In this article, we present a study on the experience of app drivers, conducted during the first half of 2020, during the lockdown months due to the Covid19 pandemic. Initially, we develop a brief literature review about the transformations in the world of work, and then a preliminary study about the Brazilian occupational structure based on available statistics from the Brazilian Institute of Geography and Statistics (IBGE). In Brazil, the unemployment rate has been on the rise in recent years, and with it the level of informality, that is, the number of workers without a signed labor contract and, therefore, without the guarantee of labor rights, has also increased. We can observe in the last three years a significant increase in the number of app drivers and delivery drivers. This category, for not having labor rights, faces precariousness, because it works for multinational companies that profit a lot, without any employment ties. This reality became even more evident in periods of crisis, as in the case of the coronavirus pandemic, when these workers could not comply with the isolation rules, because if they do not work they cannot support their families. The research intends to contribute to the understanding about the changes in work relations caused by the advance of digital tools that facilitate the exchange of services. To make this possible three specific objectives were defined: i) to map the debate in the specialized literature about labor relations mediated by the popularization of service provision apps; ii) to explore the statistics available from the Brazilian Institute of Geography and Statistics (IBGE) about the theme and present an exploratory study about the profile of app drivers. Public statistics on the economically active population will be examined and information on the occupational trajectories of service providers will be collected. Further discussion is expected about how these tools of service provision relate to work and how they modify these relationships based on the historical, economic, and political moment. They will also make it possible to contribute to producing sociodemographic information about these workers, about whom we have almost no statistics. Obesity is a chronic inflammatory disease that affects millions of people worldwide. Most studies observe the effects of a high-fat diet (HFD) in 10–12 weeks. This work investigated the effects induced by a HFD administered for 6 weeks on the nutritional status of mice and some aspects of the inflammatory response in mouse peritoneal macrophages. Male Swiss Webster mice, 2–3 months of age, were fed a control diet or HFD for 6 weeks. After this period, the mice were euthanized, and peritoneal macrophages were collected for immunoassays and assessment of biochemical parameters. A HFD was associated with increased cholesterol, insulin resistance, C-reactive protein (CRP), leptin, and serum resistin levels. Lipopolysaccharide (LPS)- stimulated adipocyte cultures of animals subjected to a HFD showed increased production of proinflammatory cytokines such as tumor necrosis factor alpha (TNF-α), interleukin-1 (IL-1), and interleukin-6 (IL-6). However, peritoneal macrophages of the HFD group showed no changes in the levels of these cytokines. LPS-stimulated peritoneal macrophages from HFD-treated animals showed a reduction in mRNA expression of TNF-α and IL-6, as well as a decrease in expression of the transcription factor nuclear factor-kappa B (NF-kB). In conclusion, HFD treatment for 6 weeks induces similar signs to metabolic syndrome and decreases the capacity of peritoneal macrophages to develop an appropriate inflammatory response to a bacterial component

AbstractSawmills are the most important wood industries in the Southern of the Espírito Santo State. The aim of this research was to analyze the operational performance of a small sawmill in Alegre, Espírito Santo state, Brazil, which process Eucalyptus sp. logs. The analysis was based on the study of lumber yield, operational efficiency and work delay ratio. The average lumber yield was 53.6% ranked as good for a small log hardwood sawmill, according to the literature. The average operational efficiency was 2.77 m³/worker/shift, and was ranked as low, even for small Brazilian sawmills, due to not classified logs, layout inadequacies, machinery bad conservation and low automation level. The average productive work was 69.6% and did not achieve the minimum rank of 75% suggested by the literature, because of the referred problems, and other ones. In general, for such sawmill the operational situation was considered poor, but it has potential to be improved. AbstractOperational performance of a small sawmill of the city of Alegre, Espírito Santo, Brazil. Introduction: The columella is an important subunit of the nose, essential for nasal architecture and facial aesthetics. The total reconstruction of the nasal columella becomes a great challenge after repairing trauma, carcinomas, and necrosis in this region. There are descriptions in the literature of numerous reconstruction techniques using different flaps, such as a frontal region flap, an infraclavicular region flap, and a unilateral and bilateral nasolabial flap. Case Report: A total reconstruction of the nasal columella after resection of basal cell carcinoma (BCC) using a bilateral nasolabial flap is reported. Conclusion: The technique proved effective for correcting the complex defect after BCC resection, with technical ease for resolution and good aesthetic and functional results

The use of animals in scientific research has contributed significantly to the development of science, promoting various advances in understanding the metabolic machinery and the discovery of treatments and preventive measures applied to human and veterinary medicine. The development and use of alternative methods is encouraged; however, in some situations, the use of animals in accordance with ethical policies is still required. Established hematological and clinical chemistry reference values in laboratory animals are essential to evaluate functional changes; however, there are few data in the literature on these values, being fundamentally a comparative basis. The aim of this investigation was the establishment of hematological and clinical chemistry reference values in common strains/stocks of mice used in animal experimentation. Blood profile (hemogram, reticulocytes and myelogram) and clinical chemistry serum determination of total protein, albumin, glucose, cholesterol, triglycerides, calcium and phosphorus were evaluated using C57BL/6, BALB/c and Swiss Webster mice, male, 2-3 months old.

Belo Horizonte, MG, Brasil. Background: For a long time, the role of CD8+ T cells in blood-stage malaria was not considered important because erythrocytes do not express major histocompatibility complex (MHC) class I proteins. While recent evidences suggest that CD8+ T cells may play an important role during the erythrocytic phase of infection by eliminating parasites, CD8+ T cells might also contribute to modulate the host response through production of regulatory cytokines. Thus, the role of CD8+ T cells during blood-stage malaria is unclear. Here, we report the phenotypic profiling of CD8+ T cells subsets from patients with uncomplicated symptomatic P. vivax malaria. Methods: Blood samples were collected from 20 Plasmodium vivax-infected individuals and 12 healthy individuals. Immunophenotyping was conducted by flow cytometry. Plasma levels of IFN-γ, TNF-α and IL-10 were determined by ELISA/CBA. Unpaired t-test or Mann–Whitney test was used depending on the data distribution. Results: P. vivax-infected subjects had lower percentages and absolute numbers of CD8+ CD45RA+ and CD8+ CD45RO+ T cells when compared to uninfected individuals (p ≤ 0.0002). A significantly lower absolute number of circulating CD8+ CD45+ CCR7+ cells (p = 0.002) was observed in P. vivax-infected individuals indicating that infection reduces the number of central memory T cells. Cytokine expression was significantly reduced in the naïve T cells from infected individuals compared with negative controls, as shown by lower numbers of IFN-γ + (p = 0.001), TNF-α+ (p < 0.0001) and IL-10+ (p < 0.0001) CD8+ T cells. Despite the reduction in the number of CD8+ memory T cells producing IFN-γ (p < 0.0001), P. vivax-infected individuals demonstrated a significant increase in memory CD8+ TNF-α+ (p = 0.016) and CD8+ IL-10+ (p = 0.004) cells. Positive correlations were observed between absolute numbers of CD8+ IL-10+ and numbers of CD8+ IFN-γ + (p < 0.001) and CD8+ TNF-α+ T cells (p ≤ 0.0001). Finally, an increase in the plasma levels of TNF-α (p = 0.017) and IL-10 (p = 0.006) and a decrease in the IFN-γ plasma level (p <0.0001) were observed in the P. vivax-infected individuals. Conclusions: P. vivax infection reduces the numbers of different subsets of CD8+ T cells, particularly the memory cells, during blood-stage of infection and enhances the number of CD8+ memory T cells expressing IL-10, which positively correlates with the number of cells expressing TNF-α and IFN-γ
