The mRNA cap-binding protein eIF4E in post-transcriptional gene expression

    Get PDF
Eukaryotic initiation factor 4E (eIF4E) has central roles in the control of several aspects of post-transcriptional gene expression and thereby affects developmental processes. It is also implicated in human diseases. This review explores the relationship between structural, biochemical and biophysical aspects of eIF4E and its function in vivo, including both long-established roles in translation and newly emerging ones in nuclear export and mRNA decay pathways

Single and multisensor Field Effect Transistors FET with a pH-sensitive Si/SiO2/SizN4/TazOs-gate and reference electrode (for single semsor) were developed and used for manufacturing the following BIO-FETs: for glucose analysis: Glucose Oxidase-FETs (GOD-FET), for penicillin G and V analysis: Penicillin G Amidase- and Penicillinase-FETs, for urea analysis: Urease-FET, and for cephalosporin C analysis: Cephalosporinase-FET. The GOD-FETswereintegrated into the FIA-system of Eppendorf (EVA) and used for monitoring the glucose concentration in microbial cultivation and production processes with recombinant_Escherichia coli K12 MF, recombinant Escherichia coli JM103, Saccharomyces cerevisiae H620, and Candida boidinii. Urease-FET-FIA was used to monitor the urea concentration in a simulatedcultivation of Cephalosporium acremonium and UREASE-FET-FIA and GOD-FET-FIA for the monitoring of urea and glucose concentrations in simulated Saccharomyces cerevisiae cultivations

New Aminoacyl-tRNA Synthetase-like Protein in Insecta with an Essential Mitochondrial Function*♦

Aminoacyl-tRNA synthetases (ARS) are modular enzymes that aminoacylate transfer RNAs (tRNA) for their use by the ribosome during protein synthesis. ARS are essential and universal components of the genetic code that were almost completely established before the appearance of the last common ancestor of all living species. This long evolutionary history explains the growing number of functions being discovered for ARS, and for ARS homologues, beyond their canonical role in gene translation. Here we present a previously uncharacterized paralogue of seryl-tRNA synthetase named SLIMP (seryl-tRNA synthetase-like insect mitochondrial protein). SLIMP is the result of a duplication of a mitochondrial seryl-tRNA synthetase (SRS) gene that took place in early metazoans and was fixed in Insecta. Here we show that SLIMP is localized in the mitochondria, where it carries out an essential function that is unrelated to the aminoacylation of tRNA. The knockdown of SLIMP by RNA interference (RNAi) causes a decrease in respiration capacity and an increase in mitochondrial mass in the form of aberrant mitochondria
