The low intestinal and hepatic toxicity of hydrolyzed fumonisin B1 correlates with its inability to alter the metabolism of sphingolipids

Fumonisins are mycotoxins frequently found as natural contaminants in maize, where they are produced by the plant pathogen Fusarium verticillioides. They are toxic to animals and exert their effects through mechanisms involving disruption of sphingolipid metabolism.Fumonisin B1 (FB1) is the predominant fumonisins in this family. FB1 is converted to its hydrolyzed analogs HFB1, by alkaline cooking (nixtamalization) or through enzymatic degradation. The toxicity of HFB1 is poorly documented especially at the intestinal level. The objectives of this study were to compare the toxicity of HFB1 and FB1 and to assess the ability of these toxins to disrupt sphingolipids biosynthesis. HFB1 was obtained by a deesterification of FB1, with a carboxylesterase. Piglets, animals highly sensitive to FB1, were exposed by gavage for 2 weeks to 2.8 µmol FB1 or HFB1/kg body weight/day. FB1 induced hepatotoxicity as indicated by the lesion score, the level of several biochemical analytes and the expression of inflammatory cytokines. Similarly, FB1 impaired the morphology of the different section of the small intestine, reduced villi height and modified intestinal cytokine expression. By contrast, HFB1 did not trigger hepatotoxicity, did not impair intestinal morphology and slightly modified the intestinal immune response. This low toxicity of HFB1 correlates with a weak alteration of the sphinganine/sphingosine ratio in the liver and in the plasma. Taken together, these data demonstrate that HFB1 does not cause intestinal or hepatic toxicity in the sensitive pig model, and slightly disrupts sphingolipids metabolism. This finding suggests that conversion to HFB1 could be a good strategy to reduce FB1 exposure

ENQUANTO A AVENTURA NÃO VEM...: CONTRIBUIÇÕES DAS LIVES DA CORRENTE DA AVENTURA PARA O CAMPO DO LAZER

The Adventure Chain is an online initiative, created by a group of researchers of adventure activities in the field of leisure, which precedes the face-to-face events related to the Brazilian and International Congresses of Adventure Activities, which were postponed due to the isolation measures necessary to contain the current COVID-19 pandemic. This initiative, in interface with the virtual environment, sought to open spaces for the continuation of reflections in the area, minimizing losses and impacts. Thus, this study, of a qualitative nature, aimed to analyze the content expressed in the Adventure Chain, involving adventure activities in the scope of leisure, contributing to the dissemination of knowledge in the area. Data were descriptively analyzed, through Content Analysis and the results indicate the concern with the pedagogical aspects followed by professional training, as topics of greater relevance discussed in the lives.  La Corriente de la Aventura es una iniciativa online, creada por un grupo de investigadores de actividades de aventura en el campo del ocio, que precede a los eventos presenciales relacionados con los Congresos Brasileños e Internacionales de Actividades de Aventura, los cuales fueron pospuestos debido a las medidas de aislamiento necesarias para contener la actual pandemia de COVID-19. Esta iniciativa, en interfaz con el entorno virtual, buscó abrir espacios para la continuación de las reflexiones en la área, minimizando pérdidas e impactos. Así, este estudio, de carácter cualitativo, tuvo como objetivo analizar los contenidos expresados ​​en la Corriente de la Aventura, involucrando actividades de aventura en el ámbito del ocio, contribuyendo a la difusión del conocimiento en el área. Los datos fueron analizados de manera descriptiva, mediante Análisis de Contenido y los resultados indican la preocupación por los aspectos pedagógicos seguida de la formación profesional, como temas de mayor relevancia discutidos en las lives.A Corrente da Aventura é uma iniciativa online, criada por um grupo de pesquisadores de atividades de aventura no campo do lazer, a qual antecede os eventos presenciais referentes aos Congressos Brasileiro e Internacional de Atividades de Aventura, que foram adiados em decorrência das medidas de isolamento social, necessárias para a contenção da atual pandemia de COVID-19. Clinical report : one year of treatment of Proteus syndrome with miransertib (ARQ 092)

A 20-yr-old man with Proteus syndrome (PS) and somatic mosaicism of the AKT1 c.49G > A p.(E17K) variant had asymmetric overgrowth of the right frontal and facial bones, asymmetric spinal overgrowth with thoracolumbar scoliosis, dilatation of the inferior vena cava, testicular cystadenoma, bilateral knee deformities, macrodactyly, and apparent intellectual disability. Miransertib (ARQ 092) is an oral, allosteric, selective pan-AKT inhibitor initially developed for cancer therapeutics, now being evaluated for the treatment of PS. After baseline evaluation, the patient started unblinded treatment of 10 mg oral miransertib daily (∼5 mg/m2/day), escalated to 30 mg daily (∼15 mg/m2/day), and then to 50 mg daily (∼25 mg/m2/day) after 3 mo of treatment. Adverse events included dry mouth, one episode of gingivostomatitis, and loose, painful dentition due to preexisting periodontal disease, all of which resolved spontaneously. After 11 mo of treatment, the patient reported improved general well-being, increased mobility of the ankle, spine, and hands, a subjective decrease in size of the right facial bone overgrowth, and reduced areas of cerebriform connective tissue nevi on the soles. Whole-body MRI findings were stable without apparent disease progression. We conclude that 1 yr of treatment with miransertib was beneficial in this case

Usabilidade de site sobre lazer para idosos

Introduction: With the increasing use of the Internet, increased demands for interactive interfaces that meet user expectations. Even with all the developments, there is still a need to improve the quality of information available on the web, especially when it comes to sites aimed at older people. Objective: Therefore, this study, qualitative in nature, aimed at evaluating the usability of a site that contains information about leisure activities for seniors, taking into focus the dimensions, layout, and operation information. Method: For this purpose, we used the Survey Site Usability on Leisure (IAUSLA-21 +) applied to a purposive sample consisted of 30 elderly, male and female students of a digital inclusion project in São Paulo. Data were analyzed descriptively using Thematic Content Analysis. Results: The results showed more strengths than weaknesses in relation to the usability of the site, especially with regard to the layout and information. Conclusion: The site had good results also in relation to user satisfaction. Suggest a few adjustments to be made ​​operational in the size of the instrument and therefore further studies to deepen this theme.Com a crescente utilização da internet, aumentaram as exigências por interfaces interativas, que atendam às expectativas dos usuários. Mesmo com toda a evolução, ainda há necessidade de melhorar a qualidade das informações disponibilizadas na web, especialmente no que tange aos sites voltados para pessoas idosas. Sendo assim, este estudo, de natureza qualitativa, teve como objetivo avaliar a usabilidade de um site que contém informações sobre lazer para idosos, tomando-se em foco as dimensões: layout, informação e operacionalização. Para tanto, utilizou-se como instrumento o Inventário de Usabilidade de Sites sobre Lazer (IAUSLA-21+), aplicado a uma amostra intencional composta por 30 idosos, de ambos os sexos, alunos de um projeto de inclusão digital da cidade de São Paulo. Os dados foram analisados descritivamente por meio de Análise de Conteúdo Temático. Os resultados apontaram mais pontos positivos do que negativos em relação à usabilidade do site, principalmente, no que se refere ao layout e à informação. O site apresentou bons resultados também quanto à satisfação dos usuários. Sugerem-se algumas adaptações na dimensão operacionalização do instrumento e, consequentemente, novos estudos que visem aprofundar essa temática

Non-disulfide-Bridge peptide 5.5 from the Scorpion Hadrurus gertschi Inhibits the growth of mycobacterium abscessus subsp. massiliense

Multi-drug resistant microorganisms have been a growing concern during the last decades due to their contribution in mortality rates worldwide. Antimicrobial peptides (AMPs) are broad spectrum antimicrobial agents that display potent microbicidal activity against a wide range of microorganisms. AMPs generally have a rapid mode of action that reduces the risk of resistance developing among pathogens. In this study, an AMP derived from scorpion venom, NDBP-5.5, was evaluated against Mycobacterium abscessus subsp. massiliense, a rapidly growing and emerging pathogen associated with healthcare infections. The minimal bactericidal concentration of NDBP-5.5, AMP quantity necessary to stop bacteria visible growth, against M. abscessus subsp. massiliense was 200 μM, a concentration that did not induce hemolysis of human red blood cells. The therapeutic index was 3.05 indicating a drug with low toxicity and therefore good clinical potential. Treatment of infected macrophages with NDBP-5.5 or clarithromycin presented similar results, reducing the bacterial load. M. abscessus subsp. massiliense-infected animals showed a decrease in the bacterial load of up to 70% when treated with NDBP-5.5. These results revealed the effective microbicidal activity of NDBP-5.5 against Mycobacterium, indicating its potential as an antimycobacterial agent
