    "Biste etwa einer von den roten Gebhardts?" - Identities between Family and Society in Klaus Kordon's "Trilogie der Wendepunkte"

    This thesis deals with the "Trilogie der Wendepunkte" ("Trilogy of Turning Points") by Klaus Kordon. The trilogy, consisting of "Die roten Matrosen" (1984), "Mit dem Rücken zur Wand" (1990), and "Der erste Frühling" (1993), tells the story of a working-class family in Berlin during the first half of the twentieth century. In the light of the important historic events taking place at that time, the adolescent protagonists are faced with the values and expectations of their family on the one hand and society on the other hand. This thesis examines their individual search for identity and how they position themselves in public and private spaces. First of all, I will clarify that the novels originally written for an adolescent audience can not only be analysed as educational and pedagogical literature but have characteristics of other genres as well. Therefore, different literary methods will be applied in order to analyse the protagonists’ identity construction. For the first part of the thesis the function of the family regarding the construction of identity will be examined. Values and moral beliefs are transferred from one generation to the other and therefore shape the individual’s identity. Especially the function of family memory and storytelling are crucial in the process of creating identity, therefore Jan and Aleida Assmann’s memory studies as well as Paul RicÏur’s concept of narrative identity will be adapted. The second part focuses on society’s role in creating identity. Social class and status as well as public spaces like school and workplace shape the individual’s identity. Considering the role theory of Lothar Krappmann, this chapter explores how the protagonists take social roles as part of their collective and personal identity. Moreover, I will apply the stages of psychosocial development by Erik H. Erikson to the protagonists’ lives and investigate how their family identity helps them to assert their identity in public space. The concluding chapter discusses the impact of differing individual needs, social requirements, and familial values on the identity construction and how the protagonists deal with conflicting expectations

    The muddled governance of state-imposed forced labour: multinational corporations, states, and cotton from China and Uzbekistan

    Forced labour persists in our global economy despite dedicated attention and eradication efforts from both the public and private sectors. Given the bounded reach and lack of enforcement by states and international organisations, the private sector has been the linchpin for eradication of forced labour globally. Utilising the brand-to-state boomerang model, this paper examines state-imposed forced labour in cotton production in China and Uzbekistan, and grapples with how interests–those of the states and the multinational corporations involved in forced labour–shape private governance outcomes. By investigating state-imposed forced labour in China (specifically, the Xinjiang Uyghur Autonomous Region) and Uzbekistan, we find that multinational corporations are reluctant to work towards eradicating forced labour when their net sales and profit are threatened by doing so. Building on the stream of international political economy research regarding how interests complicate governance effectiveness, we expose a gap in the literature on the impact of state-imposed forced labour on governance outcomes and illuminate global ramifications

    Absence of ectopic epithelial inclusions in 3,904 axillary lymph nodes examined in sentinel technique

    Intraoperative examination of sentinel axillary lymph nodes can be done by imprint cytology, frozen section, or, most recently, by PCR-based amplification of a cytokeratin signal. Using this technique, benign epithelial inclusions, representing mammary tissue displaced along the milk line, will likely generate a positive PCR signal and lead to a false-positive diagnosis of metastatic disease. To better appreciate the incidence of ectopic epithelial inclusions in axillary lymph nodes, we have performed an autopsy study, examining on 100ÎĽm step sections3,904 lymph nodes obtained from 160 axillary dissections in 80 patients. The median number of lymph nodes per axilla was 23 (15, 6, and 1 in levels 1, 2, and 3, respectively). A total of 30,450 hematoxylin-eosin stained slides were examined, as well as 8,825 slides immunostained with pan-cytokeratin antibodies. Despite this meticulous work-up, not a single epithelial inclusion was found in this study, suggesting that the incidence of such inclusions is much lower than the assumed 5% reported in the literatur

    Oxidation of p53 through DNA Charge Transport Involves a Network of Disulfides within the DNA-Binding Domain

    Transcription factor p53 plays a critical role in the cellular response to stress stimuli. We have seen that p53 dissociates selectively from various promoter sites as a result of oxidation at long-range through DNA-mediated charge transport (CT). Here, we examine this chemical oxidation and determine the residues in p53 that are essential for oxidative dissociation, focusing on the network of cysteine residues adjacent to the DNA-binding site. Of the eight mutants studied, only the C275S mutation shows decreased affinity for the Gadd45 promoter site. However, both mutations C275S and C277S result in substantial attenuation of oxidative dissociation, with C275S causing the most severe attenuation. Differential thiol labeling was used to determine the oxidation states of cysteine residues within p53 after DNA-mediated oxidation. Reduced cysteines were iodoacetamide-labeled, whereas oxidized cysteines participating in disulfide bonds were ^(13)C_2D_2-iodoacetamide-labeled. Intensities of respective iodoacetamide-modified peptide fragments were analyzed by mass spectrometry. A distinct shift in peptide labeling toward ^(13)C_2D_2-iodoacetamide-labeled cysteines is observed in oxidized samples, confirming that chemical oxidation of p53 occurs at long range. All observable cysteine residues trend toward the heavy label under conditions of DNA CT, indicating the formation of multiple disulfide bonds among the cysteine network. On the basis of these data, it is proposed that disulfide formation involving C275 is critical for inducing oxidative dissociation of p53 from DNA

    Laying Eyes on Headlights: Eye Movements Suggest Facial Features in Cars

    Humans’ proneness to see faces even in inanimate structures such as cars has long been noticed, yet empirical evidence is scarce. To examine this tendency of anthropomorphism, participants were asked to compare specific features (such as the eyes) of a face and a car front presented next to each other. Eye movement patterns indicated on which visual information participants relied to solve the task and clearly revealed the perception of facial features in cars, such as headlights as eyes or grille as nose. Most importantly, a predominance of headlights was found in attracting and guiding people’s gaze irrespective of the feature they were asked to compare – equivalent to the role of the eyes during face perception. This response to abstract configurations is interpreted as an adaptive bias of the respective inherent mechanism for face perception and is evolutionarily reasonable with regard to a »better safe than sorry« strategy

    Perfusion CT best predicts outcome after radioembolization of liver metastases: a comparison of radionuclide and CT imaging techniques

    Objective: To determine the best predictor for the response to and survival with transarterial radioembolisation (RE) with 90yttrium microspheres in patients with liver metastases. Methods: Forty consecutive patients with liver metastases undergoing RE were evaluated with multiphase CT, perfusion CT and 99mTc-MAA SPECT. Arterial perfusion (AP) from perfusion CT, HU values from the arterial (aHU) and portal venous phase (pvHU) CT, and 99mTc-MAA uptake ratio of metastases were determined. Morphologic response was evaluated after 4months and available in 30 patients. One-year survival was calculated with Kaplan-Meier curves. Results: We found significant differences between responders and non-responders for AP (P 20ml/100ml/min had a significantly (P = 0.01) higher 1-year survival, whereas an aHU value >55 HU did not discriminate survival (P = 0.12). The Cox proportional hazard model revealed AP as the only significant (P = 0.02) independent predictor of survival. Conclusion: Compared to arterial and portal venous enhancement and the 99mTc-MAA uptake ratio of liver metastases, the AP from perfusion CT is the best predictor of morphologic response to and 1-year survival with RE.     A Pro-Drug Approach for Selective Modulation of AI-2-Mediated Bacterial Cell-to-Cell Communication

    The universal quorum sensing autoinducer, AI-2, is utilized by several bacteria. Analogs of AI-2 have the potential to modulate bacterial behavior. Selectively quenching the communication of a few bacteria, in the presence of several others in an ecosystem, using analogs of AI-2 is non-trivial due to the ubiquity of AI-2 processing receptors in many bacteria that co-exist. Herein, we demonstrate that when an AI-2 analog, isobutyl DPD (which has been previously shown to be a quorum sensing, QS, quencher in both Escherichia coli and Salmonella typhimurium) is modified with ester groups, which get hydrolyzed once inside the bacterial cells, only QS in E. coli, but not in S. typhimurium, is inhibited. The origin of this differential QS inhibition could be due to differences in analog permeation of the bacterial membranes or ester hydrolysis rates. Such differences could be utilized to selectively target QS in specific bacteria amongst a consortium of other species that also use AI-2 signaling

    Efficacy and Toxicity of Different Chemotherapy Protocols for Concurrent Chemoradiation in Non-Small Cell Lung Cancer—A Secondary Analysis of the PET Plan Trial

    (1) Background: The optimal chemotherapy (CHT) regimen for concurrent chemoradiation (cCRT) is not well defined. In this secondary analysis of the international randomized PET-Plan trial, we evaluate the efficacy of different CHT. (2) Methods: Patients with inoperable NSCLC were randomized at a 1:1 ratio regarding the target volume definition and received isotoxically dose-escalated cCRT using cisplatin 80 mg/m2 (day 1, 22) and vinorelbin 15 mg/m2 (day 1, 8, 22, 29) (P1) or cisplatin 20 mg/m2 (day 1–5, 29–33) and vinorelbin 12.5 mg/m2 (day 1, 8, 15, 29, 36, 43) (P2) or carboplatin AUC1 (day 1–5, 29–33) and vinorelbin 12.5 mg/m2 (day 1, 8, 15, 29, 36, 43) (P3) or other CHT at the treating physician’s discretion. (3) Results: Between 05/2009 and 11/2016, 205 patients were randomized and 172 included in the per-protocol analysis. Patients treated in P1 or P2 had a better overall survival (OS) compared to P3 (p = 0.015, p = 0.01, respectively). Patients treated with carboplatin had a worse OS compared to cisplatin (HR 1.78, p = 0.03), but the difference did not remain significant after adjusting for age, ECOG, cardiac function creatinine and completeness of CHT. (4) Conclusions: Carboplatin doublets show no significant difference compared to cisplatin, after adjusting for possibly relevant factors, probably due to existing selection bias

    A time-resolved proteomic and prognostic map of COVID-19.

    COVID-19 is highly variable in its clinical presentation, ranging from asymptomatic infection to severe organ damage and death. We characterized the time-dependent progression of the disease in 139 COVID-19 inpatients by measuring 86 accredited diagnostic parameters, such as blood cell counts and enzyme activities, as well as untargeted plasma proteomes at 687 sampling points. We report an initial spike in a systemic inflammatory response, which is gradually alleviated and followed by a protein signature indicative of tissue repair, metabolic reconstitution, and immunomodulation. We identify prognostic marker signatures for devising risk-adapted treatment strategies and use machine learning to classify therapeutic needs. We show that the machine learning models based on the proteome are transferable to an independent cohort. Our study presents a map linking routinely used clinical diagnostic parameters to plasma proteomes and their dynamics in an infectious disease
