    Prevalence and Genotypes of Mycobacterium Avium Subspecies Paratuberculosis in Large Ruminants of Eastern Uttar Pradesh, North India

    Uttar Pradesh is the fourth largest, most populous and leading milk and meat producing state in India. Despite the huge livestock population, information on the status of paratuberculosis homogeneity and heterogeneity of Mycobacterium avium subspecies paratuberculosis (MAP) isolates of eastern Uttar Pradesh is non-existent. Present study was aimed to estimate the presence of MAP in large ruminants (Cattle and Buffaloes) of eastern Uttar Pradesh. A total 108 fecal samples were collected from farmer's herds of large ruminants (cattle and buffaloes) from different geographical regions (Chandauli, Mughalsarai, Gazipur, and Naugarh) of eastern Uttar Pradesh and screened for the presence of MAP infection using microscopic examination, direct IS900 PCR and culture on Herrold egg yolk (HEY) medium. The isolates recovered on HEY medium were subjected to molecular identification and genotyping using IS900 PCR and IS1311 PCR-REA method, respectively. Of the 108 fecal samples, 25 (23.14%) and 11 (10.18%) samples were positive for the presence of acid-fast bacilli and growth on HEY medium, respectively. Species-wise, 17.5, 7.5% and 26.5, 11.7% fecal samples from cattle and buffaloes were found positive for the presence of acid-fast bacilli and growth on HEY medium, respectively. Isolates recovered on HEY medium with mycobactin J were positive for IS900 sequence and genotyped as Bison Type using IS1311 PCR-REA method. Present study is the first report on the presence of MAP infection and ‘Bison Type' genotype of MAP in eastern Uttar Pradesh. These findings will be useful for the intervention of effective control measures in order to reduce the prevalence of MAP infection in domestic livestock species and prevent its spread to the human population in the regions

    Antibodies Elicited in Response to EBNA-1 May Cross-React with dsDNA

    Several genetic and environmental factors have been linked to Systemic Lupus Erythematosus (SLE). One environmental trigger that has a strong association with SLE is the Epstein Barr Virus (EBV). Our laboratory previously demonstrated that BALB/c mice expressing the complete EBNA-1 protein can develop antibodies to double stranded DNA (dsDNA). The present study was undertaken to understand why anti-dsDNA antibodies arise during the immune response to EBNA-1.In this study, we demonstrated that mouse antibodies elicited in response to EBNA-1 cross-react with dsDNA. First, we showed that adsorption of sera reactive with EBNA-1 and dsDNA, on dsDNA cellulose columns, diminished reactivity with EBNA-1. Next, we generated monoclonal antibodies (MAbs) to EBNA-1 and showed, by several methods, that they also reacted with dsDNA. Examination of two cross-reactive MAbs--3D4, generated in this laboratory, and 0211, a commercial MAb--revealed that 3D4 recognizes the carboxyl region of EBNA-1, while 0211 recognizes both the amino and carboxyl regions. In addition, 0211 binds moderately well to the ribonucleoprotein, Sm, which has been reported by others to elicit a cross-reactive response with EBNA-1, while 3D4 binds only weakly to Sm. This suggests that the epitope in the carboxyl region may be more important for cross-reactivity with dsDNA while the epitope in the amino region may be more important for cross-reactivity with Sm.In conclusion, our results demonstrate that antibodies to the EBNA-1 protein cross-react with dsDNA. This study is significant because it demonstrates a direct link between the viral antigen and the development of anti-dsDNA antibodies, which are the hallmark of SLE. Furthermore, it illustrates the crucial need to identify the epitopes in EBNA-1 responsible for this cross-reactivity so that therapeutic strategies can be designed to mask these regions from the immune system following EBV exposure

    Determining crystal structures through crowdsourcing and coursework

    We show here that computer game players can build high-quality crystal structures. Introduction of a new feature into the computer game Foldit allows players to build and real-space refine structures into electron density maps. To assess the usefulness of this feature, we held a crystallographic model-building competition between trained crystallographers, undergraduate students, Foldit players and automatic model-building algorithms. After removal of disordered residues, a team of Foldit players achieved the most accurate structure. Analysing the target protein of the competition, YPL067C, uncovered a new family of histidine triad proteins apparently involved in the prevention of amyloid toxicity. From this study, we conclude that crystallographers can utilize crowdsourcing to interpret electron density information and to produce structure solutions of the highest quality

    Comparative Analysis of KNN and C5.0 Algorithm for Smart City Classification

    Many governments are adopting smart city concept so as to improve living standards. Big data analytics is one of the main technologies that are able to enhance smart city services. As almost everything is becoming digitalized, a lot of data is being collected that can be beneficial in various domains. Various classifications algorithms have been developed in the last decade. Many of them have been compared. This paper presents a comparison between two classification algorithms: KNN and decision tree (C5.0). This paper is aimed to compare two or more algorithm\u27s results. The results of algorithms are central task in areas such as machine learning. For analyzing the result obtained when comparing the algorithms, the best tool used is RStudio which provides a platform for loading data to produce plots and tables

    Structural, dielectric and multiferroic properties of Er and La substituted BiFeO<SUB>3</SUB> ceramics

    Erbium (Er) and lanthanum (La) substituted BiFeO3 (BFO) ceramics have been prepared through conventional solid solution route. X-ray diffraction data indicated a gradual phase transition from rhombohedral to monoclinic structure in Bi0.9-x La0.1Erx FeO3 (x = 0.05, 0.07 and 0.1) (BLEFOx = 0.05, 0.07, 0.1) ceramics. Differential thermal analysis (DTA) measurements of BFO samples showed a ferroelectric transition at 835°C, whereas it is shifted to 792°C for BLEFO x = 0.1. The Raman spectra of BLEFOx = 0.05, 0.07, 0.1 samples showed the shift of Raman modes to higher wavenumbers and suppression of A1 modes indicating decrease in ferroelectricity. The Raman spectra also indicated the structural transformation due to Er and La substitution in BFO. On subsequent erbium doping, the intrinsic dielectric constant is found to decrease from 68 (for pure BFO) to 52 for BLEFOx = 0.05 to 43 for BLEFOx = 0.07 but increased to 89 for BLEFOx = 0.1 when compared to pure BFO. The increase in Er content resulted in the increase in spontaneous magnetization (0.1178 emu/g at 8T for BLEFOx = 0.1) due to collapse of spin cycloid structure. Ferroelectric remnant polarization of BLEFOx = 0.05 and BLEFOx = 0.07 decreases when compared to pure BFO while small remnant polarization (close to paraelectric behaviour) is evident for BLEFOx = 0.1

    Hypothyroidism and depression: Are cytokines the link?

    Context: Primary hypothyroidism has been thought of as an inflammatory condition characterized by raised levels of cytokines such as C-reactive protein (CRP), interleukin-6 (IL-6), and tumor necrosis factor-alpha (TNF-α). Depression is also well known to occur in hypothyroidism. Depression is also characterized by elevated inflammatory cytokines. We planned to study whether cytokines play an important part in linking these two conditions. Objectives: (1) To know the prevalence of depression in overt hypothyroidism due to autoimmune thyroid disease. (2) To correlate the levels of inflammatory markers with the occurrence of depression. (3) To study the effect of levothyroxine on inflammatory markers and depression. Materials and Methods: In this longitudinal, case–controlled study, 33 patients with autoimmune hypothyroidism (thyroid-stimulating hormone >10 uIU/ml) were included with 33 age-, sex-, and body max index-matched healthy controls. Individuals were tested for Serum TNF-α, IL-6, high-sensitivity-CRP (hs-CRP). They were assessed for depression using Montgomery Asberg Depression Rating Scale (MADRS) and World Health Organization Quality of Life (QOL) Scale. Patients received L Thyroxine titrated to achieve euthyroidism and were reassessed for inflammatory markers and cognitive dysfunction. Results: Nineteen patients (57%) had mild to moderate depression (MADRS >11). After 6 months of treatment, eight patients (42%) had remission of depression with significant improvement in QOL scores (P < 0.05). TNF-α, IL-6, and hs-CRP were significantly elevated in patients compared with controls and reduced with therapy but did not reach baseline as controls. The change in inflammatory markers correlated with improvement in QOL scores in social and environmental domains (P < 0.01). Conclusions: Primary autoimmune hypothyroidism is an inflammatory state characterized by elevated cytokines which decline with LT4 therapy. It is associated with depression and poor quality of life. Treatment of hypothyroidism results in alleviation of depression in the majority of patients. Similarly, patients with mild to moderate depression should be tested for hypothyroidism as simple treatment may ameliorate their depression and improves MADRS score and QOL

    BackgroundOmicron, a new variant of Severe Acute Respiratory Syndrome-Coronavirus 2 (SARS-CoV-2), was first detected in November 2021. This was believed to be highly transmissible and was reported to evade immunity. As a result, an urgent need was felt to screen all positive samples so as to rapidly identify Omicron cases and isolate them to prevent the spread of infection. Genomic surveillance of SARS-CoV-2 was planned to correlate disease severity with the genomic profile.MethodsAll the SARS-CoV-2 positive cases detected in the state of Rajasthan were sent to our Lab. Samples received from 24 November 2021 to 4 January 2022 were selected for Next-Generation Sequencing (NGS). Processing was done as per protocol on the Ion Torrent S5 System for 1,210 samples and bioinformatics analysis was done.ResultsAmong the 1,210 samples tested, 762 (62.9%) were Delta/Delta-like and other lineages, 291 (24%) were Omicron, and 157 (12.9%) were invalid or repeat samples. Within a month, the proportion of Delta and other variants was reversed, 6% Omicron became 81%, and Delta and other variants became 19%, initially all Omicron cases were seen in international travelers and their contacts but soon community transmission was seen. The majority of patients with Omicron were asymptomatic (56.7%) or had mild disease (33%), 9.2% had moderate symptoms, and two (0.7%) had severe disease requiring hospitalization, of which one (0.3%) died and the rest were (99.7%) recovered. History of vaccination was seen in 81.1%, of the previous infection in 43.2% of cases. Among the Omicron cases, BA.1 (62.8%) was the predominant lineage followed by BA.2 (23.7%) and B.1.529 (13.4%), rising trends were seen initially for BA.1 and later for BA.2 also. Although 8.9% of patients with Delta lineage during that period were hospitalized, 7.2% required oxygen, and 0.9% died. To conclude, the community spread of Omicron occurred in a short time and became the predominant circulating variant; BA.1 was the predominant lineage detected. Most of the cases with Omicron were asymptomatic or had mild disease, and the mortality rate was very low as compared to Delta and other lineages.</p
