    Microarray-based global mapping of integration sites for the retrotransposon, intracisternal A-particle, in the mouse genome

    Mammalian genomes contain numerous evolutionary harbored mobile elements, a part of which are still active and may cause genomic instability. Their movement and positional diversity occasionally result in phenotypic changes and variation by causing altered expression or disruption of neighboring host genes. Here, we describe a novel microarray-based method by which dispersed genomic locations of a type of retrotransposon in a mammalian genome can be identified. Using this method, we mapped the DNA elements for a mouse retrotransposon, intracisternal A-particle (IAP), within genomes of C3H/He and C57BL/6J inbred mouse strains; consequently we detected hundreds of probable IAP cDNA–integrated genomic regions, in which a considerable number of strain-specific putative insertions were included. In addition, by comparing genomic DNAs from radiation-induced myeloid leukemia cells and its reference normal tissue, we detected three genomic regions around which an IAP element was integrated.     Insulin resistance is often associated with atherosclerotic diseases in subjects with obesity and impaired glucose tolerance. This study examined the effects of insulin resistance on coronary risk factors in IRS-1 deficient mice, a nonobese animal model of insulin resistance. Blood pressure and plasma triglyceride levels were significantly higher in IRS-1 deficient mice than in normal mice. Impaired endothelium-dependent vascular relaxation was also observed in IRS-1 deficient mice. Furthermore, lipoprotein lipase activity was lower than in normal mice, suggesting impaired lipolysis to be involved in the increase in plasma triglyceride levels under insulin-resistant conditions. Thus, insulin resistance plays an important role in the clustering of coronary risk factors which may accelerate the progression of atherosclerosis in subjects with insulin resistance

    初・中等教育段階の日本語教育は、各国・地域の外国語教育政策に基づいて展開されており、シラバス、カリキュラム、教材もそれぞれの国・地域で統一されている場合が多く、学習者の発達段階など多くの要因を考慮しながら授業を組み立てていく必要がある。初・中等教育の教師を対象とした研修の教授法授業では、このような背景を考慮した実践的な教授活動を取り入れていくことが重要である。2003年度海外日本語教師短期研修(春期)(以下、春短期研修)における教授法授業では以下の試みを行った。できる限り教授環境を共有する国・地域別のクラス編成を重視し、多国籍のクラスの人数を少数になるようにした。さらに、それぞれの教授環境を重視し、その一方で国・地域を超えた初・中等教育に共通する課題をその枠組みの中で取り上げることとした。 共通に扱う項目としては、それぞれの教師が普段行っているであろう教授活動の流れ、「教授目標設定→授業設計→授業準備・教材作成→授業実施→授業に対する評価」の流れを教授法授業で経験させること、コンピュータの利用、高校訪問という要素を、それぞれの教師の望む程度に応じて教授活動の中に織り込ませることであった。 本稿では、5クラスそれぞれの教授法授業の実際を報告し、コンピュータ利用、高校訪問については全体について報告した。研修に参加したほとんどの教師は、教授法授業のクラス分け、扱った内容について役立ったとしているが、違う国・地域の教師との情報交流の機会をより増やすことを望む声が多かった。さらに、コンピュータ利用を勧めたことについては、IT普及の如何に関わらず好評であった。高校訪問については、半数程度が役立つとしているが、問題があったとする意見もあり、教授法授業への取り入れ方については検討を要する。今後も、初・中等教育に携わる日本語教師への研修を改善していくためには、実践に根ざした情報の蓄積が必要である

    All students in the nursing care department must take the general graduate examination. The result does not afect students\u27 acquisition of the national license now. However, it is expected that each student must pass the examination to get the license in the future. We, professors in the nursing care department, educate and encourage them through various opportunities to become good care workers. The goal of passing the examination becomes more diicult year after year as a whole, for the acknowledged level of students lowers year after year. However, the knowledge of nursing care is critically important in providing appropriate nursing care. Therefore all the professors of the department determined to educate all the students in order to pass the examination. It is a hard object for the professors to attain. We must improve our teaching skills through our faculty development activities. Last year we had many special lessons for our students to prepare the examination. The number of passing the examination improved amazingly last year, compared to the previous year. We will continue to improve our teaching skills to attain our goal, to make sure all the students pass the examinat ion

    Expression of myeloperoxidase and gene mutations in AML patients with normal karyotype: double CEBPA mutations are associated with high percentage of MPO positivity in leukemic blasts.

    The percentage of myeloperoxidase (MPO)-positive blast cells is a simple and highly significant prognostic factor in AML patients. It has been reported that the high MPO group (MPO-H), in which >50% of blasts are MPO activity positive, is associated with favorable karyotypes, while the low MPO group (≤50% of blasts are MPO activity positive, MPO-L) is associated with adverse karyotypes. The MPO-H group shows better survival even when restricted to patients belonging to the intermediate chromosomal risk group or those with a normal karyotype. It has recently been shown that genotypes defined by the mutational status of NPM1, FLT3, and CEBPA are associated with treatment outcome in patients with cytogenetically normal AML. In this study, we aimed to evaluate the relationship between MPO positivity and gene mutations found in normal karyotypes. Sixty AML patients with normal karyotypes were included in this study. Blast cell MPO positivity was assessed in bone marrow smears stained for MPO. Associated genetic lesions (the NPM1, FLT3-ITD, and CEBPA mutations) were studied using nucleotide sequencing. Thirty-two patients were in the MPO-L group, and 28 patients in the MPO-H group. FLT3-ITD was found in 11 patients (18.3%), NPM1 mutations were found in 19 patients (31.7%), and CEBPA mutations were found in 11 patients (18.3%). In patients with CEBPA mutations, the carrying two simultaneous mutations (CEBPA (double-mut)) was associated with high MPO expression, while the mutant NPM1 without FLT3-ITD genotype was not associated with MPO activity. Both higher MPO expression and the CEBPA (double-mut) genotype appeared to be associated with improved overall survival after intensive chemotherapy. Further studies are required to determine the importance of blast MPO activity as a prognostic factor, especially in CEBPA wild-type patients with a normal karyotype

    The whole blood transcriptional regulation landscape in 465 COVID-19 infected samples from Japan COVID-19 Task Force

    「コロナ制圧タスクフォース」COVID-19患者由来の血液細胞における遺伝子発現の網羅的解析 --重症度に応じた遺伝子発現の変化には、ヒトゲノム配列の個人差が影響する--. 京都大学プレスリリース. 2022-08-23.Coronavirus disease 2019 (COVID-19) is a recently-emerged infectious disease that has caused millions of deaths, where comprehensive understanding of disease mechanisms is still unestablished. In particular, studies of gene expression dynamics and regulation landscape in COVID-19 infected individuals are limited. Here, we report on a thorough analysis of whole blood RNA-seq data from 465 genotyped samples from the Japan COVID-19 Task Force, including 359 severe and 106 non-severe COVID-19 cases. We discover 1169 putative causal expression quantitative trait loci (eQTLs) including 34 possible colocalizations with biobank fine-mapping results of hematopoietic traits in a Japanese population, 1549 putative causal splice QTLs (sQTLs; e.g. two independent sQTLs at TOR1AIP1), as well as biologically interpretable trans-eQTL examples (e.g., REST and STING1), all fine-mapped at single variant resolution. We perform differential gene expression analysis to elucidate 198 genes with increased expression in severe COVID-19 cases and enriched for innate immune-related functions. Finally, we evaluate the limited but non-zero effect of COVID-19 phenotype on eQTL discovery, and highlight the presence of COVID-19 severity-interaction eQTLs (ieQTLs; e.g., CLEC4C and MYBL2). Our study provides a comprehensive catalog of whole blood regulatory variants in Japanese, as well as a reference for transcriptional landscapes in response to COVID-19 infection
