A comparative approach to confirm antibiotic-resistant microbes in the cryosphere
Antibiotic-resistant microbes pose one of the biggest challenges of the current century. While areas with proximity to human impact are closely studied, a lot is yet to learn about antimicrobial resistance in remote regions like the cryosphere. Nowadays, antibiotic (AB) resistance is considered a pollution that has reached the Earth’s most pristine areas. However, monitoring of resistant environmental bacteria therein faces several challenges that inhibit scientific progress in this field. Due to many cultivation-based antibiotic susceptibility tests being optimized for mesophilic pathogenic microorganisms, many researchers opt for expensive molecular biological approaches to detect antibiotic resistance in the cryosphere. However, some disadvantages of these methods prohibit effective comprehensive monitoring of resistant bacteria in pristine areas, hence we suggest established cultivation-based approaches when looking for antimicrobial resistance in the cryosphere. In this study, we compared two common antibiotic susceptibility tests and optimized them to meet the needs of psychrophilic microorganisms. The resulting cultures thereof originated from cryospheric habitats with differing anthropogenic impacts. The results show that these methods are applicable to detect antibiotic resistance in cryospheric habitats and could potentially increase the comparability between studies
Hybridization-related correction to the jellium model for fullerenes
We introduce a new type of correction for a more accurate description of
fullerenes within the spherically symmetric jellium model. This correction
represents a pseudopotential which originates from the comparison between an
accurate ab initio calculation and the jellium model calculation. It is shown
that such a correction to the jellium model allows one to account, at least
partly, for the sp2-hybridization of carbon atomic orbitals. Therefore, it may
be considered as a more physically meaningful correction as compared with a
structureless square-well pseudopotential which has been widely used earlier.Comment: 16 pages, 10 figure
Two European Cornus L. feeding leafmining moths, Antispila petryi Martini, 1899, sp. rev. and A. treitschkiella (Fischer von Röslerstamm, 1843) (Lepidoptera, Heliozelidae): an unjustified synonymy and overlooked range expansion
Antispila treitschkiella (Fischer von Röslerstamm, 1843) and A. petryi Martini, 1899, sp. rev. were regarded as synonymous since 1978, but are shown to be two clearly separated species with different hostplants, life histories, DNA barcodes and morphology. Antispila treitschkiella feeds on Cornus mas L., is bivoltine, and has, by following its ornamentally planted host, greatly expanded its range in north-western Europe. In contrast A. petryi feeds on the widespread native C. sanguinea L., is univoltine, and is one of only two Antispila species previously resident in the British Isles, the Netherlands and northern Europe. Consequently, the increase in abundance of A. treitschkiella in the Netherlands since the early 1990s and in Great Britain in recent years must be regarded as part of a recent expansion into north-western Europe, whereas the native A. petryi is hardly expanding and less abundant. In Britain, detailed surveys of parks and living collections confirmed the monophagy of these two species. A search of British herbarium samples provided no evidence for an earlier date of establishment. Information on recognition of all stages, including DNA barcodes, and distribution is provided, and these two species are compared with the third European Cornus L. leafminer, A. metallella (Denis & Schiffermüller, 1775)

Kinase domain mutations confer resistance to novel inhibitors targeting JAK2V617F in myeloproliferative neoplasms
The transforming JAK2V617F kinase is frequently associated with myeloproliferative neoplasms (MPNs) and thought to be instrumental for the overproduction of myeloid lineage cells. Several small molecule drugs targeting JAK2 are currently in clinical development for treatment in these diseases. We performed a high-throughput in vitro screen to identify point mutations in JAK2V617F that would be predicted to have potential clinical relevance and associated with drug resistance to the JAK2 inhibitor ruxolitinib (INCB018424). Seven libraries of mutagenized JAK2V617F cDNA were screened to specifically identify mutations in the predicted drug-binding region that would confer resistance to ruxolitinib, using a BaF3 cell-based assay. We identified 5 different non-synonymous point mutations that conferred drug resistance. Cells containing mutations had a 9 to 33-fold higher EC50 for ruxolitinib compared to native JAK2V617F. Our results further indicated that these mutations also conferred cross-resistance to all JAK2 kinase inhibitors tested, including AZD1480, TG101348, lestaurtinib (CEP-701) and CYT-387. Surprisingly, introduction of the ‘gatekeeper’ mutation (M929I) in JAK2V617F affected only ruxolitinib sensitivity (4-fold increase in EC50). These results suggest that JAK2 inhibitors currently in clinical trials may be prone to resistance as a result of point mutations and caution should be exercised when administering these drugs
Anion vacancy driven magnetism in incipient ferroelectric SrTiO3 and KTaO3 nanoparticles
Based on our analytical results [http://arxiv.org/abs/1006.3670], we predict
that undoped nanoparticles (size <10-100nm) of incipient ferroelectrics without
any magnetic ions can become ferromagnetic even at room temperatures due to the
inherent presence of a new type of magnetic defects with spin S=1, namely
oxygen vacancies, where the magnetic triplet state is the ground state in the
vicinity of the surface (magnetic shell), while the nonmagnetic singlet is the
ground state in the bulk material (nonmagnetic core). Consideration of randomly
distributed magnetic spins (S=1) had shown that magnetic properties of
incipient ferroelectric nanoparticles are strongly size and temperature
dependent due to the size and temperature dependence of their dielectric
permittivity and the effective Bohr radius proportional to permittivity. The
phase diagrams in coordinates temperature - particle radius are considered. In
particular, for particle radii less that the critical radius ferromagnetic
long-range order appears in a shell region of thickness 5 - 50 nm once the
concentration of magnetic defects exceeds the magnetic percolation threshold.
The critical radius is calculated in the mean field theory from the condition
of the magnetic defects exchange energy equality to thermal energy. For
particle radii higher than critical value only the paramagnetic phase is
possible. The conditions of the super-paramagnetic state appearance in the
assembly of nanoparticles with narrow distribution function of their sizes are

Associated reading skills in children with a history of Specific Language Impairment (SLI)
A large cohort of 200 eleven-year-old children with Specific Language Impairment (SLI) were assessed on basic reading accuracy and on reading comprehension as well as language tasks. Reading skills were examined descriptively and in relation to early language and literacy factors. Using stepwise regression analyses in which age and nonverbal IQ were controlled for, it was found that a single word reading measure taken at 7 years was unsurprisingly a strong predictor of the two different types of reading ability. However, even with this measure included, a receptive syntax task (TROG) entered when reading accuracy score was the DV. Furthermore, a test of expressive syntax/narrative and a receptive syntax task completed at 7 years entered into the model for word reading accuracy. When early reading accuracy was excluded from the analyses, early phonological skills also entered as a predictor of both reading accuracy and comprehension at 11 years. The group of children with a history of SLI were then divided into those with no literacy difficulties at 11 and those with some persisting literacy impairment. Using stepwise logistic regression, and again controlling for IQ and age, 7 years receptive syntax score (but not tests of phonology, expressive vocabulary or expressive syntax/narrative) entered as a positive predictor of membership of the ‘no literacy problems’ group regardless of whether early reading accuracy was controlled for in step one. The findings are discussed in relation to the overlap of SLI and dyslexia and the long term sequelae of language impairment
Increased amino acids levels and the risk of developing of hypertriglyceridemia in a 7-year follow-up
BACKGROUND: Recently, five branched-chain and aromatic amino acids were shown to be associated with the risk of developing type 2 diabetes (T2D). AIM: We set out to examine whether amino acids are also associated with the development of hypertriglyceridemia. MATERIALS AND METHODS: We determined the serum amino acids concentrations of 1,125 individuals of the KORA S4 baseline study, for which follow-up data were available also at the KORA F4 7Â years later. After exclusion for hypertriglyceridemia (defined as having a fasting triglyceride level above 1.70Â mmol/L) and diabetes at baseline, 755 subjects remained for analyses. RESULTS: Increased levels of leucine, arginine, valine, proline, phenylalanine, isoleucine and lysine were significantly associated with an increased risk of hypertriglyceridemia. These associations remained significant when restricting to those individuals who did not develop T2D in the 7-year follow-up. The increase per standard deviation of amino acid level was between 26 and 40Â %. CONCLUSIONS: Seven amino acids were associated with an increased risk of developing hypertriglyceridemia after 7Â years. Further studies are necessary to elucidate the complex role of these amino acids in the pathogenesis of metabolic disorders
State attachment variability across distressing situations in middle childhood
Contemporary research suggests that attachment has both a trait-like, stable component, and a state-like component that varies across contexts. In the current study, we assessed state attachment variability across comparably distressing situations in middle childhood. In two samples, children reported their expectations of maternal support in each situation. Additionally, we administered trait attachment and psychological well-being measures. Results indicated that, overall, children varied in their expectations across situations: more than half of the variance was explained by intra-individual differences across situations. Results revealed two components underlying variability: a Signal-and-Support component reflecting expectations of support-seeking and receiving, and a Back-on-Track component reflecting expectations of stress reduction and comfort. State attachment variability was related to individual differences in trait attachment: children who are more securely attached at the trait level, overall appear to vary less in their state attachment, likely due to their high mean state attachment scores across situations. When the mean state attachment scores are accounted for, more securely attached children seem to vary more, suggesting that their state attachment expectations are more sensitive to contextual fluctuations. Importantly, degree of state attachment variability explained psychological well-being over and above trait attachment
Rearrangement of cluster structure during fission processes
Results of molecular dynamics simulations of fission reactions and are presented. Dependence
of the fission barriers on isomer structure of the parent cluster is analyzed.
It is demonstrated that the energy necessary for removing homothetic groups of
atoms from the parent cluster is largely independent of the isomer form of the
parent cluster. Importance of rearrangement of the cluster structure during the
fission process is elucidated. This rearrangement may include transition to
another isomer state of the parent cluster before actual separation of the
daughter fragments begins and/or forming a "neck" between the separating
fragments
