Arrow Plot: a new graphical tool for selecting up and down regulated genes and genes differentially expressed on samples subgroups

Background: A common task in analyzing microarray data is to determine which genes are differentially expressed across two (or more) kind of tissue samples or samples submitted under experimental conditions. Several statistical methods have been proposed to accomplish this goal, generally based on measures of distance between classes. It is well known that biological samples are heterogeneous because of factors such as molecular subtypes or genetic background that are often unknown to the experimenter. For instance, in experiments which involve molecular classification of tumors it is important to identify significant subtypes of cancer. Bimodal or multimodal distributions often reflect the presence of subsamples mixtures. Consequently, there can be genes differentially expressed on sample subgroups which are missed if usual statistical approaches are used. In this paper we propose a new graphical tool which not only identifies genes with up and down regulations, but also genes with differential expression in different subclasses, that are usually missed if current statistical methods are used. This tool is based on two measures of distance between samples, namely the overlapping coefficient (OVL) between two densities and the area under the receiver operating characteristic (ROC) curve. The methodology proposed here was implemented in the open-source R software. Results: This method was applied to a publicly available dataset, as well as to a simulated dataset. We compared our results with the ones obtained using some of the standard methods for detecting differentially expressed genes, namely Welch t-statistic, fold change (FC), rank products (RP), average difference (AD), weighted average difference (WAD), moderated t-statistic (modT), intensity-based moderated t-statistic (ibmT), significance analysis of microarrays (samT) and area under the ROC curve (AUC). On both datasets all differentially expressed genes with bimodal or multimodal distributions were not selected by all standard selection procedures. We also compared our results with (i) area between ROC curve and rising area (ABCR) and (ii) the test for not proper ROC curves (TNRC). We found our methodology more comprehensive, because it detects both bimodal and multimodal distributions and different variances can be considered on both samples. Another advantage of our method is that we can analyze graphically the behavior of different kinds of differentially expressed genes. Conclusion: Our results indicate that the arrow plot represents a new flexible and useful tool for the analysis of gene expression profiles from microarrays

As memórias de tradução: um estudo de caso na AP - PORTUGAL

Relatório de estágio de mestrado em Estudos Interculturais Português/Chinês (área de especialização em Tradução, Formação e Comunicação Empresarial)O presente relatório apresentará as atividades desenvolvidas durante o estágio curricular do Mestrado em Estudos Interculturais Português/Chinês: Tradução, Formação e Comunicação Empresarial, realizado na empresa de tradução AP | PORTUGAL, num período compreendido de seis meses, a partir de fevereiro 2017 até julho 2017. Focar-se-á principalmente na tarefa principal que desenvolvi ao longo do estágio, dedicada à Gestão das Memórias de Tradução. O primeiro capítulo visa apresentar a empresa com uma breve apreciação global do estágio, no segundo capítulo irá ser feita uma apresentação da Tradução Assistida por Computador: CAT (Computer Assisted Translation Tools), irá também ser feita uma abordagem às memórias de tradução recorrendo à descrição da tarefa principal desenvolvida no estágio curricular, nomeadamente, a gestão das memórias de tradução: em que consistiu este projeto, e a enumeração de erros e inconsistências encontradas ao longo do mesmo. No terceiro capítulo será feita uma descrição das três fases do projeto da gestão das memórias de tradução, sendo também apresentadas outras atividades desenvolvidas, sendo estas: o DTP – Desktop Publishing, a tradução, revisão e releitura, o controlo de qualidade, as transcrições, alinhamentos para a criação de memórias de tradução e por fim uma breve descrição dos guias de instrução de trabalho.This report will present the activities developed during the internship of the Master's Degree in Portuguese / Chinese Intercultural Studies: Translation, Training and Business Communication, carried out in the translation company AP | PORTUGAL, in a period of six months, from February 2017 to July 2017. It will focus mainly on the main task that I developed during the internship, dedicated to the Management of Translation Memories. This first chapter aims to present the company with an overall assessment of the internship, in the second chapter will be presented the Computer Assisted Translation (CAT), and an approach to the translation memories will be made using the description of the main task developed during the internship, namely the management of translation memories: in which consisted this project, and the enumeration of errors and inconsistencies found throughout this project. In the third chapter, a brief description of the 3 phases of the management of translation memories project will be made, as well as other activities such as: DTP - Desktop Publishing, translation, revision and proofreading, quality control, transcriptions, alignments for creating translation memories and finally a brief description of work instruction guides.本实习报告将介绍我在AP|PORTUGAL 翻译公司进行的实习活动。该实习从2017 年2 月开始，到同一年7 月结束，前后共计6 个月。这篇报告将着重介绍我在实习期间 完成的主要工作，即翻译记忆管理。 第一章旨在呈现我的实习概况，第二章将介绍计算机辅助翻译（CAT），通过描 述在实习期间的主要工作，我将陈述翻译记忆的方法，即翻译记忆管理法。这一方法 包含本项目，以及对于错误和不一致性的举例说明。 第三章将描述翻译记忆管理的三个步骤,以及其它活动，如：DTP - 桌面出版， 翻译，修订和校对，质量控制，翻译，创建翻译记忆库，和简要的工作说明手册

Gestão de recursos humanos na administração pública: um caso de um hospital público

O presente relatório tem como principal objetivo descrever as atividades desenvolvidas no âmbito do estágio curricular, inserido na componente não letiva do Mestrado em Contabilidade e Gestão Pública, ministrado pelo Instituto Superior de Contabilidade e Administração de Coimbra. O estágio foi realizado no serviço de recursos humanos do Hospital de Santo Espírito da Ilha Terceira e teve como principal intuito desenvolver atividades relacionadas com a administração de pessoal, em termos de carreiras, remunerações, assiduidade, mobilidade, seleção e recrutamento. Para a realização do estágio foi fundamental quer a aplicação de conceitos apreendidos nas Unidades Curriculares da componente letiva do Mestrado em Contabilidade e Gestão Pública, quer a aprendizagem que foi sendo obtida em contexto real de trabalho. Deste modo, foi possível adquirir competências profissionais na área de Gestão de Recursos Humanos, obter experiência em contexto de trabalho e compreender a dinâmica e respetivo funcionamento do serviço de Gestão de Recursos Humanos do Hospital de Santo Espírito da Ilha Terceira

Planeamento urbanístico e gestão do património arqueológico : a zona intra muros do centro histórico de Coimbra

Dissertação de mestrado em Arqueologia e Território, apresentada à Faculdade de Letras da Universidade de CoimbraEsta dissertação pretendeu fazer o diagnóstico dos parâmetros e modus operandi da prática e gestão arqueológica na cidade de Coimbra, com principal enfoque na zona intra-muros do Centro Histórico dessa urbe, no tempo actual. E, a partir desses dados, apontar metodologias e técnicas de gestão do património arqueológico municipal. Partiu-se do estado da investigação arqueológica para o conhecimento da cidade, com o objectivo de caracterizar o espaço definido para este estudo. Para tal, registaram-se, analisaram-se e traduziram-se em cartografia: o edificado (O que se vê), as intervenções arqueológicas (O substracto) e apontaram-se metodologias de trabalho conducentes à prática e gestão patrimonial. Tendo em atenção o estudo efectuado, propuseram-se novas sementes conducentes a um renovado modelo de gestão, não sem antes se fazer uma análise aos conceitos e problemáticas da arqueologia em meio urbano. A conjugação destes dois pilares (prática arqueológica e modelos de gestão) deve estar umbilicalmente ligada aos mecanismos legais de protecção e planeamento urbanístico de responsabilidade de cada município. Assim, foram apontadas as ferramentas (virtuais e legais) que nos pareceram mais adequadas para a gestão da arqueologia urbana.The present study is intended to observe the nowadays methodology and «modus operandi» of the actual real archaeological practice and management in the city of Coimbra, specially as far as the Historic Center inside the ancient Wall is concerned. Based upon the observed data we tried to point out more rational ways of doing things, regarding the best possible preservation of the archaeological city treasures. The retrospective observational design of this work was fullfiled, starting from the «state of the art» of archeology today to the real knowledge of the field, in order to frame the space to be looked at. So, the following items were observed, analysed and mapped : the built (what can be seen); different level of intervention ( substrate) ;subsequently some different methodologies are being indicated. Our aim has been to determine, by both observation and study of a « data corpus » about the prevalence of minor or major distance towards a renewed approach to models, having, after all, concluded that the scientific archaeological methods must always be strongly linked to the legal regulations of urban protection and planning of each and every City Hall all freely different among them

The experience of the persons with critical condition hospitalized in an intensive care unit

Objective: to assess the experience of persons with critical condition during hospitalization in an intensive care unit. Method: study within the qualitative paradigm using a phenomenological descriptive approach. The phenomenological approach allowed accessing the reports about the experience of twelve participants who agreed to be interviewed. Results: the essential structure that reflects the nature of the experience of persons with critical condition during hospitalization in an intensive care unit consists of three components: feeling trapped – the loss of control over the body; between life and death; and the need for security. Conclusion: it is an experience of suffering, felt in a unique and singular manner. Waking up in the intensive care unit was scary and hopeless. The existence is compromised; life and death are a constant. These persons need to feel secure, valuing the immediate care provided by the nurses. O processo artístico e biográfico na obra do escultor Paulo Neves

Mestrado em Criação Artística ContemporâneaA presente dissertação analisa a vida e obra do artista português Paulo Neves. Consideração da obra do artista, de que forma o seu trabalho é influenciado pelo espaço onde nasceu, reside e trabalha e onde construiu a Galeria O Mato. O artista é autor de uma obra vasta que poderá ser abordada por diferentes pontos de vista, a nossa investigação considera o caso de estudo do artista através da análise das suas principais obras de forma a perceber a génese do seu trabalho e do seu processo artístico. O estudo foi abordado em dois momentos espelhados no corpo da dissertação que é dividido em duas partes: O primeiro capítulo apresenta a biografia do artista e um enquadramento histórico, cultural e artístico da época em que o seu trabalho começou a ser produzido. Essa análise incidirá sobre as décadas de 80 e 90, uma vez que correspondem ao período no qual o artista iniciou a sua obra. Correspondentemente atendemos às exposições que realizou nesse período e ainda às recentes produções, analisando o que foi escrito pela crítica. A segunda parte procura verificar a influência da experiencia do autor nas suas obras e perceber o seu processo artístico na sua relação com os materiais e o lugar na construção da obra. Finalmente, procuraremos concluir o estudo tentando perceber a construção atendendo à forte relação com a matéria e tentando verificar a influência do lugar onde reside.This dissertation analyzes the life and work of Portuguese artist Paulo Neves. It discusses the artist's work and how his work was influenced by the place where he was born, where he lives and works and where he built the gallery O Mato. The artist has authored a vast work, which can be approached from different points of view, our research is a case study of the artist through the analysis of his major work in order to understand the genesis of his work and his artistic process. The body of the dissertation is divided into two mirrored parts: The first part is a biographical study of the artist and a historical, cultural and artistic framework of the period when his work was starting to be produced. This analysis will focus on the 80's and 90's, since they correspond to the period in which the artist began his work. Correspondingly we discuss the exhibitions that took place during this period and also the recent productions, analyzing what has been written by critics. The second part tries to determine the influence of the experience of the author in his work and to understand the artistic process in its relation with the material and space in the construction of his work. Finally, we conclude the study trying to understand the construction given his strong relationship with the material and trying to understand the influence of the place where he lives
