    SAM domain-containing N-terminal region of SAMHD1 plays a crucial role in its stabilization and restriction of HIV-1 infection

    SAMHD1 restricts human immunodeficiency virus type 1 (HIV-1) infection in a cell-type specific manner. Other than primary monocyte derived cells and resting CD4+ T cells, the SAMHD1-mediated HIV-1 block was reported only in phorbol 12-myristate 13-acetate (PMA)-differentiated THP-1 and U937 monocyte cell lines. We previously reported that SAMHD1 restricted HIV-1 infection in TE671 rhabdomyosarcoma cells in addition to these cell lines. In this study, we compared the amounts of the full-length SAMHD1 and its deletion mutants, SAM domain containing N-terminal fragment (residues 1-119, SAMHD1n) and HD domain containing C-terminal fragment (120-626, SAMHD1c) in U937, TE671, and HeLa cells. The results showed that the full-length SAMHD1 and SAMHD1n proteins were significantly more abundant than the SAMHD1c protein in TE671 and differentiated U937 cells. The proteasome inhibitor MG132 increased the amount of the SAMHD1c and the SAMHD1c-fused GFP proteins. In contrast, the fusion of the SAMHD1n to the APOBEC3G protein inhibited Vif-induced proteasomal degradation in TE671 and in differentiated U937 cells. These results indicated that the SAMHD1 C-terminal HD domain-containing region leads the SAMHD1 to proteasomal degradation, and the SAMHD1 N-terminal SAM domain-containing region stabilizes the protein. Our study showed that the SAMHD1 protein expression is post-translationally regulated and the significance of SAM and HD domains for the full-length SAMHD1 protein stability. Further, we suggest that the SAM domain-containing N-terminal region participate in the cell-type specific restrictive function of SAMHD1 against HIV-1 infection, by protein stabilization

    Detection of deviance in Japanese kanji compound words

    Reading fluency is based on the automatic visual recognition of words. As a manifestation of the automatic processing of words, an automatic deviance detection of visual word stimuli can be observed in the early stages of visual recognition. To clarify whether this phenomenon occurs with Japanese kanji compounds-since their lexicality is related to semantic association-we investigated the brain response by utilizing three types of deviants: differences in font type, lexically correct or incorrect Japanese kanji compound words and pseudo-kanji characters modified from correct and incorrect compounds. We employed magnetoencephalography (MEG) to evaluate the spatiotemporal profiles of the related brain regions. The study included 22 adult native Japanese speakers (16 females). The abovementioned three kinds of stimuli containing 20% deviants were presented during the MEG measurement. Activity in the occipital pole region of the brain was observed upon the detection of font-type deviance within 250 ms of stimulus onset. Although no significant activity upon detecting lexically correct/incorrect kanji compounds or pseudo-kanji character deviations was observed, the activity in the posterior transverse region of the collateral sulcus (pCoS)-which is a fusiform neighboring area-was larger when detecting lexically correct kanji compounds than when detecting pseudo-kanji characters. Taken together, these results support the notion that the automatic detection of deviance in kanji compounds may be limited to a low-level feature, such as the stimulus stroke thickness.Peer reviewe

    High Level Estradiol Induces EBV Reactivation and EBV gp350/220(+)CD138(+) Double-positive B Cell Population in Graves’ Disease Patients and Healthy Controls

    Graves’ disease occurs predominantly in women. Epstein-Barr virus (EBV) mainly persists in human B lymphocytes, and its reactivation stimulates antibody production. We previously suggested that the EBV reactivation-induced production of TRAb and IgM at 100 nM estradiol (pregnant level) was lower than that at 0 nM estradiol and that class switch recombination may be increased by estradiol. In this study, we examined the effect of estradiol on EBV reactivation. We identified the expression of EBV-glycoprotein 350/220 (gp350/220) in the late phase of reactivation and plasma cell differentiation of EBV-infected cells using 72A1 antibody and CD138 antibody, respectively. We found the mean ratio of gp 350/220(+) CD138(+) cells at 100 nM estradiol was higher than that at 0 nM estradiol. These results suggested that EBV-infected cells could survive with keeping the ability of antibody production in 100 nM estradiol, which is consistent with the improvement of Graves’ disease during maternity and exacerbation postpartum

    Unraveling the antifungal activity of a South American rattlesnake toxin crotamine

    Crotamine is a highly basic peptide from the venom of Crotalus durissus terrificus rattlesnake. Its common gene ancestry and structural similarity with the beta-defensins, mainly due to an identical disulfide bond pattern, stimulated us to assess the antimicrobial properties of native, recombinant, and chemically synthesized crotamine. Antimicrobial activities against standard strains and clinical isolates were analyzed by the colorimetric microdilution method showing a weak antibacterial activity against both Gram-positive and Gram-negative bacteria [MIC (Minimum Inhibitory Concentration) of 50->200 mu g/mL], with the exception of Micrococcus luteus [MIC ranging from 1 to 2 mu g/mL]. No detectable activity was observed for the filamentous fungus Aspergillus fumigatus and Trichophyton rubrum at concentrations up to 125 mu g/mL. However, a pronounced antifungal activity against Candida spp., Trichosporon spp., and Cryptococcus neoformans [12.5-50.0 mu g/mL] was observed. Chemically produced synthetic crotamine in general displayed MIC values similar to those observed for native crotamine, whereas recombinant crotamine was overridingly more potent in most assays. On the other hand, derived short linear peptides were not very effective apart from a few exceptions. Pronounced ultrastructure alteration in Candida albicans elicited by crotamine was observed by electron microscopy analyses. the peculiar specificity for highly proliferating cells was confirmed here showing potential low cytotoxic effect of crotamine against nontumoral mammal cell lines (HEK293, PC12, and primary culture astrocyte cells) compared to tumoral B16F10 cells, and no hemolytic activity was observed. Taken together these results suggest that, at low concentration, crotamine is a potentially valuable anti-yeast or candicidal agent, with low harmful effects on normal mammal cells, justifying further studies on its mechanisms of action aiming medical and industrial applications.     Calcium sparks enhance the tissue fluidity within epithelial layers and promote apical extrusion of transformed cells

    In vertebrates, newly emerging transformed cells are often apically extruded from epithelial layers through cell competition with surrounding normal epithelial cells. However, the underlying molecular mechanism remains elusive. Here, using phospho-SILAC screening, we show that phosphorylation of AHNAK2 is elevated in normal cells neighboring RasV12 cells soon after the induction of RasV12 expression, which is mediated by calcium-dependent protein kinase C. In addition, transient upsurges of intracellular calcium, which we call calcium sparks, frequently occur in normal cells neighboring RasV12 cells, which are mediated by mechanosensitive calcium channel TRPC1 upon membrane stretching. Calcium sparks then enhance cell movements of both normal and RasV12 cells through phosphorylation of AHNAK2 and promote apical extrusion. Moreover, comparable calcium sparks positively regulate apical extrusion of RasV12-transformed cells in zebrafish larvae as well. Hence, calcium sparks play a crucial role in the elimination of transformed cells at the early phase of cell competition

    ZAK Inhibitor PLX4720 Promotes Extrusion of Transformed Cells via Cell Competition

    Previous studies have revealed that, at the initial step of carcinogenesis, transformed cells are often eliminated from epithelia via cell competition with the surrounding normal cells. In this study, we performed cell competition-based high-throughput screening for chemical compounds using cultured epithelial cells and confocal microscopy. PLX4720 was identified as a hit compound that promoted apical extrusion of RasV12-transformed cells surrounded by normal epithelial cells. Knockdown/knockout of ZAK, a target of PLX4720, substantially enhanced the apical elimination of RasV12 cells in vitro and in vivo. ZAK negatively modulated the accumulation or activation of multiple cell competition regulators. Moreover, PLX4720 treatment promoted apical elimination of RasV12-transformed cells in vivo and suppressed the formation of potentially precancerous tumors. This is the first report demonstrating that a cell competition-promoting chemical drug facilitates apical elimination of transformed cells in vivo, providing a new dimension in cancer preventive medicine

    The thymus has an important role in the immune reaction. The lymphocytes localized both in thymic cortex and in medulla, known as T cells, proliferate in an environment of the epithelial reticular cells. T cells educated to react against non-self materials enter the circulation and function in peripheral immunity. Thymus of the mouse has almost developed and grown to its natal size by the 18 days gestation. After the birth thymus grows at a somewhat slower rate until puberty. It subsequently shows a decrease both in size and in weight. That is due to the decrease in the lymphocyte-accumulated region and to the partial replacement by fat tissue. The process is so-called age"involution". Fat cells are classified as connective-tissue cells. They must develop and proliferate in the capsule or in the trabecula of the thymus after birth, because no typical fat cells containing lipid droplets are observed at birth.We examined when fat cells appeared in the thymus after the birth. Fat cells are known to express leptin, so that the appearance of leptin mRNA was detected by the PCR method. The expression of leptin was detected as early as in the thymus of the 3 week-old mouse and increased with the age. It is sugested that fat tissue development begins immediately after the birth and precedes to the age involution in the thymus, although that the cortex and medulla develops until puberty

    Enhanced Nogo-P3 amplitudes of mothers compared with non-mother women during an emotional Go/Nogo task

    Background: It is known that emotion regulatory responses of humans are changed by the experiences they have, but in particular, they are changed by becoming a mother. A recent study has found how a woman's emotion regulatory response to a child's crying changes after becoming a mother. However, mothers' emotion regulatory responses other than those to children and the association between emotion regulatory response and parental stress are still unknown. Methods: Eighteen healthy Japanese females (nine mothers and nine non-mothers) participated in the experiment. They performed an emotional Go/Nogo task, with facial expressions of others (angry, happy, and neutral faces) used as emotional stimuli. The percentage of correct responses, response time, and event-related potentials (ERPs) during the task was measured. Results: This comparison revealed that the mother group had a larger P3 (Nogo-P3) amplitude than the non-mother group when Nogo trials were held. This indicates that in mothers, there was greater activation of the behavioral inhibition-related brain areas than in non-mother women when they inhibited inappropriate behavior following recognition of facial expressions of others. In addition, in the mother group, there was a negative correlation between parental stress levels and Nogo-P3 amplitudes evoked by angry faces. This suggests that there is a relation between the level of parental stress of mothers and their emotion regulatory responses to angry faces. Conclusions: Our results demonstrate that mothers' emotion regulatory processes may differ from those of non-mothers in response, not only to a child's crying but also to expressions of emotions by others, and also suggest that the inhibitory recognition activity of mothers can be affected by parental stres
