    The development of recombinant Adenoviral vaccines to target pneumovirus infection

    Respiratory Syncytial Virus (RSV) is a member of the pneumovirus genus (family Paramyxoviridae, subfamily Pneumovirinae). RSV is an important respiratory virus of both infants and the elderly, representing an underappreciated burden on health care systems. In addition, re-infections can occur despite the presence of pre-existing immunity, suggesting that immunological memory to RSV is incomplete. To date, treatment of RSV infection is limited to the provision of supportive care and no effective vaccine is available. Although several are currently under investigation, these candidates focus upon the delivery of the F and G antigens of RSV to stimulate the immune system, rather than the internal antigens, which may provide cross protection between different subtypes of RSV. Vaccine development has been greatly hindered by the lack of an appropriate animal model in which to study vaccine efficacy and pneumovirus pathogenesis. Pneumonia virus of mice (PVM) is also a member of the Pneumovirus genus and, like RSV infection of humans, causes a bronchiolitis and fatal pneumonia in its natural host, the mouse. PVM has been proposed as an appropriate model system in which to both study pneumovirus pathogenesis and vaccine efficacy. The PVM model system was adapted to investigate a potential vaccination strategy to address the lack of an available RSV vaccine. Replication deficient recombinant adenovirus serotype 5 (rAd5) vectors were constructed which expressed the F, M and N genes of PVM J3666, in addition to a control construct, which expressed the LacZ gene of E. coli. The constructs were administered via the intranasal route to BALB/c mice and were able to elicit complete protection against a lethal dose of pathogenic PVM J3666, in both short-term experiments and in a long-term experiment, up to 20 weeks post immunisation. The protection effect elicited by the constructs was observed when administered in a single dose, and in alternative mouse strains, C3H/He-mg and C57BL/6, which had differing immunity haplotypes. The rAd5 vectors generated a PVM specific IgG humoral response to PVM and Ad5 antigen which did not correlate as the primary mediator of protection. The rAd5 candidate expressing the N gene of PVM was shown to induce IFNγ secreting T-cells. The use of a peptide library of PVM N protein determined that a specific response could be identified towards the amino acids N41-90, N81-130, N161-210 and N281-330.     A single consumption of curry improved postprandial endothelial function in healthy male subjects: a randomized, controlled crossover trial

    BACKGROUND: Curry, one of the most popular foods in Japan, contains spices that are rich in potentially antioxidative compounds, such as curcumin and eugenol. Oxidative stress is thought to impair endothelial function associated with atherosclerosis, a leading cause of cardiovascular events. The aim of this study was to determine whether a single consumption of curry meal would improve endothelial function in healthy men. METHODS: Fourteen healthy male subjects (BMI 23.7 ± 2.7 kg/m(2); age 45 ± 9 years) were given a single serving of curry meal or spice-free control meal (180 g of curry or control and 200 g of cooked rice; approximately 500 kcal in total) in a randomized, controlled crossover design. Before and 1 hr after the consumption, fasting and postprandial flow-mediated vasodilation (FMD) responses and other parameters were measured. RESULTS: The consumption of the control meal decreased FMD from 5.8 ± 2.4% to 5.1 ± 2.3% (P = 0.039). On the other hand, the consumption of the curry meal increased FMD from 5.2 ± 2.5% to 6.6 ± 2.0% (P = 0.001), and the postprandial FMD after the curry meal was higher than that after the control meal (P = 0.002). Presence of spices in the curry did not alter significantly the systemic and forearm hemodynamics, or any biochemical parameters including oxidative stress markers measured. CONCLUSIONS: These findings suggest that the consumption of curry ameliorates postprandial endothelial function in healthy male subjects and may be beneficial for improving cardiovascular health.     Evaluation of a new histologic staging and grading system for primary biliary cirrhosis in comparison with classical systems

    Recently, our research team proposed a new histologic staging and grading system for primary biliary cirrhosis (PBC) that takes into account necroinflammatory activity and histologic heterogeneity. The present study aimed to confirm the usefulness of the new evaluation system. A total of 152 liver biopsy specimens and clinical data (including outcomes in patients with PBC before treatment with ursodeoxycholic acid) were analyzed with respect to the new system. Staging was evaluated on the basis of 3 histologic components (fibrosis, bile duct loss, and deposition of orcein-positive granules), and grading was assessed on the basis of chronic cholangitis activity and hepatitis activity. Concurrently, the classical systems, that is, the Scheuer and Ludwig staging systems, were also assessed and compared with our new system. PBC cases showed different distributions in each stage of the 3 systems. The new staging and grading system reflected liver dysfunctions before specific treatment. This was on a par with the results obtained using the classical systems. Development of cirrhosis-related conditions correlated well with the new staging system compared with the 2 classical staging systems, and in particular, the amount of deposition of orcein-positive granules could reflect development of cirrhosis-related conditions (scores 0-1 versus scores 2-3 groups, P < .0001). In conclusion, the new PBC staging system was demonstrated to reflect clinicolaboratory features, and its progression was associated with the development of cirrhosis-related conditions. © 2013 Elsevier Inc.     Muscular contractions in the zebrafish embryo are necessary to reveal thiuram-induced notochord distortions

It has been reported that dithiocarbamates, including thiuram (tetramethylthiuram disulfide), cause wavy distortions of the notochord in zebrafish and other fish embryos. In the present study, we investigated the mechanism underlying the toxicity of thiuram in zebrafish embryos. When embryos were exposed to thiuram (2-1,000 nM: 0.48-240 µg/L) from 3 hours post fertilization (hpf) (30% epiboly) until 24 hpf (Prim-5), all embryos develop wavy notochords, disorganized somites, and have shortened yolk sac extensions. The thiuram response was specific and did not cause growth retardation or mortality at 24 hpf. The thiuram-dependent responses showed the same concentration dependence with a waterborne EC50 values of approximately 7 nM. Morphometric measurements revealed that thiuram does not affect the rate of notochord lengthening. However, the rate of overall body lengthening was significantly reduced in thiuram exposed animals. Other dithiocarbamates, such as ziram, caused similar malformations to thiuram. While expression of genes involved in somitogenesis was not affected, the levels of notochord specific transcripts were altered before and after the onset of malformations. Distortion of the notochord started precisely at 18 hpf, which is concomitant with onset of spontaneous rhythmic trunk contractions. Abolishment of spontaneous contractions using tricaine, α-bungarotoxin, and a paralytic mutant sofa potato, resulted in normal notochord morphology in the presence of thiuram.     A dehydrated space-weathered skin cloaking the hydrated interior of Ryugu

    Without a protective atmosphere, space-exposed surfaces of airless Solar System bodies gradually experience an alteration in composition, structure and optical properties through a collective process called space weathering. The return of samples from near-Earth asteroid (162173) Ryugu by Hayabusa2 provides the first opportunity for laboratory study of space-weathering signatures on the most abundant type of inner solar system body: a C-type asteroid, composed of materials largely unchanged since the formation of the Solar System. Weathered Ryugu grains show areas of surface amorphization and partial melting of phyllosilicates, in which reduction from Fe3+ to Fe2+ and dehydration developed. Space weathering probably contributed to dehydration by dehydroxylation of Ryugu surface phyllosilicates that had already lost interlayer water molecules and to weakening of the 2.7 µm hydroxyl (–OH) band in reflectance spectra. For C-type asteroids in general, this indicates that a weak 2.7 µm band can signify space-weathering-induced surface dehydration, rather than bulk volatile loss

    Remote assistance-focused strength training and nutritional guidance in preventing frailty on body composition and muscle strength in community-dwelling Japanese older adults during COVID-19 pandemic

    This study aimed to investigate whether the implementation of a frailty prevention program using remote assistance combined with face-to-face guidance or solely through a remote assistance method is effective for community-dwelling Japanese older adults. Forty-nine community-dwelling older adults (16 men, age: 70.0 ± 4.8 years) were classified into the following three groups: remote plus face-to-face guidance group (RFGG; n = 17), remote guidance group (RGG; n = 17), and control group (CG; n = 15). The RFGG performed face-to-face supervised strength training once a week for the first 4 weeks, plus similar exercises once a week at home, followed by 8 weeks of performing the same exercises twice a week at home while watching an instructional digital versatile disc (DVD) for a total of 12 weeks. They received one face-to-face nutritional guidance session, followed by guidance over the telephone once a month for 3 months. The RGG performed the same exercises at home while watching the DVD twice weekly and received nutritional guidance over the telephone once a month for 3 months (12 weeks). Only pre- and post-intervention surveys in the CG were conducted. When compared to pre-intervention measurements, RFGG lean body mass, lower limb muscle mass, and grip strength increased significantly, while fat mass percentage decreased significantly (p &lt; 0.05). In the RGG, no significant changes were observed in the physical characteristics. These results suggest that a frailty prevention program not only using a remote guidance but also combined with a face-to-face guidance may be effective for older adults
