    Clostridium difficile ribotypes in humans and animals in Brazil

    Clostridium difficile is an emerging enteropathogen responsible for pseudomembranous colitis in humans and diarrhoea in several domestic and wild animal species. Despite its known importance, there are few studies aboutC. difficile polymerase chain reaction (PCR) ribotypes in Brazil and the actual knowledge is restricted to studies on human isolates. The aim of the study was therefore to compare C. difficileribotypes isolated from humans and animals in Brazil. Seventy-six C. difficile strains isolated from humans (n = 25), dogs (n = 23), piglets (n = 12), foals (n = 7), calves (n = 7), one cat, and one manned wolf were distributed into 24 different PCR ribotypes. Among toxigenic strains, PCR ribotypes 014/020 and 106 were the most common, accounting for 14 (18.4%) and eight (10.5%) samples, respectively. Fourteen different PCR ribotypes were detected among human isolates, nine of them have also been identified in at least one animal species. PCR ribotype 027 was not detected, whereas 078 were found only in foals. This data suggests a high diversity of PCR ribotypes in humans and animals in Brazil and support the discussion of C. difficile as a zoonotic pathogen

    Outbreak of Type C Botulism in Backyard Poultry in Santa Catarina, Brazil

    Background: Botulism is a disease caused by the ingestion of neurotoxin produced by Clostridium botulinum, characterized by flaccid paralysis, which can lead to high mortality. They have seven types of neurotoxins (A, B, C, D, E, F, and G) and, in birds, most cases are attributed to type C. They are considered sources of botulinum toxins where the decomposition of organic matter occurs, like stagnant water and rotting food. The main feature of the disease in birds is ascending symmetric flaccid paralysis. The present study aims to describe an outbreak of type C botulism in backyard poultry in the state of Santa Catarina, Southern Brazil.Case: A visit was made to the property with 160 backyard poultry with a history of high mortality in the municipality of Agrolândia, Santa Catarina. Clinical signs were characterized by paralysis of the pelvic limbs, neck and pendular wings, which progressed to death within 48 h. There was a mortality rate of 37.5% (60/160) between March and May 2019. These birds were kept in an overcrowded environment, with different species (chickens, ducks, teals, and turkeys) fed irregularly. The water supplied was provided from kitchen exhaust, accumulating in puddles on the floor that contained organic matter residues such as animal feces, food waste and bone fragments. The disposal of the carcasses of birds that died was in the same enclosure, buried superficially, facilitating the access of other birds to dig them up and consume them. Necropsy was performed on 2 chickens and one duck, no macroscopic or histopathological lesions were observed. Blood, liver, and gastrointestinal content samples were sent for research and identification of botulinum toxin through the serum neutralization test in mice. The presence of type C botulinum toxin was confirmed in the liver chicken of one sampled animals.Discussion: The identification of type C botulism toxin enabled the characterization of the outbreak, which is the toxin most associated with episodes of botulism in birds. It is not always possible to identify the origin of the infection, as intoxication can occur by ingestion of water contaminated with organic waste, however, in this outbreak, as sources of poisoning in birds, it was specified and occurred due to the ingestion of water with organic matter that was stagnant in the floor of the enclosure, and also by ingesting contaminated carcasses present in the area. In subsistence farming, botulism outbreaks are reported with greater frequency in the Northeast and Southeast of Brazil generally, cases in which sanitary conditions and incorrect carcass disposal favor the occurrence of the disease. As noted in the present study, high mortality is a common feature of botulism. The evaluated signs and developed evolution were similar to previous studies, which ranged from 14 to 72 h. The absence of macroscopic and histopathological changes is commonly reported in cases of botulism in domestic animals, since botulinum toxin only causes functional changes, with no tissue damage. The association of clinical signs, epidemiology, post mortem evaluation and detection of type C botulinum toxin concludes the diagnosis of botulism. Avian influenza and Newcastle disease are important diseases that have neurological conditions and high mortality that should be distinguished from botulism. This report confirms the need to associate history, clinical signs, absence of lesions with laboratory research for the diagnosis of botulism in birds.     Diagnóstico anatomopatológico, imuno-histoquímico e biomolecular de um caso de miocardite por Clostridium chauvoei em um bovino

    The aim of this study was to report a case of clostridial myocarditis in a bovine in Brazil with emphasis on the pathological findings, isolation and molecular identification associated with the in situ localization of C. chauvoei. The animal, a male Brangus bull with nine months of age, was found dead without prior clinical signs. Multifocal and coalescent areas of necrosis were observed in the myocardium. Rod cells in the cardiac muscle fibers were positive immunostaining for C. chauvoei, while this bacterium was also isolated and identified by polymerase chain reaction (PCR).O objetivo deste estudo foi relatar um caso de miocardite clostridial em um bovino no Brasil, com ênfase nos achados patológicos, isolamento e identificação molecular do agente e visualização in situ de C. chauvoei. O animal, um macho da raça Brangus com nove meses de idade, foi encontrado morto sem sinais clínicos prévios. Foram observadas áreas de necrose multifocal e coalescente no miocárdio, com a presença de bastonetes imunomarcados para C. chauvoei nas fibras musculares cardíacas. Uma estirpe de C. chauvoei foi isolada do tecido e sua identidade foi confirmada por reação em cadeia da polimeras (PCR)

    Detecção de agentes zoonóticos e fatores de risco associados a diarreia em cães de um município do semiárido brasileiro

    Os animais domésticos são colonizados por uma extensa diversidade de bactérias comensais e patogênicas. Particularmente, os de estimação têm sido apontados como reservatórios de algumas bactérias específicas, como Salmonella spp. e Clostridioides (Clostridium) difficile. Esses animais também podem estar infectados por helmintos potencialmente zoonóticos. No entanto, pouco se sabe sobre os fatores de risco e frequência desses patógenos em cães no Brasil. O objetivo neste estudo foi avaliar a presença de Salmonella spp., C. difficile e helmintos com potencial zoonótico em cães no município de Barra (Bahia, Brasil), bem como fatores de risco associados à ocorrência de diarreia. Para isso, foram obtidas as fezes de 40 cães. Cada tutor respondeu a um questionário epidemiológico para identificar episódios anteriores de diarreia nos animais e fatores de risco associados à presença dos patógenos avaliados. Ancylostoma spp. foi detectado em cinco cães (12,5%), enquanto Salmonella spp. foi isolado em 12,5% dos animais. C. difficile não foi isolado neste estudo. A presença de Ancylostoma spp. foi associada a fezes secas e ausência de vermifugação por mais de 12 meses. Cães positivos para Salmonella spp. foram cinco vezes mais propensos a ter diarreia do que aqueles negativos para este patógeno. Animais alimentados com dieta à base de carne crua e em contato com outros animais apresentaram maior probabilidade de apresentar episódio de diarreia nos últimos 12 meses. Pela primeira vez, Salmonella enterica sorovar Oranienburg foi isolado em um cão. Este trabalho sugere que se deve dar atenção à alimentação dos cães e à desparasitação regular para mitigar os riscos impostos por estes dois patógenos zoonóticos
