IBD Serology and Disease Outcomes in African Americans with Crohn's Disease
Backgrounds: Recent studies have identified the role of serologic markers in characterizing disease phenotype, location, complications, and severity among Northern Europeans (NE) with Crohn's disease (CD). However, very little is known about the role of serology in CD among African Americans (AA). Our study explored the relationship between serology and disease phenotype in AA with CD, while controlling for genetic ancestry. Methods: AAs with CD were enrolled as participants through multicenter collaborative efforts. Serological levels of IgA anti-Saccharomyces cervisiae antibody (ASCA), IgG ASCA, E. coli outermembrane porin C, anti-CBir1, and ANCA were measured using enzyme-linked immunosorbent assays. Genotyping was performed using Illumina immunochip technology; an admixture rate was calculated for each subject. Multiple imputation by chained equations was performed to account for data missing at random. Logistic regression was used to calculate adjusted odds ratio (OR) for associations between serological markers and both complicated disease and disease requiring surgery. Results: A total of 358 patients were included in the analysis. The majority of our patients had inflammatory, noncomplicated disease (58.4%), perianal disease (55.7%), and documented colonic inflammation (86.8%). On multivariable analysis, both IgG ASCA and OmpC were associated with complicated disease (OR, 2.67; 95% CI, 1.67-4.28; OR, 2.23; 95% CI, 1.41-3.53, respectively) and disease requiring surgery (OR, 2.51; 95% CI, 1.49-4.22; OR, 3.57; 95% CI, 2.12-6.00). NE admixture to the African genome did not have any associations or interactions in relation to clinical outcome. Conclusions: Our study comprises the largest cohort of AAs with CD. The utility of serological markers for the prognosis of CD in NE applies equally to AA populations
Transverse-Longitudinal Asymmetry in the Quasielastic 3He→(e→, e′) Reaction
The transverse-longitudinal asymmetry ATL′ in 3He→(e→, e′) quasielastic scattering at momentum transfer Q2=0.14 (GeV/c)^2 has been measured to be 1.52 ± 0.55(stat) ± 0.15(syst)%. The plane wave impulse approximation (PWIA) prediction for this measurement ranges from 2.1% to 2.9%, where the variation is due to uncertainty in the initial state wave function, nucleon form factors, and off-shell prescription. The data may suggest a suppression with respect to the PWIA, which has also been observed for the unpolarized longitudinal response function
Conservation agriculture as a climate change mitigation strategy in Zimbabwe
There is a need to quantify agriculture’s potential to sequester carbon (C) to inform global approaches aimed at mitigating climate change effects. Many factors including climate, crop, soil management practices, and soil type can influence the contribution of agriculture to the global carbon cycle. The objective of this study was to investigate the C sequestration potential of conservation agriculture (CA) (defined by minimal soil disturbance, maintaining permanent soil cover, and crop rotations). This study used micrometeorological methods to measure carbon dioxide (CO2) flux from several alternative CA practices in Harare, central Zimbabwe. Micrometeorological methods can detect differences in total CO2 emissions of agricultural management practices; our results show that CA practices produce less CO2 emissions. Over three years of measurement, the mean and standard error (SE) of CO2 emissions for the plot with the most consistent CA practices was 0.564 ± 0.0122 g CO2 m−2 h−1, significantly less than 0.928 ± 0.00859 g CO2 m−2 h−1 for the conventional tillage practice. Fluorescent cyanine dyes for the quantification of low amounts of dsDNA
In this research six cyanine fluorophores for the quantification of dsDNA in the pg-ng range, without amplification, are compared under exactly identical conditions: EvaGreen, SYBR Green, PicoGreen, AccuClear, AccuBlue NextGen and YOYO-1. The fluorescence intensity as a function of the amount of dsDNA is measured at the optimal wavelengths for excitation and emission and for each dye the limit of detection and the response linearity at low levels of dsDNA are determined. No linear range was found for SYBR Green and YOYO-1 for pg-ng quantities of dsDNA. EvaGreen, PicoGreen, AccuClear and AccuBlue NextGen show good linearity in the pg-ng range. AccuClear exhibits the widest linear range of 3 pg–200 ng, whereas AccuBlue NextGen turned out to have the highest sensitivity of the tested dyes with a limit of detection of 50 pg
Neuropeptides in anterior pituitary development
Recent studies using biotechnological methods have achieved significant advances in our knowledge of molecular mechanisms underlying
pituitary gland development and the differentiation of pituitary cytotypes. A large number of neuropeptides have been reported in the adult
pituitary gland as well as in the central and peripheral nervous system. The early presence of neuropeptides during pituitary development is
reviewed here. Neuromedin U (NmU), galanin and the polypeptide 7B2 have been localised to different endocrine cells of the gland. Their
expression seems to be manifold even though it is temporally and spatially regulated. There is now firm immunocytochemical evidence
that neuropeptides are present during morphogenesis of the pituitary and can be present simultaneously with all pituitary hormones.
