    Apoptosis of non-parasitized red blood cells in malaria: a putative mechanism involved in the pathogenesis of anaemia

    <p>Abstract</p> <p>Background</p> <p>Severe anaemia is a common complication of <it>Plasmodium falciparum </it>malaria in hyperendemic regions. Premature elimination of non-parasitized red blood cells (nRBC) has been considered as one mechanism involved in the genesis of severe malaria anaemia. It has been reported that apoptosis can occur in RBC and, consequently, this cell death process could contribute to anaemia. This study was performed to evaluate the susceptibility of nRBC to apoptosis in a malaria anaemia murine model.</p> <p>Methods</p> <p>Balb/c mice were intraperitonially inoculated with 1 × 10<sup>6 </sup><it>P. yoelii </it>17XL parasitized RBC (pRBC) and, then, parasitaemia and anaemia were monitored. Apoptosis in both pRBC and nRBC was assessed during early and late phases of infection by flow cytometry using Syto 16 and annexin V-PE double staining and forward scatter measurement.</p> <p>Results</p> <p>As expected, experimental infection of Balb/c mice with <it>Plasmodium yoelii </it>17XL parasites was characterized by progressive increase of parasitaemia and acute anaemia, leading to death. Flow cytometry analysis showed that a number of pRBC was in the apoptotic process. It was noteworthy that the increase of nRBC apoptosis levels occurred in the late phase of infection, when anaemia degree was notably accentuated, while no significant alteration was observed in the early phase.</p> <p>Conclusion</p> <p>The increased levels of nRBC apoptosis herein firstly reported, in malaria infection could represent a putative mechanism worsening the severity of malarial anaemia.</p

    Hypothalamic S1p/s1pr1 Axis Controls Energy Homeostasis In Middle-aged Rodents: The Reversal Effects Of Physical Exercise

Here, we found that reduction of hypothalamic S1PR1 expression occurs in an age-dependent manner, and was associated with defective thermogenic signaling and weight gain. To address the physiological relevance of these findings, we investigated the effects of chronic and acute exercise on the hypothalamic S1PR1/STAT3 axis. Chronic exercise increased S1PR1 expression and STAT3 phosphorylation in the hypothalamus, restoring the anorexigenic and thermogenic signals in middle-aged mice. Acutely, exercise increased sphingosine-1-phosphate (S1P) levels in the cerebrospinal fluid (CSF) of young rats, whereas the administration of CSF from exercised young rats into the hypothalamus of middle-aged rats at rest was sufficient to reduce the food intake. Finally, the intracerebroventricular (ICV) administration of S1PR1 activators, including the bioactive lipid molecule S1P, and pharmacological S1PR1 activator, SEW2871, induced a potent STAT3 phosphorylation and anorexigenic response in middle-aged rats.     Valor nutritivo de pastagens de capim-elefante manejadas sob sistema convencional e agroecológico.

    O capim-elefante é utilizado, na sua grande maioria, em sistemas convencionais de produção animal. O objetivo deste trabalho foi comparar o valor nutritivo do capimelefante em sistemas de manejo agroecológico e convencional, quanto a proteína bruta (PB), fibra em detergente neutro (FDN) e digestibilidade in vitro da matéria seca (DIVMS). Foram usados quatro piquetes, com 0,12ha cada um. No sistema convencional, o capim-elefante foi estabelecido singularmente. No sistema agroecológico, o plantio foi feito em linhas afastadas de 3m. Nas entrelinhas, estabeleceu-se a aveia e o azevém no período hibernal, enquanto que no período estival permitiu-se o desenvolvimento de espécies espontâneas. A adubação foi feita com fertilizantes orgânicos (150kg ha-1 de N). No sistema agroecológico, foram conduzidos sete pastejos, de 24/04/2004 a 05/05/2005. Na pastagem convencional, usouse a mesma quantidade de N (uréia), sendo conduzidos quatro ciclos de pastejo, de 06/10/2004 a 05/05/2005. Para ambos os sistemas foram utilizadas vacas da raça Holandês, recebendo complementação alimentar de 3,5kg dia-1 de concentrado com 20% de proteína bruta, constituindo-se nos animais experimentais. Nas avaliações, considerou-se a massa de forragem inicial com base na matéria seca (MS), os componentes botânicos da pastagem e estruturais do capimelefante. As análises de qualidade foram feitas em amostras de pastejo simulado. O delineamento experimental foi o inteiramente casualizado com dois tratamentos, convencional e agroecológico, duas repetições (piquetes) e em parcelas incompletas subdivididas no tempo (pastejos). Houve interação (P<0,05) entre tratamentos e pastejos em todas as variáveis. Na pastagem agroecológica, o modelo que melhor se ajustou foi o cúbico para todas as variáveis, em função do tempo de pastejo. Na pastagem convencional, a PB e a DIVMS ajustaram-se melhor ao modelo linear, com taxa positiva de crescimento, sendo observado comportamento inverso para FDN, com o decorrer dos pastejos. Tanto na pastagem convencional quanto na agroecológica encontraram-se associações negativas entre lâmina foliar do capim-elefante com PB e DIVMS e positiva com FDN. Ambos os sistemas apresentaram teores qualitativos elevados das pastagens, considerando-se a adubação, o manejo e o tempo de utilização

    Reductions in the Prevalence and Incidence of Geohelminth Infections following a City-wide Sanitation Program in a Brazilian Urban Centre

    In the city of Salvador, a large urban centre in Northeast Brazil, a city-wide sanitation intervention started in 1997, aiming to improve the sewerage coverage of households from 26% to 80%. Our aim was to study the impact of the intervention on the prevalence and incidence of geohelminths in the school-aged population. The longitudinal study comprised two cohorts: from the beginning of 1997 to 1998, where data was collected before the intervention, and at the end of 2003 to 2004, after the intervention. Copro-parasitological examinations were carried out on every individual from both cohorts, at the start and nine months later. Demographic, socio-economic, and environmental data were collected using semi-structured questionnaires. The variables utilized to demonstrate the effects of intervention, when utilized together in a multivariate model, accounted for a 100% observed reduction in the prevalence ratio (PR) and incidence ratio (IR). As well as proving that the variables associated with the effect of the program intervention were mediators in this reduction, the reduction in the PR and IR between these periods demonstrated that modifications to the urban environment, particularly those associated with sanitary sewage systems, affected the health of the population, significantly reducing the prevalence of geohelminths

    Esta comunicação apresenta a análise de livros didáticos de Química para o ensino médio, indicados no PNLD 2012 (Brasil, 2011), em relação a contextualização dos conteúdos químicos, como resultado de atividade desenvolvida com bolsistas do Programa Institucional de Bolsa de Iniciação a Docência do curso de Licenciatura em Química da Universidade Federal da Bahia (PIBID/UFBA), contando com a participação dos estudantes matriculados na disciplina Ensino de Química no Contexto, no semestre 2011.2. Foram consideradas três categorias de análise, baseadas em Silva (2007) e os resultados apontaram que dos livros analisados, três apresentam a contextualização como exemplificação e/ou informação do cotidiano, um apresenta contextualização relacionada ao movimento CTS e um outro aproxima-se também desse tipo de contextualização, embora tenha como foco a construção do conhecimento

    Industrial scale isolation, structural and spectroscopic characterization of epiisopiloturine from Pilocarpus microphyllus stapf leaves: a promising alkaloid against schistosomiasis

    This paper presents an industrial scale process for extraction, purification, and isolation of epiisopiloturine (EPI) (2(3H)- Furanone,dihydro-3-(hydroxyphenylmethyl)-4-[(1-methyl-1H-imidazol-4-yl)methyl]-, [3S-[3a(R*),4b]]), which is an alkaloid from jaborandi leaves (Pilocarpus microphyllus Stapf). Additionally for the first time a set of structural and spectroscopic techniques were used to characterize this alkaloid. EPI has shown schistomicidal activity against adults and young forms, as well as the reduction of the egg laying adult worms and low toxicity to mammalian cells (in vitro). At first, the extraction of EPI was done with toluene and methylene chloride to obtain a solution that was alkalinized with ammonium carbonate. The remaining solution was treated in sequence by acidification, filtration and alkalinization. These industrial procedures are necessary in order to remove impurities and subsequent application of the high performance liquid chromatography (HPLC). The HPLC was employed also to remove other alkaloids, to obtain EPI purity higher than 98%. The viability of the method was confirmed through HPLC and electrospray mass spectrometry, that yielded a pseudo molecular ion of m/z equal to 287.1 Da. EPI structure was characterized by single crystal X-ray diffraction (XRD), 1H and 13C nuclear magnetic resonance (NMR) in deuterated methanol/chloroform solution, vibrational spectroscopy and mass coupled thermal analyses. EPI molecule presents a parallel alignment of the benzene and the methyl imidazol ring separated by an interplanar spacing of 3.758 Å indicating a π-π bond interaction. The imidazole alkaloid melts at 225°C and decomposes above 230°C under air. EPI structure was used in theoretical Density Functional Theory calculations, considering the single crystal XRD data in order to simulate the NMR, infrared and Raman spectra of the molecule, and performs the signals attribution

    Hypothalamic S1P/S1PR1 axis controls energy homeostasis

    Sphingosine 1-phosphate receptor 1 (S1PR1) is a G-protein-coupled receptor for sphingosine-1-phosphate (S1P) that has a role in many physiological and pathophysiological processes. Here we show that the S1P/S1PR1 signalling pathway in hypothalamic neurons regulates energy homeostasis in rodents. We demonstrate that S1PR1 protein is highly enriched in hypothalamic POMC neurons of rats. Intracerebroventricular injections of the bioactive lipid, S1P, reduce food consumption and increase rat energy expenditure through persistent activation of STAT3 and the melanocortin system. Similarly, the selective disruption of hypothalamic S1PR1 increases food intake and reduces the respiratory exchange ratio. We further show that STAT3 controls S1PR1 expression in neurons via a positive feedback mechanism. Interestingly, several models of obesity and cancer anorexia display an imbalance of hypothalamic S1P/S1PR1/STAT3 axis, whereas pharmacological intervention ameliorates these phenotypes. Taken together, our data demonstrate that the neuronal S1P/S1PR1/STAT3 signalling axis plays a critical role in the control of energy homeostasis in rats
