    Aerosol and Cloud Interaction Observed From High Spectral Resolution Lidar Data

    Recent studies utilizing satellite retrievals have shown a strong correlation between aerosol optical depth (AOD) and cloud cover. However, these retrievals from passive sensors are subject to many limitations, including cloud adjacency (or 3D) effects, possible cloud contamination, uncertainty in the AOD retrieval. Some of these limitations do not exist in High Spectral Resolution Lidar (HSRL) observations; for instance, HSRL observations are not a ected by cloud adjacency effects, are less prone to cloud contamination, and offer accurate aerosol property measurements (backscatter coefficient, extinction coefficient, lidar ratio, backscatter Angstrom exponent,and aerosol optical depth) at a neospatial resolution (less than 100 m) in the vicinity of clouds. Hence, the HSRL provides an important dataset for studying aerosol and cloud interaction. In this study, we statistically analyze aircraft-based HSRL profiles according to their distance from the nearest cloud, assuring that all profile comparisons are subject to the same large-scale meteorological conditions. Our results indicate that AODs from HSRL are about 17% higher in the proximity of clouds (approximately 100 m) than far away from clouds (4.5 km), which is much smaller than the reported cloud 3D effect on AOD retrievals. The backscatter and extinction coefficients also systematically increase in the vicinity of clouds, which can be explained by aerosol swelling in the high relative humidity (RH) environment and/or aerosol growth through in cloud processing (albeit not conclusively). On the other hand, we do not observe a systematic trend in lidar ratio; we hypothesize that this is caused by the opposite effects of aerosol swelling and aerosol in-cloud processing on the lidar ratio. Finally, the observed backscatter Angstrom exponent (BAE) does not show a consistent trend because of the complicated relationship between BAE and RH.     Pfam is a database of protein families that currently contains 7973 entries (release 18.0). A recent development in Pfam has enabled the grouping of related families into clans. Pfam clans are described in detail, together with the new associated web pages. Improvements to the range of Pfam web tools and the first set of Pfam web services that allow programmatic access to the database and associated tools are also presented.     A preliminary report of an educational intervention in practice management

    BACKGROUND: Practice management education continues to evolve, and little information exists regarding its curriculum design and effectiveness for resident education. We report the results of an exploratory study of a practice management curriculum for primary care residents. METHODS: After performing a needs assessment with a group of primary care residents at Wright State University, we designed a monthly seminar series covering twelve practice management topics. The curriculum consisted of interactive lectures and practice-based application, whenever possible. We descriptively evaluated two cognitive components (practice management knowledge and skills) and the residents' evaluation of the curriculum. RESULTS: The mean correct on the knowledge test for this group of residents was 74% (n = 12) and 91% (n = 12) before and after the curriculum, respectively. The mean scores for the practice management skill assessments were 2.62 before (n = 12), and 3.65 after (n = 12) the curriculum (modified Likert, 1 = strongly disagree, 5 = strongly agree). The residents rated the curriculum consistently high. CONCLUSIONS: This exploratory study suggests that this curriculum may be useful in developing knowledge and skills in practice management for primary care residents. This study suggests further research into evaluation of this curriculum may be informative for practice-based education

    Targeting tumour re-wiring by triple blockade of mTORC1, epidermal growth factor, and oestrogen receptor signalling pathways in endocrine-resistant breast cancer

    Background Endocrine therapies are the mainstay of treatment for oestrogen receptor (ER)-positive (ER+) breast cancer (BC). However, resistance remains problematic largely due to enhanced cross-talk between ER and growth factor pathways, circumventing the need for steroid hormones. Previously, we reported the anti-proliferative effect of everolimus (RAD001-mTORC1 inhibitor) with endocrine therapy in resistance models; however, potential routes of escape from treatment via ERBB2/3 signalling were observed. We hypothesised that combined targeting of three cellular nodes (ER, ERBB, and mTORC1) may provide enhanced long-term clinical utility. Methods A panel of ER+ BC cell lines adapted to long-term oestrogen deprivation (LTED) and expressing ESR1wt or ESR1Y537S, modelling acquired resistance to an aromatase-inhibitor (AI), were treated in vitro with a combination of RAD001 and neratinib (pan-ERBB inhibitor) in the presence or absence of oestradiol (E2), tamoxifen (4-OHT), or fulvestrant (ICI182780). End points included proliferation, cell signalling, cell cycle, and effect on ER-mediated transactivation. An in-vivo model of AI resistance was treated with monotherapies and combinations to assess the efficacy in delaying tumour progression. RNA-seq analysis was performed to identify changes in global gene expression as a result of the indicated therapies. Results Here, we show RAD001 and neratinib (pan-ERBB inhibitor) caused a concentration-dependent decrease in proliferation, irrespective of the ESR1 mutation status. The combination of either agent with endocrine therapy further reduced proliferation but the maximum effect was observed with a triple combination of RAD001, neratinib, and endocrine therapy. In the absence of oestrogen, RAD001 caused a reduction in ER-mediated transcription in the majority of the cell lines, which associated with a decrease in recruitment of ER to an oestrogen-response element on the TFF1 promoter. Contrastingly, neratinib increased both ER-mediated transactivation and ER recruitment, an effect reduced by the addition of RAD001. In-vivo analysis of an LTED model showed the triple combination of RAD001, neratinib, and fulvestrant was most effective at reducing tumour volume. Gene set enrichment analysis revealed that the addition of neratinib negated the epidermal growth factor (EGF)/EGF receptor feedback loops associated with RAD001. Conclusions Our data support the combination of therapies targeting ERBB2/3 and mTORC1 signalling, together with fulvestrant, in patients who relapse on endocrine therapy and retain a functional ER

    The Luminosity Function and Color-Magnitude Diagram of the Globular Cluster M12

    In this paper we present the V and I luminosity functions and color-magnitude diagrams derived from wide-field (23 arcmin by 23 arcmin) BVI photometry of the intermediate metallicity ([Fe/H]=-1.3) Galactic globular cluster M12. Using observed values (and ranges of values) for the cluster metallicity, reddening, distance modulus, and age we compare these data to recent alpha-enhanced stellar evolution models for low mass metal-poor stars. We describe several methods of making comparisons between theoretical and observed luminosity functions in order to isolate the evolutionary timescale information the luminosity functions contain. We find no significant evidence of excesses of stars on the red giant branch, although the morphology of the subgiant branch in the observed luminosity function does not match theoretical predictions in a satisfactory way. Current uncertainties in Teff-color transformations (and possibly also in other physics inputs to the models) make more detailed conclusions about the subgiant branch morphology impossible. Given the recent constraints on cluster ages from the WMAP experiment (Spergel et al. 2003), we find that good fitting models that do not include He diffusion (both color-magnitude diagrams and luminosity functions) are too old (by approximately 1-2 Gyr) to adequately represent the cluster luminosity function. The inclusion of helium diffusion in the models provides an age reduction (compared to non-diffusive models) that is consistent with the age of the universe being 13.7+/-0.2 Gyr (Bennett et al. 2003).Comment: 63 pages, 29 figures, accepted for ApJ.     Distances and ages of NGC 6397, NGC 6752 and 47 Tuc

    Distances and absolute ages for the globular clusters NGC6397, NGC6752, and 47 Tuc are obtained using Main Sequence Fitting. Accurate reddenings and metal abundances (errors of 0.005 mag and 0.04 dex) are obtained by comparing B-V, b-y colours and UVES spectra for clusters TO-stars and subgiants with similar data for Hipparcos subdwarfs, using a reddening free temperature indicator (Halpha). Errors in distances and ages are 0.07 mag and 1 Gyr. NGC6397 and NGC6752 have ages of 13.5\pm 1.1 and 13.4\pm 1.1 Gyr, while 47 Tuc is probably about 2.6 Gyr younger, in agreement with results obtained by techniques sensitive to relative ages. Considering possible systematic errors, the age of the oldest globular clusters is 13.4\pm 0.8\pm 0.6 Gyr (random and systematic errors). This age is fully compatible with WMAP results, and indicates that the oldest Galactic globular clusters formed within the first 1.7 Gyr after the Big Bang, corresponding to a redshift of z\geq 2.5, in a standard LambdaCDM model. The epoch of formation of the (inner halo) globular clusters lasted about 2.6 Gyr, ending at a time corresponding to a redshift of z\geq 1.3. Once combined with values of Ho given by WMAP and by the HST Key Project, our age estimate provides a robust upper limit at 95% level of confidence of Omega_M<0.57, indicating the need for a vacuum energy independent of results from type Ia SNe and clusters of galaxies. Using cluster distances, we derive a value of M_V(HB)=(0.22\pm 0.05)([Fe/H]+1.5)+(0.56\pm 0.07) for the zero point of the relation between the horizontal branch absolute magnitude and metallicity. When combined with the apparent average luminosity of the RR Lyrae stars in the LMC by Clementini et al. (2003), this zero point provides a new estimate of the distance modulus to the LMC: (m-M)o=18.50\pm 0.09.Comment: 16 pages, 5 encapsulated figures, accepted for publication in A&

    Fission-fusion dynamics over large distances in raven non-breeders

    The influence of fission-fusion dynamics, i.e., temporal variation in group size and composition, on social complexity has been studied in large-brained mammals that rely on social bonds. Little is known about birds, even though some species like ravens have recently received attention for their socio-cognitive skills and use of social bonds. While raven breeders defend territories year-round, non-breeders roam through large areas and form groups at food sources or night roosts. We here examined the fission-fusion patterns of non-breeding ravens over years, investigating whether birds meet repeatedly either at the same or at different locations. We combined four large datasets: presence-absence observations from two study sites (Austria, Italy) and GPS-tracking of ravens across two study areas (Austria, France). As expected, we found a highly dynamic system in which individuals with long phases of temporary settlement had a high probability of meeting others. Although GPS-tagged ravens spread out over thousands of square kilometres, we found repeated associations between almost half of the possible combinations at different locations. Such a system makes repeated interactions between individuals at different sites possible and likely. High fission-fusion dynamics may thus not hinder but shape the social complexity of ravens and, possibly, other long-term bonded birds

    The International Surface Pressure Databank version 2

    The International Surface Pressure Databank (ISPD) is the world's largest collection of global surface and sea-level pressure observations. It was developed by extracting observations from established international archives, through international cooperation with data recovery facilitated by the Atmospheric Circulation Reconstructions over the Earth (ACRE) initiative, and directly by contributing universities, organizations, and countries. The dataset period is currently 1768–2012 and consists of three data components: observations from land stations, marine observing systems, and tropical cyclone best track pressure reports. Version 2 of the ISPD (ISPDv2) was created to be observational input for the Twentieth Century Reanalysis Project (20CR) and contains the quality control and assimilation feedback metadata from the 20CR. Since then, it has been used for various general climate and weather studies, and an updated version 3 (ISPDv3) has been used in the ERA-20C reanalysis in connection with the European Reanalysis of Global Climate Observations project (ERA-CLIM). The focus of this paper is on the ISPDv2 and the inclusion of the 20CR feedback metadata. The Research Data Archive at the National Center for Atmospheric Research provides data collection and access for the ISPDv2, and will provide access to future versions
