    Overexpression of TRPM7 suppresses AngII-induced phenotypic changes in hypertensive human vascular smooth muscle cells

    Purpose: To study the role of TRPM7 in suppressing AngII-induced phenotype changes in human vascular smooth muscle cells (HAVSMCs).Methods: Relative levels of TRPM7, α-SMA and OPN in AngII-induced HAVSMCs were determined, and the regulatory effects of TRPM7 on levels of α-SMA and OPN, wound healing effect, 5-ethynyl-2’- deoxyuridine (EdU)-positive ratio, and viability in AngII-induced HAVSMCs were assessed. A total of 159 patients with essential hypertension attending The Second Xiangya Hospital, Central South University, Changsha, China, and 70 healthy controls were enrolled; after treatment with angiotensin receptor blocker (ARB), changes in SBP, DBP and plasma level of TRPM7 in hypertension patients were examined. Potential correlation between TRPM7 level with SBP and DBP was assessed.Results: TRPM7 and α-SMA were downregulated, while OPN was upregulated in AngII-induced HAVSMCs, but this trend was partially reversed by losartan treatment (p < 0.05). Overexpression of TRPM7 reversed the expression changes of α-SMA and OPN in AngII-induced HAVSMCs. In addition, the overexpression of TRPM7 significantly inhibited the enhanced migratory and proliferative capacities in AngII-induced HAVSMCs (p < 0.05). After ARB treatment, SBP, DBP and plasma TRPM7 were significantly reduced in hypertension patients, while TRPM7 level positively correlated with SBP and DBP in hypertensive patients.Conclusion: Overexpression of TRPM7 blocked AngII-induced phenotype changes in HAVSMCs under the pathological circumstance of hypertension.&nbsp

    A Hybrid Model Based on Support Vector Machine for Bus Travel-Time Prediction

    Effective bus travel time prediction is essential in transit operation system. An improved support vector machine (SVM) is applied in this paper to predict bus travel time and then the efficiency of the improved SVM is checked. The improved SVM is the combination of traditional SVM, Grubbs’ test method and an adaptive algorithm for bus travel-time prediction. Since error data exists in the collected data, Grubbs’ test method is used for removing outliers from input data before applying the traditional SVM model. Besides, to decrease the influence of the historical data in different stages on the forecast result of the traditional SVM, an adaptive algorithm is adopted to dynamically decrease the forecast error. Finally, the proposed approach is tested with the data of No. 232 bus route in Shenyang. The results show that the improved SVM has good prediction accuracy and practicality

    Control of Temperature and Aluminum Fluoride Concentration Based on Model Prediction in Aluminum Electrolysis

    The temperature and the aluminum fluoride (AlF3) concentration of electrolyte greatly affect the current efficiency and energy consumption in aluminum electrolysis. This paper developed a new kind of algorithm to control the temperature and AlF3 concentration of electrolyte for 300 kA prebake aluminum production cells by altering the setting cell voltage and the AlF3 adding rate. One is liquidus model which can be used to calculate AlF3 concentration of electrolyte by some technical parameters and target superheat and the other is the temperature models which can determine the required cell voltage by the technical parameters and target temperature. Consequently, the AlF3 addition rate can be decided by the AlF3 concentration. The temperature and AlF3 concentration of electrolyte in the production are measured and the differences between the measurement and the corresponding calculation are used as feedback to correct the model. The coefficients in the model are also revised according to the external conditions. This system has been used in an aluminum company for 2 years. Both the temperature and the AlF3 concentration of electrolyte can be controlled as required and the energy consumption of the aluminum production was much decreased

    An optimal fractional-order accumulative Grey Markov model with variable parameters and its application in total energy consumption

    In this paper, we propose an optimal fractional-order accumulative Grey Markov model with variable parameters (FOGMKM (1, 1)) to predict the annual total energy consumption in China and improve the accuracy of energy consumption forecasting. The new model is built upon the traditional Grey model and utilized matrix perturbation theory to study the natural and response characteristics of a system when the structural parameters change slightly. The particle swarm optimization algorithm (PSO) is used to determine the number of optimal fractional order and nonlinear parameters. An experiment is conducted to validate the high prediction accuracy of the FOGMKM (1, 1) model, with mean absolute percentage error (MAPE) and root mean square error (RMSE) values of 0.51% and 1886.6, respectively, and corresponding fitting values of 0.92% and 6108.8. These results demonstrate the superior fitting performance of the FOGMKM (1, 1) model when compared to other six competitive models, including GM (1, 1), ARIMA, Linear, FAONGBM (1, 1), FGM (1, 1) and FOGM (1, 1). Our study provides a scientific basis and technical references for further research in the finance as well as energy fields and can serve well for energy market benchmark research

    Ago-2-Mediated Slicer Activity Is Essential for Anti-Flaviviral Efficacy of RNAi

    RNA interference can be mediated by fully complementary siRNA or partially complementary miRNA. siRNAs are widely used to suppress viral replication and the fully complementary siRNA bound Ago-2 in the RISC is known to degrade the target RNA. Although other argonaute proteins lacking slicer activity can also bind oligonucleotides with both si and miRNA structures, whether they can also contribute to antiviral effects is not entirely clear. We tested si and miRNA structured oligos for target repression in dual luciferase assays as well as for inhibition of Dengue and West Nile virus replication in ES cells expressing individual Ago proteins. In luciferase assays, both fully complementary and partially complementary oligos effectively repressed their targets in all individual Ago expressing cell lines, although the efficacy with fully complementary oligos was higher in Ago-2+ cells. However, partially complementary oligos had no effect on virus replication in any cell line, while fully complementary siRNAs were highly effective in Ago-2 expressing, but not in cells expressing other Ago proteins. This occurred irrespective of whether the target sequences were located in the coding region or 3′UTR of the virus. We conclude that Ago-2 slicer activity is essential for anti-viral efficacy of siRNAs and miRNA-mediated translational repression/transcript destabilization is too weak to suppress the abundantly expressed flaviviral proteins
