    Classical and molecular epidemiology of campylobacter, in particular Campylobacter jejuni, in the Alberta beef industry

    This research used classical and molecular epidemiology tools to assess the potential importance of feedlot cattle as Campylobacter reservoirs. The project was conducted from November 2004 to September 2005 in southern Alberta. Fresh pen-floor fecal samples were collected from commercial feedlot cattle near slaughter weight in seven feedlots. Overall, 87% of 2,776 fecal samples were culture positive for Campylobacter species (86% of 1,400 in winter, 88% of 1,376 in summer), and 69% of 1,486 Campylobacter positive isolates were identified as Campylobacter jejuni. After accounting for clustering within pen and feedlot, the number of days-on-feed and feedlot size were associated (p ¡Ü 0.05) with Campylobacter species isolation rates. Retail ground beef was collected from 60 grocery stores (four chains, three cities). None of the 1,200 packages were culture positive for Campylobacter species. Polymerase chain reaction (PCR) results from a subset of samples (n=142) indicated that 48% of packages were positive for Campylobacter DNA. By species, 14.8% (21/142), 26.8% (38/142) and 1.4% (2/142) of packages were PCR positive for C. jejuni, C. coli and C. hyointestinalis DNA, respectively. The collection period (1, 2, 3 or 4) was associated (p ¡Ü 0.05) with the odds of detecting Campylobacter species DNA using PCR. Oligonucleotide DNA microarrays were used as a platform for comparative genomic hybridization (CGH) analysis of 87 C. jejuni isolates (46 bovine, 41 human) obtained within the same geographical regions and time frame. Of the 13 CGH clusters identified based on overall comparative genomic profile similarity, nine contained human and cattle isolates, three contained only human isolates, and one contained only cattle isolates. In addition, human clinical and feedlot cattle C. jejuni isolates were compared on a gene-by-gene basis and only a small number of the 1,399 genes tested were unequally distributed between the two groups (p ¡Ü 0.05). The high isolation rates of Campylobacter species and C. jejuni reported here may have implications for food safety, public health and environmental contamination. Our findings suggest that feedlot cattle and human C. jejunistrains are very similar and may be endemic within southern Alberta

    Genes coding for virulence determinants of Campylobacter jejuni in human clinical and cattle isolates from Alberta, Canada, and their potential role in colonization of poultry

    Forty nine Campylobacter jejuni isolates from cattle feces collected from Alberta feedlots and 50 clinical C. jejuni isolates from people in Alberta were tested for the presence of 14 genes encoding putative virulence factors by PCR. These included genes implicated in adherence and colonization (flaC, cadF, docC, racR, jlpA, peb1, and dnaJ), invasion (virB11, ciaB, pldA, and iamA) and protection against harsh conditions (htrA, cbrA, and sodB). The genes examined were widely distributed in both the cattle fecal isolates and the human isolates. Of the isolates tested, 67% contained all of the genes except virB11. The cadF gene was found in 100% of the isolates tested. The presence or absence of virulence-associated genes was not associated with the ability of the organism to colonize birds. All of the C. jejuni isolates used to challenge birds were able to colonize the animals regardless of virulence gene profile. While some diversity in the profile of the occurrence of virulence-associated genes in C. jejuni exists, the distribution of these putative virulence-associated genes isolates from feedlotcattle feces and humans in Alberta was similar. In addition it was not possible to predict the ability of the selected isolates tocolonize young chicks based on the presence of these genes coding for virulence determinants.     Prevalence and molecular characterization of Clostridium difficile isolated from feedlot beef cattle upon arrival and mid-feeding period

    <p>Abstract</p> <p>Background</p> <p>The presence of indistinguishable strains of <it>Clostridium difficile </it>in humans, food animals and food, as well as the apparent emergence of the food-animal-associated ribotype 078/toxinotype V as a cause of community-associated <it>C. difficile </it>infection have created concerns about the potential for foodborne infection. While studies have reported <it>C. difficile </it>in calves, studies of cattle closer to the age of harvest are required. Four commercial feedlots in Alberta (Canada) were enrolled for this study. Fecal samples were collected at the time of arrival and after acclimation (< 62, 62-71 or > 71 days on feed). Selective culture for <it>Clostridium difficile </it>was performed, and isolates were characterized by ribotyping and pulsed-field gel electrophoresis. A logistic regression model was built to investigate the effect of exposure to antimicrobial drugs on the presence of <it>C. difficile</it>.</p> <p>Results</p> <p><it>Clostridium difficile </it>was isolated from 18 of 539 animals at the time of feedlot arrival (CI = 2.3-6.1) and from 18 of 335 cattle at mid-feeding period (CI = 2.9-13.1). Overall, there was no significant difference in the prevalence of <it>C. difficile </it>shedding on arrival versus mid-feeding period (<it>P </it>= 0.47). No association between shedding of the bacterium and antimicrobial administration was found (<it>P </it>= 0.33). All the isolates recovered were ribotype 078, a toxinotype V strain with genes encoding toxins A, B and CDT. In addition, all strains were classified as NAP7 by pulsed field gel electrophoresis (PFGE) and had the characteristic 39 base pairs deletion and upstream truncating mutation on the <it>tcd</it>C gene.</p> <p>Conclusions</p> <p>It is apparent that <it>C. difficile </it>is carried in the intestinal tracts of a small percentage of feedlot cattle arriving and later in the feeding period and that ribotype 078/NAP7 is the dominant strain in these animals. Herd management practices associated with <it>C. difficile </it>shedding were not identified, however further studies of the potential role of antimicrobials on <it>C. difficile </it>acquisition and shedding are required.</p

    DNA methylation patterns of behavior-related gene promoter regions dissect the gray wolf from domestic dog breeds

    A growing body of evidence highlights the relationship between epigenetics, especially DNA methylation, and population divergence as well as speciation. However, little is known about how general the phenomenon of epigenetics-wise separation of different populations is, or whether population assignment is, possible based on solely epigenetic marks. In the present study, we compared DNA methylation profiles between four different canine populations: three domestic dog breeds and their ancestor the gray wolf. Altogether, 79 CpG sites constituting the 65 so-called CpG units located in the promoter regions of genes affecting behavioral and temperamental traits (COMT, HTR1A, MAOA, OXTR, SLC6A4, TPH1, WFS1)-regions putatively targeted during domestication and breed selection. Methylation status of buccal cells was assessed using EpiTYPER technology. Significant inter-population methylation differences were found in 52.3% of all CpG units investigated. DNA methylation profile-based hierarchical cluster analysis indicated an unambiguous segregation of wolf from domestic dog. In addition, one of the three dog breeds (Golden Retriever) investigated also formed a separate, autonomous group. The findings support that population segregation is interrelated with shifts in DNA methylation patterns, at least in putative selection target regions, and also imply that epigenetic profiles could provide a sufficient basis for population assignment of individuals

Morley ([email protected])Cattle Enrolled in Study: September 2007-January 2010.Escherichia coli was isolated from the feces of beef cattle from four commercial feedlots in Alberta, Canada. Each individual was sampled twice, once upon arrival to the feedlot, and again at ≥33 days on feed (DOF). A total of 2,725 isolates were collected from 923 individuals. Antibiotic susceptibility of these isolates to 19 different antimicrobial drugs was determined by broth microdilution and/or disk diffusion assay. Prevalence of resistance to each drug, by sampling time, was calculated (see Figure 2 in associated publication).     Antimicrobial Resistance in Escherichia coli Recovered from Feedlot Cattle and Associations with Antimicrobial Use.

    The objectives of this study were to estimate the prevalence of antimicrobial resistance (AMR) and to investigate the associations between exposures to antimicrobial drugs (AMDs) and AMR in fecal non-type specific Escherichia coli (NTSEC) recovered from a large population of feedlot cattle. Two-stage random sampling was used to select individually identified cattle for enrollment, which were sampled at arrival and then a second time later in the feeding period. Advanced regression techniques were used to estimate resistance prevalences, and to investigate associations between AMD exposures in enrolled cattle and penmates and AMR identified in NTSEC recovered from the second sample set. Resistance was most commonly detected to tetracycline, streptomycin, and sulfisoxazole, and was rarely identified for critically important AMDs. All cattle were exposed to AMDs in feed, and 45% were treated parenterally. While resistance prevalence generally increased during the feeding period, most AMD exposures were not significantly associated with AMR outcomes. Exposures of enrolled cattle to tetracycline were associated with increased resistance to tetracycline and trimethoprim sulfa, while beta-lactam exposures were associated with decreased likelihood of detecting streptomycin resistance. Pen-level AMD exposure measures were not associated with resistance outcomes. These findings suggest that tetracycline treatment of feedlot cattle can be associated with modest increases in risk for recovery of resistant NTSEC, but the numerous treatments with an advanced macrolide (tulathromycin) were not associated with detectable increases in resistance in NTSEC. All cattle were exposed to in-feed treatments of tetracycline and this could limit the ability to identify the full impact of these exposures, but these exposures varied for enrolled cattle varied, providing an opportunity to evaluate a dose response. While AMD exposures were not associated with detectably increased risks for resistance to critically important AMDs, rare resistance outcomes and infrequent exposure to other important AMDs (e.g., cephalosporins) limited our ability to rigorously investigate questions regarding factors that can influence resistance to these important AMDs
