    RBR E3 ubiquitin ligases: new structures, new insights, new questions

    The RBR (RING-BetweenRING-RING) or TRIAD [two RING fingers and a DRIL (double RING finger linked)] E3 ubiquitin ligases comprise a group of 12 complex multidomain enzymes. This unique family of E3 ligases includes parkin, whose dysfunction is linked to the pathogenesis of early-onset Parkinson's disease, and HOIP (HOIL-1-interacting protein) and HOIL-1 (haem-oxidized IRP2 ubiquitin ligase 1), members of the LUBAC (linear ubiquitin chain assembly complex). The RBR E3 ligases share common features with both the larger RING and HECT (homologous with E6-associated protein C-terminus) E3 ligase families, directly catalysing ubiquitin transfer from an intrinsic catalytic cysteine housed in the C-terminal domain, as well as recruiting thioester-bound E2 enzymes via a RING domain. Recent three-dimensional structures and biochemical findings of the RBRs have revealed novel protein domain folds not previously envisioned and some surprising modes of regulation that have raised many questions. This has required renaming two of the domains in the RBR E3 ligases to more accurately reflect their structures and functions: the C-terminal Rcat (required-for-catalysis) domain, essential for catalytic activity, and a central BRcat (benign-catalytic) domain that adopts the same fold as the Rcat, but lacks a catalytic cysteine residue and ubiquitination activity. The present review discusses how three-dimensional structures of RBR (RING1-BRcat-Rcat) E3 ligases have provided new insights into our understanding of the biochemical mechanisms of these important enzymes in ubiquitin biology. The views of Skármeta, Allende and others are reported and a list of possible Post-Boom characteristics is suggested. It is argued that there are difficulties in the way of relating the Post-Boom easily to Postmodernism, but that the notion of Post-colonialism may prove helpful in future criticism

    A hybrid computer solution of the swing equation

    Models for the governor and exciter control systems are presented and the development of a hybrid computing system including these models is described. A technique for including the governor and exciter effects into the solution of the power system swing equation is also discussed. A hybrid computing system consisting of an SCC 650 digital computer, hybrid interface, and an EAI-TR-48 analog computer is used to study five test cases. The test cases involve basically the same example system with different values for fault clearing times and machine inertias. The results of each test case are briefly discussed with relevance to the importance of the inclusion of the control system effects.     On the Dark Side: Conrad\u27s The Secret Sharer and Valenzuela\u27s La palabra asesino

    Conrad’s famous “The Secret Sharer” and the short story “La palabra asesino” [“The Word ‘Killer’” in its English translation] by the Argentine Luisa Valenzuela both concern psychological self-exploration and self-discovery, through contact with a killer, a situation which challenges conventional moral standards. It is suggested that a comparison between the two stories may throw reciprocal light on both of them. In each story an act or acts of murder becomes a trigger which sets off a train of psychological events, somewhat different in the two cases. Discussion of the differences highlights the authors\u27 priorities and the significance they attach to the darker side of the human personality. Both stories are highly ambiguous; but the ambiguity serves a different purpose in each case. Conrad is concerned with psychological doubling ; Valenzuela with exploration of aspects of the human personality which in turn may be related to aspects of Argentina\u27s collective personality as it expressed itself during the “Dirty War.” An examination of the different forces in play in the two stories improves our understanding of both

    Sources of age determination errors for sablefish (Anoplopoma fimbria)

    This study was undertaken to resolve problems in age determination of sablefish (Anoplopoma fimbria). Aging of this species has been hampered by poor agreement (averaging less than 45%) among age readers and by differences in assigned ages of as much as 15 years. Otoliths from fish that had been injected with oxytetracycline (OTC) and that had been at liberty for known durations were used to determine why age determinations were so difficult and to help determine the correct aging procedure. All fish were sampled from Oregon southwards, which represents the southern part of their range. The otoliths were examined with the aid of image processing. Some fish showed little or no growth on the otolith after eight months at liberty, whereas otoliths from other fish grew substantially. Some fish lay down two prominent hyaline zones within a single year, one in the summer and one in the winter. We classified the otoliths by morphological type and found that certain types are more likely to lay down multiple hyaline zones and other types are likely to lay down little or no zones. This finding suggests that some improvement could be achieved by detailed knowledge of the growth characteristics of the different types. This study suggests that it may not be possible to obtain reliable ages from sablefish otoliths. At the very least, more studies will be required to under-stand the growth of sablefish otoliths
