Eradication of Candida albicans persister cell biofilm by the membranotropic peptide gH625
Biofilm formation poses an important clinical trouble due to resistance to antimicrobial agents; therefore, there is an urgent demand for new antibiofilm strategies that focus on the use of alternative compounds also in combination with conventional drugs. Drug-tolerant persisters are present in Candida albicans biofilms and are detected following treatment with high doses of amphotericin B. In this study, persisters were found in biofilms treated with amphotericin B of two clinical isolate strains, and were capable to form a new biofilm in situ. We investigated the possibility of eradicating persister-derived biofilms from these two Candida albicans strains, using the peptide gH625 analogue (gH625-M). Confocal microscopy studies allowed us to characterize the persister-derived biofilm and understand the mechanism of interaction of gH625-M with the biofilm. These findings confirm that persisters may be responsible for Candida biofilm survival, and prove that gH625-M was very effective in eradicating persister-derived biofilms both alone and in combination with conventional antifungals, mainly strengthening the antibiofilm activity of fluconazole and 5-flucytosine. Our strategy advances our insights into the development of effective antibiofilm therapeutic approaches
Critical Steps of Plasmodium falciparum Ookinete Maturation
The egress and fertilization of Plasmodium gametes and development of a motile ookinete are the first crucial steps that mediate the successful transmission of the malaria parasites from humans to the Anopheles vector. However, limited information exists about the cell biology and regulation of this process. Technical impediments in the establishment of in vitro conditions for ookinete maturation in Plasmodium falciparum and other human malaria parasites further constrain a detailed characterization of ookinete maturation. Here, using fluorescence microscopy and immunolabeling, we compared P. falciparum ookinete maturation in Anopheles coluzzii mosquitoes in vivo and in cell culture in vitro. Our results identified two critical steps in ookinete maturation that are regulated by distinct mosquito factors, thereby highlighting the role of the mosquito environment in the transmission efficiency of malaria parasites

Polymicrobial antibiofilm activity of the membranotropic peptide gH625 and its analogue
Abstract This work illustrates a new role for the membranotropic peptide gH625 and its derivative gH625-GCGKKK in impairing formation of polymicrobial biofilms. Mixed biofilms composed of Candida and bacterial species cause frequently infections and failure of medical silicone devices and also show a major drug resistance than single-species biofilms. Inhibition and eradication of biofilms were evaluated by complementary methods: XTT-reduction, and crystal violet staining (CV). Our results indicate that gH625-GCGKKKK, better than the native peptide, strongly inhibited formation of mixed biofilms of clinical isolates of C. tropicalis/S. marcescens and C. tropicalis/S. aureus and reduced the biofilm architecture, interfering with cell adhesion and polymeric matrix, as well as eradicated the long-term polymicrobial biofilms on silicone surface
Total Cross Sections for Neutron Scattering
Measurements of neutron total cross-sections are both extensive and extremely
accurate. Although they place a strong constraint on theoretically constructed
models, there are relatively few comparisons of predictions with experiment.
The total cross-sections for neutron scattering from O and Ca are
calculated as a function of energy from ~MeV laboratory energy with a
microscopic first order optical potential derived within the framework of the
Watson expansion. Although these results are already in qualitative agreement
with the data, the inclusion of medium corrections to the propagator is

Polydopamine-Coated Magnetic Iron Oxide Nanoparticles: From Design to Applications
Magnetic iron oxide nanoparticles have been extensively investigated due to their applications in various fields such as biomedicine, sensing, and environmental remediation. However, they need to be coated with a suitable material in order to make them biocompatible and to add new functionalities on their surface. This review is intended to give a comprehensive overview of recent advantages and applications of iron oxide nanoparticles coated by polydopamine film. The synthesis method of magnetic nanoparticles, their functionalization with bioinspired materials and (in particular) with polydopamine are discussed. Finally, some interesting applications of polydopamine-coated magnetic iron oxide nanoparticles will be pointed out
Towards a comprehensive taxonomy for characterizing robots
Every day a new robot is developed with advanced characteristics and technical qualities. The increasingly rapid growth of robots and their characteristics demands bridging between the application requirements and the robot specifications. This process requires a supporting conceptual structure that can capture as many robot qualities as possible. Presenting robot characteristics through the proposed conceptual structure would enable designers to optimize robot capabilities against application requirements. It would also help application developers to select the most appropriate robot. Without a formal structure, an accurate linking between the robot domain and the application domain is not possible. This paper presents a novel theoretical representation that can capture robot features and capabilities and express them as descriptive dimensions to be used to develop a capability profile. The profile is intended to unify robot description and presentation. The proposed structure is reinforced with several layers, sections, categorizations and levels to allow a detailed explanation of robot characteristics. It is hoped that the proposed structure will influence the design, development, and testing of robots for specific applications. At the same time, it would help in highlighting the corresponding outlines in robot application requirements
Stem cell therapies for ischemic cardiovascular diseases
Myocardial infarction results in loss of cardiac muscle and deficiency in cardiac performance. Likewise, peripheral artery disease can result in critical limb ischemia leading to reduced mobility, non-healing ulcers, gangrene and amputation. Both of these common conditions diminish quality of life and enhance risk of mortality. Successful advances in treatment have led to more people surviving incidences of myocardial infarction or living with peripheral artery disease. However, the current treatments are inadequate in repairing ischemic tissue. Over the last 5 years, a vast number of patents have been submitted concerning the use of stem cells, which correlates with the exponential growth in stem cell publications. Exploiting stem cell therapy offers a real potential in replacing ischemic tissue with functional cells. In this paper, we review recent patents concerning stem cell therapy that have the potential to provide or potentiate novel treatment for ischemic cardiovascular disease. In addition, we evaluate the promise of the inventions by describing some clinical trials that are currently taking place, as well as considering how current research on ischemic cardiovascular disease may change the patent landscape in the future
A first attempt to evaluate the toxicity to Phaeodactylum tricornutum Bohlin exposed to rare earth elements
The increasing use and demand of rare earth elements in many emerging technologies is leading to a potentially higher input to the marine environment. This study compared for the first time the effect of lanthanum (La), cerium (Ce), neodymium (Nd), samarium (Sm), europium (Eu), gadolinium (Gd), dysprosium (Dy), and erbium (Er) to the microalga Phaeodactylum tricornutum Bohlin. The algal growth inhibition was investigated after 72 h of exposure. The median effect concentrations (EC50) ranged from 0.98 mg/L to 13.21 mg/L and elements were ranked as follows: Gd > Ce > Er > La > Eu > Nd > Dy > Sm. The comparison of predicted no effect concentrations (PNEC) for hazard and risk assessment with measured environmental concentrations showed that ecological risks deriving from REEs could be present, but limited to specific environments like estuarine waters. The results support evidence of actions to manage the REE impact in seawater environments, looking to improve the monitoring tailored to the different and dynamic nature of ecosystems
