    Macular Microcysts in Mitochondrial Optic Neuropathies: Prevalence and Retinal Layer Thickness Measurements.

    PurposeTo investigate the thickness of the retinal layers and to assess the prevalence of macular microcysts (MM) in the inner nuclear layer (INL) of patients with mitochondrial optic neuropathies (MON).MethodsAll patients with molecularly confirmed MON, i.e. Leber's Hereditary Optic Neuropathy (LHON) and Dominant Optic Atrophy (DOA), referred between 2010 and 2012 were enrolled. Eight patients with MM were compared with two control groups: MON patients without MM matched by age, peripapillary retinal nerve fiber layer (RNFL) thickness, and visual acuity, as well as age-matched controls. Retinal segmentation was performed using specific Optical coherence tomography (OCT) software (Carl Zeiss Meditec). Macular segmentation thickness values of the three groups were compared by one-way analysis of variance with Bonferroni post hoc corrections.ResultsMM were identified in 5/90 (5.6%) patients with LHON and 3/58 (5.2%) with DOA. The INL was thicker in patients with MON compared to controls regardless of the presence of MM [133.1±7μm vs 122.3±9μm in MM patients (p<0.01) and 128.5±8μm vs. 122.3±9μm in no-MM patients (p<0.05)], however the outer nuclear layer (ONL) was thicker in patients with MM (101.4±1mμ) compared to patients without MM [77.5±8mμ (p<0.001)] and controls [78.4±7mμ (p<0.001)]. ONL thickness did not significantly differ between patients without MM and controls.ConclusionThe prevalence of MM in MON is low (5-6%), but associated with ONL thickening. We speculate that in MON patients with MM, vitreo-retinal traction contributes to the thickening of ONL as well as to the production of cystic spaces

    ABSTRACTThe Mar Piccolo Basin is a coastal brackish marine ecosystem located along the northern coast of the Gulf of Taranto (Southern Italy). Despite the ecological relevance of the area (Site of Community Importance IT9130004, Regional Reserve 'Palude La Vela' EUAP1189), the entire basin is subjected to intensive human usage. The main activities include extensive mussel farming, important industrial activities, a military harbor and densely populated shores. The goal of our study was to spatially quantify human pressure within the basin and its relationship with biocoenoses. A broad set of data was integrated including acoustic remote data (obtained using a multibeam echosounder and side scan sonar devices), direct observations obtained by SCUBA diving and from a trawled camera, an orthophoto and ESRI® Imagery Basemap.     Early Renal Involvement in Cats with Natural Feline Morbillivirus Infection

    Feline morbillivirus (FeMV) is a newly discovered paramyxovirus infecting domestic cats and its role in the pathogenesis of feline chronic kidney disease (CKD) has been suggested, however not confirmed. The primary aim of the study was to evaluate the renal damage associated with FeMV infection in cats. In this retrospective study, clinical and clinicopathological data were compared among 14 FeMV naturally infected, 21 CKD and 22 healthy cats. FeMV positive cats had serum chemistry analytes and main urine chemistry results similar to the healthy subjects. FeMV positive cats had significantly decreased urine specific gravity (median 1054, range 1022-1065) and urine creatinine (median 227.23 mg/dL, range 83.02-489.75) when compared with healthy cats (median 1067, range 1040-1080, P < 0.001; median 406.50 mg/dL, range 195.32-575.58; P < 0.001, respectively). Urine protein:creatinine ratio (UPC) results of FeMV and CKD were not different (median 0.20, range 0.08-1.03; median 0.23, range 0.10-0.80, respectively), however UPC results were significantly increased in both groups, if compared with healthy cats (median 0.1, range 0.04-0.250, P < 0.01). Based on clinical data, serum creatinine concentration, urine specific gravity and UPC results, CKD was suspected by clinicians in 3/14 FeMV cats. Urine protein sodium-dodecyl-sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) in 10/13 (77%) FeMV cats indicated a tubular pattern, with a decrease of uromodulin and an increase in the number and intensity of low molecular weight proteins. FeMV infection can be associated with different grades of renal dysfunction ranging from mild tubular proteinuria with less concentrated urine to azotemia in cats younger than those typically affected by CKD

    A real-time RT-PCR assay for molecular identification and quantitation of feline morbillivirus RNA from biological specimens.

    The aim of this study was to develop a real-time RT-PCR to detect and quantitate feline morbillivirus (FeMV) RNA in biological samples. Primers and probe were targeted on a conserved region of FeMV P/V/C gene. To validate the assay with field samples, a total number of specimens of cats have been recruited including 264 urine and blood samples and compared with a generic RT-PCR targeting the L protein encoding gene of morbilliviruses. In addition, 385 tissue samples from 35 carcasses of cats have been also employed. RNA titres were low in all tested samples. Results also indicated the absence of cross-reaction with related morbilliviruses and existing pathogens of cats. However, in tissues with low levels of FeMV RNA, the presence of viral antigen was confirmed by immunohistochemistry targeting the N viral protein. This newly described assay allows for a rapid, accurate and reliable quantitative detection of FeMV RNA that can be applied for diagnostics and research studies

    Extraction of DNA from Choline Chloride Feed Additive (CC) and from derived Pre-Mixes (PMCC) and Screening of CC and PMCC for (a) presence of rice and (b) presence of Bt63

    Choline Chloride 60% (CC) is a feed additive that is imported in significant quantities from China. In a number of cases GM rice, harbouring the event Bt63, has been found in imported CC batches but not in derived pre-mixes. The Member States were asked to provide positive CC samples and derived pre-mixes to the EU-RL GMFF in order to allow the EU-RL GMFF establishing practical approaches to DNA extraction and the subsequent testing for presence of (a) rice and (b) the Bt63 rice event. This technical note has been derived based on data provided from National Control Laboratories and experience made by the EU-RL GMFF when re-testing nine feed additive samples and ten pre-mixes samples. The EU-RL GMFF found that the extraction of DNA from the feed additive Choline Chloride (CC) does not normally pose specific problems. It can be carried out following the available protocols or using available standard DNA extraction kits. The extraction of DNA from derived pre-mixes (PMCC) is, however, posing problems because of observed strong inhibition and the presence of DNA from additional sources than those present in the CC. The EU-RL GMFF has tried a number of protocols and extraction kits on PMCC samples. On this basis it is concluded that it is advisable to carefully purify the DNA, to verify the possible presence of inhibition effects and eventually to try to reduce any inhibition observed. Concerning testing the extracted DNA for rice and Bt63 rice event presence, this does not pose a problem in case of the CC while for PMCC rice was not anymore detectable or was detected in trace amounts. In any case testing for Bt63 presence was impossible. A possible explanation could be that much of the extracted DNA is from other plants than rice and hence the concentration of rice DNA is below the LOD of the rice-taxon specific method. In order to verify this, the EU-RL GMFF has analysed the available rice (taxon-) specific methods by means of bio-informatics and (partly) in the laboratory and concludes that they all should be suitable for specifically and reliably detecting rice from the Oryza sativa species.     Grand Rounds: Could Occupational Exposure to n-Hexane and Other Solvents Precipitate Visual Failure in Leber Hereditary Optic Neuropathy?

    CONTEXT: Leber hereditary optic neuropathy (LHON) is a maternally inherited loss of central vision related to pathogenic mutations in the mitochondrial genome, which are a necessary but not sufficient condition to develop the disease. Investigation of precipitating environmental/occupational (and additional genetic) factors could be relevant for prevention. CASE PRESENTATION: After a 6-month period of occupational exposure to n-hexane and other organic solvents, a 27-year-old man (a moderate smoker) developed an optic neuropathy. The patient had a full ophthalmologic and neurologic investigation, including standardized cycloergometer test for serum lactic acid levels and a skeletal muscle biopsy. His exposure history was also detailed, and he underwent genetic testing for LHON mitochondrial DNA mutations. The patient suffered a sequential optic neuropathy with the hallmarks of LHON and tested positive for the homoplasmic 11778G → A/ND4 mutation. Routine laboratory monitoring revealed increased concentrations of urinary 2.5 hexandione (n-hexane metabolite) and hippuric acid (toluene metabolite) in the period immediately preceding the visual loss. DISCUSSION: In a subject carrying an LHON mutation, the strict temporal sequence of prolonged appreciable occupational exposure followed by sudden onset of visual loss must raise a suspicion of causality (with a possible further interaction with tobacco smoke). RELEVANCE: In this article, we add to the candidate occupational/environmental triggers of LHON and highlight the need for appropriate case–control (and laboratory) studies to validate the causal effect of mixed toxic exposures

    The balloon-borne large-aperture submillimeter telescope for polarimetry: BLAST-Pol

    The Balloon-borne Large Aperture Submillimeter Telescope for Polarimetry (BLAST-Pol) is a suborbital mapping experiment designed to study the role played by magnetic fields in the star formation process. BLAST-Pol is the reconstructed BLAST telescope, with the addition of linear polarization capability. Using a 1.8 m Cassegrain telescope, BLAST-Pol images the sky onto a focal plane that consists of 280 bolometric detectors in three arrays, observing simultaneously at 250, 350, and 500 um. The diffraction-limited optical system provides a resolution of 30'' at 250 um. The polarimeter consists of photolithographic polarizing grids mounted in front of each bolometer/detector array. A rotating 4 K achromatic half-wave plate provides additional polarization modulation. With its unprecedented mapping speed and resolution, BLAST-Pol will produce three-color polarization maps for a large number of molecular clouds. The instrument provides a much needed bridge in spatial coverage between larger-scale, coarse resolution surveys and narrow field of view, and high resolution observations of substructure within molecular cloud cores.
