Validierung des Endlosschleifentetsts (EST) unter power sowie work-limit Vorgabe
Ziel der vorliegenden Diplomarbeit ist die Validierung des Endlosschleifentests (EST) unter der power sowie work-limit Instruktion. Eine zentrale Fragestellung ist, ob der Endlosschleifentest (EST), welcher das räumliche Vorstellungsvermögen erfasst, und unter der power Bedingung als Rasch-Homogen gilt, auch unter der work-limit Bedingung Eindimensionalität im Sinne von Rasch aufweist. Außerdem wird der Frage nachgegangen, ob die work-limit Vorgabe, die im Gegensatz zur power Vorgabe eine Zeitdruckkomponente enthält, zusätzliche Informationen, bezüglich der Leistung der Versuchspersonen liefert, sowie ob es aufgrund der work-limit Bedingung höhere korrelative Bezüge zu bestimmten Außenkriterien gibt. Die work-limit Bedingung des EST ist mit den strengen Kriterien des Rasch-Modells vereinbar, jedoch konnte durch die work-limit Bedingung im Bezug auf die Leistungen der Versuchspersonen keine zusätzlich diagnostisch relevanten Informationen gewonnen werden. Bezüglich der Außenkriterien, lassen die Ergebnisse darauf schliessen, dass sich die beiden Vorgabebedingungen nur geringfügig in der Höhe der Korrelationen unterscheiden.The objective of this thesis is to validate the Endless-loops-test (ger.: Endlosschleifentest – EST; Gittler & Arendasy, 2003) under the power and the work-limit condition. The EST determines the spatial ability of the test person. It has been examined before that under the power condition the EST can be described as Rasch homogeneous. The key question to be answered is whether or not the same holds true under the work-limit condition. Furthermore it was tested if the EST results under the work-limit condition reveal additional information about the performance of the test person. In addition it was examined if higher correlative scores can be found in respect of certain external criteria. The work-limit condition of the EST indeed is Rasch homogeneous – even under the strict criteria of the Rasch model. However the predictive capability of the performance of the test persons under the work-limit condition of the EST did not improve in a meaningful way. Regarding the external criteria the results conclude that the correlation between the power and the work-limit condition differ only in a minor way
Decompactification of space or time in large N QCD_2
QCD_2 with fundamental quarks on a cylinder is solved to leading order in the
1/N expansion, including the zero mode gluons. As a result of the
non-perturbative dynamics of these gauge degrees of freedom, the compact
space-time direction gets effectively decompactified. In a thermodynamic
interpretation, this implies that there is no pressure of order N and that the
chiral condensate of order N is temperature independent. These findings are
consistent with confinement of quarks, rule out both chiral and deconfining
phase transitions in the finite temperature 't Hooft model, and help to resolve

Biocomputational prediction of non-coding RNAs in model cyanobacteria
<p>Abstract</p> <p>Background</p> <p>In bacteria, non-coding RNAs (ncRNA) are crucial regulators of gene expression, controlling various stress responses, virulence, and motility. Previous work revealed a relatively high number of ncRNAs in some marine cyanobacteria. However, for efficient genetic and biochemical analysis it would be desirable to identify a set of ncRNA candidate genes in model cyanobacteria that are easy to manipulate and for which extended mutant, transcriptomic and proteomic data sets are available.</p> <p>Results</p> <p>Here we have used comparative genome analysis for the biocomputational prediction of ncRNA genes and other sequence/structure-conserved elements in intergenic regions of the three unicellular model cyanobacteria <it>Synechocystis </it>PCC6803, <it>Synechococcus elongatus </it>PCC6301 and <it>Thermosynechococcus elongatus </it>BP1 plus the toxic <it>Microcystis aeruginosa </it>NIES843. The unfiltered numbers of predicted elements in these strains is 383, 168, 168, and 809, respectively, combined into 443 sequence clusters, whereas the numbers of individual elements with high support are 94, 56, 64, and 406, respectively. Removing also transposon-associated repeats, finally 78, 53, 42 and 168 sequences, respectively, are left belonging to 109 different clusters in the data set. Experimental analysis of selected ncRNA candidates in <it>Synechocystis </it>PCC6803 validated new ncRNAs originating from the <it>fabF-hoxH </it>and <it>apcC-prmA </it>intergenic spacers and three highly expressed ncRNAs belonging to the Yfr2 family of ncRNAs. Yfr2a promoter-<it>luxAB </it>fusions confirmed a very strong activity of this promoter and indicated a stimulation of expression if the cultures were exposed to elevated light intensities.</p> <p>Conclusion</p> <p>Comparison to entries in Rfam and experimental testing of selected ncRNA candidates in <it>Synechocystis </it>PCC6803 indicate a high reliability of the current prediction, despite some contamination by the high number of repetitive sequences in some of these species. In particular, we identified in the four species altogether 8 new ncRNA homologs belonging to the Yfr2 family of ncRNAs. Modelling of RNA secondary structures indicated two conserved single-stranded sequence motifs that might be involved in RNA-protein interactions or in the recognition of target RNAs. Since our analysis has been restricted to find ncRNA candidates with a reasonable high degree of conservation among these four cyanobacteria, there might be many more, requiring direct experimental approaches for their identification.</p
Clinical forensic radiology in strangulation victims: forensic expertise based on magnetic resonance imaging (MRI) findings
Based on only one objective and several subjective signs, the forensic classification of strangulation incidents concerning their life-threatening quality can be problematic. Reflecting that it is almost impossible to detect internal injuries of the neck with the standard forensic external examination, we examined 14 persons who have survived manual and ligature strangulation or forearm choke holds using MRI technique (1.5-T scanner). Two clinical radiologists evaluated the neck findings independently. The danger to life was evaluated based on the "classical” external findings alone and in addition to the radiological data. We observed hemorrhaging in the subcutaneous fatty tissue of the neck in ten cases. Other frequent findings were hemorrhages of the neck and larynx muscles, the lymph nodes, the pharynx, and larynx soft tissues. Based on the classical forensic strangulation findings with MRI, eight of the cases were declared as life-endangering incidents, four of them without the presence of petechial hemorrhage but with further signs of impaired brain function due to hypoxia. The accuracy of future forensic classification of the danger to life will probably be increased when it is based not only on one objective and several subjective signs but also on the evidence of inner neck injuries. However, further prospective studies including larger cohorts are necessary to clarify the value of the inner neck injuries in the forensic classification of surviving strangulation victim
AAV Vectors for FRET-Based Analysis of Protein-Protein Interactions in Photoreceptor Outer Segments
Fluorescence resonance energy transfer (FRET) is a powerful method for the detection and quantification of stationary and dynamic protein-protein interactions. Technical limitations have hampered systematic in vivo FRET experiments to study protein-protein interactions in their native environment. Here, we describe a rapid and robust protocol that combines adeno-associated virus (AAV) vector-mediated in vivo delivery of genetically encoded FRET partners with ex vivo FRET measurements. The method was established on acutely isolated outer segments of murine rod and cone photoreceptors and relies on the high co-transduction efficiency of retinal photoreceptors by co-delivered AAV vectors. The procedure can be used for the systematic analysis of protein-protein interactions of wild type or mutant outer segment proteins in their native environment. Conclusively, our protocol can help to characterize the physiological and pathophysiological relevance of photoreceptor specific proteins and, in principle, should also be transferable to other cell types
Emergence of Skyrme crystal in Gross-Neveu and 't Hooft models at finite density
We study two-dimensional, large field theoretic models (Gross-Neveu
model, 't Hooft model) at finite baryon density near the chiral limit. The same
mechanism which leads to massless baryons in these models induces a breakdown
of translational invariance at any finite density. In the chiral limit baryonic
matter is characterized by a spatially varying chiral angle with a wave number
depending only on the density. For small bare quark masses a sine-Gordon kink
chain is obtained which may be regarded as simplest realization of the Skyrme
crystal for nuclear matter. Characteristic differences between confining and

Finishing the euchromatic sequence of the human genome
The sequence of the human genome encodes the genetic instructions for human physiology, as well as rich information about human evolution. In 2001, the International Human Genome Sequencing Consortium reported a draft sequence of the euchromatic portion of the human genome. Since then, the international collaboration has worked to convert this draft into a genome sequence with high accuracy and nearly complete coverage. Here, we report the result of this finishing process. The current genome sequence (Build 35) contains 2.85 billion nucleotides interrupted by only 341 gaps. It covers ∼99% of the euchromatic genome and is accurate to an error rate of ∼1 event per 100,000 bases. Many of the remaining euchromatic gaps are associated with segmental duplications and will require focused work with new methods. The near-complete sequence, the first for a vertebrate, greatly improves the precision of biological analyses of the human genome including studies of gene number, birth and death. Notably, the human enome seems to encode only 20,000-25,000 protein-coding genes. The genome sequence reported here should serve as a firm foundation for biomedical research in the decades ahead
