defaecation and altering the colonic environment. We aimed to assess the impact of psyllium on faecal microbiota, whose key role in gut physiology is being increasingly recognised. We performed two randomised, placebo-controlled, double-blinded trials comparing 7 days of psyllium with a placebo (maltodextrin) in 8 healthy volunteers and 16 constipated patients respectively. We measured the patients’ gastrointestnal (GI) transit, faecal water content, short-chain fatty acid (SCFA) and the stool microbiota composition. While psyllium supplement had a small but significant effect on the microbial composition of healthy adults (increasing Veillonella and decreasing Subdoligranulum), in constipated subjects there were greater effects on the microbial composition (increased Lachnospira, Faecalibacterium, Phascolarctobacterium, Veillonella and Sutterella and decreased uncultured Coriobacteria and Christensenella) and alterations in the levels of acetate and propionate. We found several taxa to be associated with altered GI transit, SCFAs and faecal water content in these patients. Significant increases in three genera known to produce butyrate, Lachnospira, Roseburia and Faecalibacterium, correlated with increased faecal water. In summary, psyllium supplementation increased stool water and this was associated with significant changes in microbiota, most marked in constipated patients
