"Gestresste" Mitochondrien werden isoliert : ein Protein schlĂ€gt die BrĂŒcke zwischen QualitĂ€tskontrolle und Dynamik
Mitochondrien sind die Kraftwerke unserer Zellen. In ihnen findet die Zellatmung statt, die unseren Körper mit lebenswichtiger Energie versorgt. ZusĂ€tzlich teilen sich die Zellorganellen und verschmelzen wieder miteinander im Minutentakt. Was aber passiert, wenn Teile dieses dynamischen Geflechts Defekte aufweisen? Die Antwort dazu könnte ein Protein sein, das auf zwei verschiedene Weisen in die Mitochondrien-Membranen eingebaut wird. Liegt keine kurze Form des Proteins vor, ist das ein Hinweis dafĂŒr, dass die Organellen defekt sind. Die Mitochondrien verbrennen die mit der Nahrung zugefĂŒhrten Kohlenhydrate und Fette unter Verbrauch von Sauerstoff zu Kohlendioxid und Wasser. Bei diesem Vorgang, der Zellatmung, wird ĂŒber eine Reihe von Proteinkomplexen ein elektrochemisches Potenzial aufgebaut, das zur Produktion des EnergietrĂ€gers ATP (Adenosintriphosphat) genutzt wird. ATP kann aus den Mitochondrien abtransportiert werden und steht somit als eine Art Treibstoff fĂŒr alle Stoffwechselprozesse zur VerfĂŒgung. Die Arbeit der Mitochondrien ist der Hauptgrund fĂŒr unseren tĂ€glichen Sauerstoffbedarf. AuĂerdem tragen die Nano-Kraftwerke der Zelle dazu bei, unsere Körpertemperatur auf 37 °C aufrechtzuerhalten. Aufgrund dieser zentralen Funktionen ist es nicht verwunderlich, dass eine Reihe von Krankheiten beim Menschen durch den Funktionsverlust von Mitochondrien verursacht oder beeinflusst wird. Das sind in erster Linie neurologische oder muskulĂ€re Erkrankungen, aber auch Diabetes, Fettleibigkeit, verschiedene Formen von Krebs und Alterungsprozesse. Folglich ist es von immenser Bedeutung zu verstehen, wie Mitochondrien funktionieren, wie sie ihre FunktionalitĂ€t aufrechterhalten und gegebenenfalls repariert oder entsorgt werden können. Dem können wir am Wissenschaftsstandort Frankfurt hervorragend nachgehen, da sich einige international ausgewiesene Forschungsgruppen in den Fachbereichen Medizin, Biologie, Chemie und am Max-Planck-Institut fĂŒr Biophysik mit verschiedenen Aspekten der mitochondrialen Biologie befassen. In zahlreichen interdisziplinĂ€ren Kooperationen wird so versucht, dieses komplexe System besser zu verstehen
Diffusions Tensor Bildgebung der Mamma: eine Pilotstudie
Die diffusionsgewichtete Magnetresonanztomographie (Diffusion Weighted
Imaging, DWI) basiert auf der inkohÀrenten Brownschen
Molekularbewegung. Dabei stellt die diffusionsgewichtete MRT eine schnelle und kontrastmittelfreie Methode dar, welche in mehreren Vorarbeiten der letzten Jahre eine hohe Genauigkeit in der Differenzierung und Detektion maligner Neoplasien der Mamma erreicht hat. Der Vorgang der Diffusion im Gewebe ist unterschiedlichen Graden der Isotropie unterworfen, welche sich durch einen Tensor beschreiben lĂ€sst. Wurde in bisherigen Untersuchungen an der Mamma lediglich die gemittelte DiffusivitĂ€t in Form des âApparent Diffusion Coefficientâ (ADC) gemessen, so ist die Messung der zusĂ€tzlichen Information der Isotropie, ausgedrĂŒckt als âfraktionelle Anisotropieâ (FA) viel versprechend hinsichtlich zusĂ€tzlicher Informationen ĂŒber die mikroskopische Beschaffenheit des Gewebes. Die Abbildung wird als âDiffusion Tensor Imagingâ (DTI) bezeichnet. Die vorliegende Studie sollte die Machbarkeit und den klinischen Nutzen der DTI der Mamma zur Detektion und Differentialdiagnose pathologischer LĂ€sionen der Mamma evaluieren. In die vorliegende Untersuchung wurden randomisiert prospektiv 67 Patientinnen mit anreichernden Herdbefunden in der MR-Mammographie eingeschlossen (medianes Alter 54,6a). Alle waren in einem Zeitraum von 11 Monaten durch die Klinik fĂŒr GynĂ€kologie des UniversitĂ€tsklinikums Jena aufgrund unklarer oder suspekter Vorbefunde zur MRM ĂŒberwiesen worden. Der bilateralen dynamischen MRM (1,5T, dynamische T1w FLASH2D Messung vor und nach i.v. Injektion von 0,1mmol/kg KG Gd-DTPA bei 3 ml/s Injektionsrate) folgte die unilaterale DTI Messung (TR 8900 ms, TE 139 ms, 6 Richtungen, zwei Diffusionswichtungen: b=0 s/mm2 und b= 1000 s/mm2). Mittels Scannersoftware wurden parametrische Karten von ADC und FA berechnet. AnschlieĂend erfolgte nach Fusion der anatomischen und DTI Daten die Messung von mehreren Regions-of-Interest (ROI) in anatomisch korrespondierenden Regionen der kontrastmittelverstĂ€rkten T1w
(KM-T1w) sowie DWI Sequenzen. ZunĂ€chst wurde die gesamte LĂ€sion (LĂ€siongesamt), folgend der Teil der LĂ€sion mit am stĂ€rksten herabgesetzter DiffusivitĂ€t (LĂ€sionZentrum) gemessen. Es folgten Messungen in makroskopisch gesundem Parenchym sowie dem Rauschen ausserhalb der Mamma. Ausden normalisierten SignalintensitĂ€ten von LĂ€siongesamt und Parenchym wurde das Kontrast-zu-Rausch-VerhĂ€ltnis (CNR) berechnet und mittels Wilcoxon Test zwischen KM-T1w und DWI verglichen. ADC und FA Werte wurden zwischen benignen und malignen LĂ€sionen sowie Parenchym mittels Mann-Whitney-U Test verglichen. Zur Identifikation unabhĂ€ngiger differentialdiagnostischer PrĂ€diktoren folgte eine schrittweise binĂ€r logistische Regressionsanalyse (RĂŒckwĂ€rts Methode), zur Quantifizierung der diagnostischen Genauigkeit eine Receiver-Operating-Characteristics (ROC) Analyse. In 59 Patientinnen mit 54 malignen und 17 benignen LĂ€sionen wurden erfolgreiche DTI Messungen durchgefĂŒhrt. Das CNR zwischen KM-T1w und DTI war gleichwertig (P=0,191). Sowohl ADC als auch FA Werte unterschieden sich signifikant (P<0,002) zwischen benignen und malignen Herdbefunden. ADC Werte in malignen LĂ€sionen waren dabei niedriger als die entsprechenden Werte in benignen Herden und Parenchym. Im Gegensatz dazu fanden sich niedrigste FA Werte in benignen Herden. Maligne LĂ€sionen zeigten höhere Werte, gefolgt von höchsten FA Werten in gesundem Parenchym. Die Regressionsanalyse identifizierte ADCZentrum als alleinigen unabhĂ€ngigen differentialdiagnostischen PrĂ€diktor. Die Area under the Curve (AUC) wurde mit 0,899 gemessen, es ergaben sich eine SensitivitĂ€t von 92,6% sowie eine SpezifitĂ€t von 88,2%. Das Ă€quivalente CNR der DTI Sequenz ist Hinweis auf eine zur KM-T1w gleichwertige Sichtbarkeit von Herdbefunden. Nicht nur ADC Werte, sondern auch die FA differierte zwischen den verschiedenen untersuchten Gewebetypen
An optimizational approach for an algorithmic reassembly of fragmented objects
In Cambodia close to the Thai border, lies the Angkor-style temple of Banteay Chhmar. Like all nearly forgotten temples in remote places, it crumbles under the ages. By today most of it is only a heap of stones. Manually reconstructing these temples is both complex and challenging: The conservation team is confronted with a pile of stones, the
original position of which is generally unkown. This reassembly task resembles a large-scale 3D puzzle. Usually, it is resolved by a team of specialists who analyze each stone, using their experience and knowledge of Khmer culture. Possible solutions are tried and retried and the stones are placed in different locations until the correct one is found. The major drawbacks of this technique are: First, since the stones are moved continuously they are further damaged, second, there is a threat to the safety of the workers due to handling very heavy weights, and third because of the high complexity and labour-intensity of the
work it takes several months up to several years to solve even a small part of the puzzle.
These risks and conditions motivated the development of a virtual approach to reassemble the stones, as computer algorithms are theoretically capable of enumerating all
potential solutions in less time, thereby drastically reducing the amount of work required for handling the stones. Furthermore the virtual approach has the potential to reduce the on-site costs of in-situ analysis. The basis for this virtual puzzle algorithm are high-resolution 3D models of more than one hundred stones. The stones can be viewed as polytopes with approximately cuboidal form although some of them contain additional indentations. Exploiting these and related geometric features and using a priori knowledge of the orientation of each stone speeds up the process of matching the stones.
The aim of the current thesis is to solve this complex large-scale virtual 3D puzzle. In order to achieve this, a general workflow is developed which involves 1) to simplify
the high-resolution models to their most characteristic features, 2) apply an advanced similarity analysis and 3) to match best combinations as well as 4) validate the results.
The simplification step is necessary to be able to quickly match potential side-surfaces. It introduces the new concept of a minimal volume box (MVB) designed to closely and storage efficiently resemble Khmer stones.Additionally, this reduced edge-based model is used to segment the high-resolution data according to each side-surface. The second step presents a novel technique allowing to conduct a similarity analysis of virtual temple stones. It is based on several geometric distance functions which determine the relatedness of a potential match and is capable of sorting out unlikely ones. The third step employs graph theoretical methods to combine the similarity values into a correct solution of this large-scale 3D puzzle. The validation demonstrates the high quality and robustness of this newly constructed puzzle workflow.
The workflow this thesis presents virtually puzzles digitized stones of fallen straight Khmer temple walls. It is able to virtually and correctly reasemble up to 42 digitized stones requiring a minimum of user-interaction

Ziel des Projektes war es mit Bezug auf die allgemeinen Klimaschutzziele des Landes Hessen Grundlagen fĂŒr den bisher wenig beachteten Markt der AuĂer-Haus-Verpflegung (AHV) zu erheben. DafĂŒr sollte eine Treibhausgas (THG) -Bilanzierung in ausgewĂ€hlten GroĂkĂŒchen durchgefĂŒhrt werden. Basierend auf den Ergebnissen wurden Optimierungsfelder erarbeitet und aufgezeigt. Zur Gewinnung der entsprechenden Grundlagen wurden sowohl standortbezogene Bilanzierungen als auch die Berechnungen von EinzelmenĂŒs angestrebt. Die Ergebnisse der Bilanzierung sowie die aufgezeigten Optimierungsfelder bzw. THG-Einsparpotenziale, könnten als Basis dienen fĂŒr die Ausarbeitung von vertiefenden Klimaschutzzielen im Bereich der AHV im hessischen Klimaschutzplan 2025
Pharmacodynamic Effects of an Angiotensin II Receptor-Antagonist in Phase IâComparison between Healthy Subjects and Patients with Hypertension
Biomarkers are increasingly used to provide decision making data early in phase I by showing Proof of Mechanism or Proof of Concept (PoM/PoC). For antihypertensive agents, the administration of multiple doses (md) to hypertensive patients is assumed to be necessary for an early go/no-go decision. We compared the effects of an Angiotensin II receptor antagonist (ARA) on Plasma Renin and blood pressure (BP) following an oral single dose (sd) and once daily md for seven days to healthy volunteers and patients with essential hypertension (diastolic BP 95 mmHg to 114 mmHg; systolic BP 130 mmHg to 200 mmHg). Methods: 5â12 healthy male subjects/dose received 10 mg to 300 mg ARA sd and 50 to 300 mg md for 7 days; patients (9â10/dose) received 20 mgâ400 mg ARA for 7 days. The studies were designed as randomized, single-blind, placebo-controlled, group comparison or crossover dose-escalation studies. Plasma Renin and BP were monitored up to 24 hours after dosing. Results: Plasma Renin showed a high interindividual variability in both healthy volunteers and patients. Healthy subjects showed a dose- and time-related increase in plasma Renin after sd from 40 mg to 300 mg and md of 50 mg to 300 mg (p < 0.05 for doses of 200 mg and 300 mg). In patients, increases in plasma Renin occurred at 8 hours and beyond starting at sd of 100 mg and md of 50 mg (p < 0.05 for the dose of 400 mg). While healthy volunteers showed no relevant decrease in BP, in hypertensive patients a reduction in BP in doses of 100 mg to 400 mg occurred (p < 0.05); effects were more pronounced after md compared to sd. Conclusion: Early PoM for an antihypertensive agent can be shown by use of laboratory biomarkers following sd to healthy subjects. PoC can be achieved after sd in hypertensive patients. Administration of sd to healthy volunteers is sufficient for an early go/no-decision
The Cards and Lottery Task: Validation of a New Paradigm Assessing Decision Making Under Risk in Individuals With Severe Obesity
Background: A growing body of research demonstrated impaired executive functions in
individuals with severe obesity, including increased sensitivity to reward and impulsive
decision making under risk conditions. For the assessment of decision making in patients
with severe obesity, studies widely used the Iowa Gambling Task (IGT) or the Delay
Discounting Task (DDT), which cover short-term or long-term consequences of decisions
only. A further development originating from the field of addiction research is the Cards
and Lottery Task (CLT), in which each decision made has conflicting immediate and longterm
consequences at the same time. The present study aimed to validate the CLT in
individuals with severe obesity.
Methods: Patients with severe obesity (N = 78, 67% women, 42.9 ± 10.4 years old, body
mass index of 48.1 ± 8.3 kg/m2) were included. Convergent validity was evaluated using
the computerized Delay Discounting Task and well-established self-report questionnaires
assessing different aspects of impulsivity. For discriminant validity, CLT performance was
compared between symptom groups characterized by high versus low impulsivity. The
taskâs clinical validity was evaluated based on associations with general and eating
disorder psychopathology, and body mass index. Test-retest reliability was determined by
administering the CLT in n = 31 participants without weight-loss treatment one year later.
The taskâs sensitivity to change due to weight loss was evaluated by retesting n = 32
patients one year after receiving obesity surgery.
Results: The number of advantageous decisions in the CLT was significantly positively
associated with delay discounting and effortful control, and significantly negatively
correlated with behavioral impulsivity. CLT performance differed significantly between
individuals with and without symptoms of attention-deficit/hyperactivity disorder and
between samples with severe obesity and healthy controls. Clinically, CLT performance
was significantly associated with general, but not eating disorder psychopathology. The
CLT showed moderate test-retest reliability after one year in weight-stable individuals and
was sensitive to change in those undergoing obesity surgery.
Conclusions: This study identified the CLT to be a highly promising, new complex
measure of short- and long-term decision making with good reliability and validity in
individuals with severe obesity. Future studies should assess its association with the IGT
and predictive value for real-life health behavior
Changes in visual attention towards food cues after obesity surgery: An eye-tracking study
Research documented the effectiveness of obesity surgery (OS) for long-term weight loss and improvements in medical and psychosocial sequelae, and general cognitive functioning. However, there is only preliminary evidence for changes in attentional processing of food cues after OS. This study longitudinally investigated visual attention towards food cues from pre- to 1-year post-surgery. Using eye tracking (ET) and a Visual Search Task (VST), attentional processing of food versus non-food cues was assessed in n = 32 patients with OS and n = 31 matched controls without weight-loss treatment at baseline and 1-year follow-up. Associations with experimentally assessed impulsivity and eating disorder psychopathology and the predictive value of changes in visual attention towards food cues for weight loss and eating behaviors were determined. During ET, both groups showed significant gaze duration biases to non-food cues without differences and changes over time. No attentional biases over group and time were found by the VST. Correlations between attentional data and clinical variables were sparse and not robust over time. Changes in visual attention did not predict weight loss and eating disorder psychopathology after OS. The present study provides support for a top-down regulation of visual attention to non-food cues in individuals with severe obesity. No changes in attentional processing of food cues were detected 1-year post-surgery. Further studies are needed with comparable methodology and longer follow-ups to clarify the role of biased visual attention towards food cues for long-term weight outcomes and eating behaviors after OS
Conditional Deletion of LRP1 Leads to Progressive Loss of Recombined NG2-Expressing Oligodendrocyte Precursor Cells in a Novel Mouse Model
The low-density lipoprotein receptor-related protein 1 (LRP1) is a transmembrane
receptor, mediating endocytosis and activating intracellular signaling cascades. LRP1 is highly
expressed in the central nervous system (CNS), especially in oligodendrocyte precursor cells
(OPCs). Previous studies have suggested LRP1 as a regulator in early oligodendrocyte development,
repair of chemically induced white matter lesions, and cholesterol homeostasis. To circumvent
embryonic lethality observed in the case of global LRP1 deletion, we generated a new inducible
conditional knockout (KO) mouse model, which enabled an NG2-restricted LRP1 deficiency
(NG2-CreERT2ct2/wtxR26eGFPflox/floxxLRP1flox/flox). When characterizing our triple transgenic mouse
model, we noticed a substantial and progressive loss of recombined LRP1-deficient cells in the
oligodendrocyte lineage. On the other hand, we found comparable distributions and fractions of
oligodendroglia within the Corpus callosum of the KO and control animals, indicating a compensation
of these deficits. An initial study on experimental autoimmune encephalomyelitis (EAE) was
performed in triple transgenic and control mice and the cell biology of oligodendrocytes obtained
from the animals was studied in an in vitro myelination assay. Differences could be observed in these
assays, which, however, did not achieve statistical significance, presumably because the majority
of recombined LRP1-deficient cells has been replaced by non-recombined cells. Thus, the analysis
of the role of LRP1 in EAE will require the induction of acute recombination in the context of the
disease process. As LRP1 is necessary for the survival of OPCs in vivo, we assume that it will play an
important role in myelin repair
