    Molecular Analysis of N6-Methyladenine Patterns in \u3cem\u3eTetrahymena thermophila\u3c/em\u3e Nuclear DNA

    We have cloned two DNA fragments containing 5\u27-GATC-3\u27 sites at which the adenine is methylated in the macronucleus of the ciliate Tetrahymena thermophila. Using these cloned fragments as molecular probes, we analyzed the maintenance of methylation patterns at two partially and two uniformly methylated sites. Our results suggest that a semiconservative copying model for maintenance of methylation is not sufficient to account for the methylation patterns we found during somatic growth of Tetrahymena. Although we detected hemimethylated molecules in macronuclear DNA, they were present in both replicating and nonreplicating DNA. In addition, we observed that a complex methylation pattern including partially methylated sites was maintained during vegetative growth. This required the activity of a methylase capable of recognizing and modifying sites specified by something other than hemimethylation. We suggest that a eucaryotic maintenance methylase may be capable of discriminating between potential methylation sites to ensure the inheritance of methylation patterns

    Diffusion, dimensionality and noise in transcriptional regulation

    The precision of biochemical signaling is limited by randomness in the diffusive arrival of molecules at their targets. For proteins binding to the specific sites on the DNA and regulating transcription, the ability of the proteins to diffuse in one dimension by sliding along the length of the DNA, in addition to their diffusion in bulk solution, would seem to generate a larger target for DNA binding, consequently reducing the noise in the occupancy of the regulatory site. Here we show that this effect is largely cancelled by the enhanced temporal correlations in one dimensional diffusion. With realistic parameters, sliding along DNA has surprisingly little effect on the physical limits to the precision of transcriptional regulation.Comment: 8 pages, 2 figure

    A statistical study of the global structure of the ring current

    [1] In this paper we derive the average configuration of the ring current as a function of the state of the magnetosphere as indicated by the Dst index. We sort magnetic field data from the Combined Release and Radiation Effects Satellite (CRRES) by spatial location and by the Dst index in order to produce magnetic field maps. From these maps we calculate local current systems by taking the curl of the magnetic field. We find both the westward (outer) and the eastward (inner) components of the ring current. We find that the ring current intensity varies linearly with Dst as expected and that the ring current is asymmetric for all Dst values. The azimuthal peak of the ring current is located in the afternoon sector for quiet conditions and near midnight for disturbed conditions. The ring current also moves closer to the Earth during disturbed conditions. We attempt to recreate the Dst index by integrating the magnetic perturbations caused by the ring current. We find that we need to multiply our computed disturbance by a factor of 1.88 ± 0.27 and add an offset of 3.84 ± 4.33 nT in order to get optimal agreement with Dst. When taking into account a tail current contribution of roughly 25%, this agrees well with our expectation of a factor of 1.3 to 1.5 based on a partially conducting Earth. The offset that we have to add does not agree well with an expected offset of approximately 20 nT based on solar wind pressure

    In economics comparative analysis plays the same role as experimental research in physics. In this paper we closely examine several methodological problems related to comparative analysis by investigating the specific example of grain markets in China and France respectively. This enables us to answer a question in economic history which has so far remained pending, namely whether or not market integration progressed in the 18th century. In economics as in physics, before being accepted any new result has to be checked and re-checked by different researchers. This is what we call the replication and comparison procedures.     Topological Speed Limits to Network Synchronization

    We study collective synchronization of pulse-coupled oscillators interacting on asymmetric random networks. We demonstrate that random matrix theory can be used to accurately predict the speed of synchronization in such networks in dependence on the dynamical and network parameters. Furthermore, we show that the speed of synchronization is limited by the network connectivity and stays finite, even if the coupling strength becomes infinite. In addition, our results indicate that synchrony is robust under structural perturbations of the network dynamics.Comment: 5 pages, 3 figure

    Solving Pure Yang Mills in 2+1 Dimensions

    We analytically compute the spectrum of the spin zero glueballs in the planar limit of pure Yang-Mills theory in 2+1 dimensions. The new ingredient is provided by our computation of a new non-trivial form of the ground state wave-functional. The mass spectrum of the theory is determined by the zeroes of Bessel functions, and the agreement with large N lattice data is excellent.Comment: 4 page letter; version to appear in Physical Review Letter

    Heisenberg Hamiltonian description of multiple-sublattice itinerant-electron systems: General considerations and applications to NiMnSb and MnAs

    We consider magnetic systems where the magnetic sublattices can be unambiguously separated into sublattices of inducing and induced moments. The concrete numerical calculations are performed for half-metallic ferromagnetic Heusler compound NiMnSb and hexagonal phase of MnAs. In both systems, Mn atoms possess a robust atomic moment that is much larger than the induced moments of other atoms. It is shown that the treatment of the induced moments as independent variables of the Heisenberg Hamiltonian leads to artificial features in the spin-wave spectrum. We show that the artificial features of the model do not have a dramatic influence on the estimated value of the Curie temperature. This is demonstrated within both mean-field approximation and random-phase approximation. It is shown that the calculational scheme where the induced moments are assumed to fully adjust their values and directions to the adiabatic magnetic configurations of the inducing moments is free from the artificial feature in the spin-wave spectra. In this scheme, the exchange interaction between the inducing and induced moments appears as renormalization of the exchange interactions between inducing moments. It is shown that the redistribution of the exchange interactions has strong influence on the estimated value of the Curie temperature because of the decreased number of the degrees of freedom in the thermodynamic model. Different schemes of the mapping of the systems on the Heisenberg Hamiltonian are examined. The similarities and differences in the properties of NiMnSb and MnAs are discussed

    Solar pond power plant feasibility study for Davis, California

    The feasibility of constructing a solar pond power plant at Davis, California was studied. Site visits, weather data compilation, soil and water analyses, conceptual system design and analyses, a material and equipment market survey, conceptual site layout, and a preliminary cost estimate were studied. It was concluded that a solar pond power plant is technically feasible, but economically unattractive. The relatively small scale of the proposed plant and the high cost of importing salt resulted in a disproportionately high capital investment with respect to the annual energy production capacity of the plant. Cycle optimization and increased plant size would increase the economical attractiveness of the proposed concept
