    Liquid film formation: prediction accuracy of different numerical approaches

    In counteracting fouling phenomenon in gas turbines, which leads to system inefficiencies and performance degradation, water washing technique is very often adopted. Water droplets sprays are injected and, hitting the solid surfaces, remove the dirt deposition. Among the collateral undesirable phenomena related to water washing, blades erosion and liquid film formation are the most remarkable. Despite the former issue was extensively assessed by the authors in previous works, up to the authors' knowledge the risk of liquid film formation due to water washing was scarcely investigated. Liquid film formation and spreading on a solid surface is a complex phenomenon involving a large number of physical events, such as: droplets impact on a solid surface, splashing phenomena, liquid film dragging under the effect of the carrier phase and droplets separation from the film in proximity of geometry discontinuities. In this paper, an extensively used experimental test case involving all these phenomena was used to test different numerical wall film models available in literature. The test case consists in the injection of a liquid jet in a high velocity crossflow. Some of the liquid jet mass impacts on the opposite solid surface generating a wall film which develops under the dragging effect of the crossflow. A Lagrangian approach was used to track the suspended droplets within the flow field by also considering the turbulent dispersion by means of a Random Walk model. Droplets-wall interaction is considered according to the Stanton-Rutland model, which provides the outcome of a collision (deposit, rebound or splashing), depending on the local impact conditions. If a droplet sticks on a solid boundary, a liquid film generates. Droplets atomization is also accounted for by using the Madabhushi model while Friederich separation model was selected to take into account the detachment of droplets from the film at the geometry edge. Three different numerical simulations have been performed based on different approaches used to solve the liquid film evolution, namely Eulerian one-way coupling, Eulerian two-way coupling and Lagrangian two-way coupling. Numerical results have been compared with the experimental ones from both a qualitative and a quantitative point of view. The wall film shape, its spatial distribution and the variation of the film thickness of the wall centreline have been compared between experimental and numerical simulations proving that the Lagrangian 2-way coupling approach better reproduces the liquid film dynamics observed in the experiments

    Il paper presenta i risultati di una ricerca-azione sul tema della povertà energetica nelle aree interne. L’indagine è stata promossa e condotta dallo Spi-Cgil (Sindacato dei pensionati italiani), in collaborazione con la Fondazione Giuseppe Di Vittorio. L’indagine sul campo è stata condotta su tutto il territorio nazionale attraverso un disegno di campionamento per quote a due stadi: i comuni periferici e ultra-periferici, selezionati con procedura casuale, come unità di primo stadio; le persone di età maggiore 64 anni residenti negli stessi comuni, come unità di secondo stadio. Gli scopi della ricerca sono: i) fornire strumenti interpretativi per definire, misurare e contrastare il fenomeno della povertà energetica; ii) indagare la vulnerabilità e la povertà economica ed energetica nelle aree periferiche ed ultra-periferiche quali ambiti privilegiati per la costruzione di pratiche innovative di investimento sociale place-based e di nuovo welfare locale da cui ripartire per rispondere ai nuovi bisogni di protezione dei soggetti più fragili; iii) rafforzare il ruolo della contrattazione sociale e territoriale dei sindacati nella lotta alla povertà energetica e nella costruzione di una transizione ecologica giusta

    Investigation of antibacterial activity of new classes of essential oils derivatives

    Essential oils (EOs) have deserved much attention in the past decades for their antimicrobial activity, since many of them have demonstrated efficacy against food-borne pathogenic and spoilage microorganisms. Moreover, they have potential application in animal nutrition as multifunctional feed supplements, avoiding or diminishing the use of antibiotics in livestock. However, low solubility and bioavailability as well as volatility and marked aromatic note are important limitations in food and feed applications. In this study we present the synthesis, characterization and evaluation of the antibacterial activity of new thymol, carvacrol and menthol derivatives. The new compounds have been designed to overcome the limitations of the precursors, such as poor water solubility and volatility, still maintaining a good antimicrobial profile. We evaluated the activity of the synthetized compounds against pathogens causing important foodborne diseases, . i.e. . Clostridium perfringens, . Salmonella typhimurium, . Salmonella enteritidis and . Escherichia coli. The low MICs and MBCs values for some of the studied compounds, combined with water solubility and negligible cytotoxicity towards HT-29 human cells, confirmed the potential use for EOs derivatives in the food industry

    GERIATRIC NEPHROLOGY: AN OVERVIEW

    Introduction: Chronic kidney disease (CKD) is a highly prevalent condition and its prevalence is increasing worldwide, particularly in adults aged ≥ 70 years. Epidemiological studies showed that as many as 20–54% of the older adults suffer from CKD in stages 3-5. Nevertheless the question whether this lower eGFR is a consequence of kidney disease or if it is the result of a physiological aging is still debated, even if it implies a reduced renal reserve and vulnerability to drugs overdose with increased risk of acute kidney injury (AKI). Materials and methods: PubMed search was conducted for available English literature, describing the actual knowledge about specific and frequent issues reported in the acute and chronic kidney disease in older adults. Prospective and retrospective studies, as well as meta-analyses and latest systematic reviews were included. Results:Most of the studies examined and reviewed were discarded for wrong population or intervention or deemed unfit. Only 103 met the inclusion criteria for the review. The studies included in the review were grouped into two areas: chronic and acute kidney disease in older adults and we have analysed the peculiar and frequently found issues in this population. Conclusions: The geriatric population is increasing worldwide.We should consider peculiar aspects of this population, such as sarcopenia, malnutrition, psychological and cognitive deficits and increased risk of AKI, in order to reach a good quality of life, with improved doctor / patient relationship, a greater adherence to therapy, a reduction in health care costs, and if possible, adequate "end of life", as far as it is approved by the patient and his family. The achievement of these objectives requires an organized work in multidisciplinary teams that evaluate overall the geriatric patient

    Pr-favoured variants of the bacteriophytochrome from the plant pathogen Xanthomonas campestris hint on light regulation of virulence-associated mechanisms

    Red/far-red light-sensing bacteriophytochrome photoreceptor (BphP) pathways play key roles in bacterial physiology and ecology. These bilin-binding proteins photoswitch between two states, Pr (red absorbing) and Pfr (far-red absorbing). The isomerization of the chromophore and the downstream structural changes result in the light signal transduction. The agricultural pathogen Xanthomonas campestris pv. campestris (Xcc) code for a single bathy-like type BphP (XccBphP), previously shown to negatively regulate several light-mediated biological processes involved in virulence. Here, we generated three different full-length variants with single amino acid changes within its GAF domain that affect the XccBphP photocycle favouring its Pr state: L193Q, L193N and D199A. While D199A recombinant protein locks XccBphP in a Pr-like state, L193Q and L193N exhibit a significant enrichment of the Pr form in thermal equilibrium. The X-ray crystal structures of the three variants were solved, resembling the wild-type protein in the Pr state. Finally, we studied the effects of altering the XccBphP photocycle on the exopolysaccharide xanthan production and stomatal aperture assays as readouts of its bacterial signalling pathway. Null-mutant complementation assays show that the photoactive Pr-favoured XccBphP variants L193Q and L193N tend to negatively regulate xanthan production in vivo. In addition, our results indicate that strains expressing these variants also promote stomatal apertures in challenged plant epidermal peels, compared to wild-type Xcc. The findings presented in this work provide new evidence on the Pr state of XccBphP as a negative regulator of the virulence-associated mechanisms by light in Xcc.Fil: Conforte, Valeria Paola.     Role of SIRT-3, p-mTOR and HIF-1\u3b1 in Hepatocellular Carcinoma Patients Affected by Metabolic Dysfunctions and in Chronic Treatment with Metformin

    The incidence of hepatocellular carcinoma deriving from metabolic dysfunctions has increased in the last years. Sirtuin- (SIRT-3), phospho-mammalian target of rapamycin (p-mTOR) and hypoxia-inducible factor- (HIF-1\u3b1) are involved in metabolism and cancer. However, their role in hepatocellular carcinoma (HCC) metabolism, drug resistance and progression remains unclear. This study aimed to better clarify the biological and clinical function of these markers in HCC patients, in relation to the presence of metabolic alterations, metformin therapy and clinical outcome. A total of 70 HCC patients were enrolled: 48 and 22 of whom were in early stage and advanced stage, respectively. The expression levels of the three markers were assessed by immunohistochemistry and summarized using descriptive statistics. SIRT-3 expression was higher in diabetic than non-diabetic patients, and in metformin-treated than insulin-treated patients. Interestingly, p-mTOR was higher in patients with metabolic syndrome than those with different etiology, and, similar to SIRT-3, in metformin-treated than insulin-treated patients. Moreover, our results describe a slight, albeit not significant, benefit of high SIRT-3 and a significant benefit of high nuclear HIF-1\u3b1 expression in early-stage patients, whereas high levels of p-mTOR correlated with worse prognosis in advanced-stage patients. Our study highlighted the involvement of SIRT-3 and p-mTOR in metabolic dysfunctions that occur in HCC patients, and suggested SIRT-3 and HIF-1\u3b1 as predictors of prognosis in early-stage HCC patients, and p-mTOR as target for the treatment of advanced-stage HCC

    Structural basis for the Pr-Pfr long-range signaling mechanism of a full-length bacterial phytochrome at the atomic level

    Phytochromes constitute a widespread photoreceptor family that typically interconverts between two photostates called Pr (red light-absorbing) and Pfr (far-red light-absorbing). The lack of full-length structures solved at the (near-)atomic level in both pure Pr and Pfr states leaves gaps in the structural mechanisms involved in the signal transmission pathways during the photoconversion. Here, we present the crystallographic structures of three versions from the plant pathogen Xanthomonas campestris virulence regulator XccBphP bacteriophytochrome, including two full-length proteins, in the Pr and Pfr states. The structures show a reorganization of the interaction networks within and around the chromophore-binding pocket, an α-helix/β-sheet tongue transition, and specific domain reorientations, along with interchanging kinks and breaks at the helical spine as a result of the photoswitching, which subsequently affect the quaternary assembly. These structural findings, combined with multidisciplinary studies, allow us to describe the signaling mechanism of a full-length bacterial phytochrome at the atomic level.Fil: Otero, Lisandro Horacio.     Low-protein diets for chronic kidney disease patients: The Italian experience

    open20Nutritional treatment has always represented a major feature of CKD management. Over the decades, the use of nutritional treatment in CKD patients has been marked by several goals. The first of these include the attainment of metabolic and fluid control together with the prevention and correction of signs, symptoms and complications of advanced CKD. The aim of this first stage is the prevention of malnutrition and a delay in the commencement of dialysis. Subsequently, nutritional manipulations have also been applied in association with other therapeutic interventions in an attempt to control several cardiovascular risk factors associated with CKD and to improve the patient's overall outcome.     Phosphorylation of SRSF1 is modulated by replicational stress

    DNA ligase I-deficient 46BR.1G1 cells show a delay in the maturation of replicative intermediates resulting in the accumulation of single- and double-stranded DNA breaks. As a consequence the ataxia telangiectasia mutated protein kinase (ATM) is constitutively phosphorylated at a basal level. Here, we use 46BR.1G1 cells as a model system to study the cell response to chronic replication-dependent DNA damage. Starting from a proteomic approach, we demonstrate that the phosphorylation level of factors controlling constitutive and alternative splicing is affected by the damage elicited by DNA ligase I deficiency. In particular, we show that SRSF1 is hyperphosphorylated in 46BR.1G1 cells compared to control fibroblasts. This hyperphosphorylation can be partially prevented by inhibiting ATM activity with caffeine. Notably, hyperphosphorylation of SRSF1 affects the subnuclear distribution of the protein and the alternative splicing pattern of target genes. We also unveil a modulation of SRSF1 phosphorylation after exposure of MRC-5V1 control fibroblasts to different exogenous sources of DNA damage. Altogether, our observations indicate that a relevant aspect of the cell response to DNA damage involves the post-translational regulation of splicing factor SRSF1 which is associated with a shift in the alternative splicing program of target genes to control cell survival or cell death
