    The EMSO-Azores deep sea observatory: 8 years of operation

    The EMSO-Azores deep sea observatory is a component of the EMSO ERIC. It focuses on two main questions: What are the feedbacks between volcanism, deformation, seismicity, and hydrothermalism at a slow spreading mid-ocean ridge and how does the hydrothermal ecosystem couple with these sub-seabed processes? The infrastructure comprises 2 sea monitoring nodes, autonomous instruments and a set of site studies experiments. It has been deployed in 2010 in the Lucky Strike vent field and acquires multidisciplinary data since then.Peer Reviewe

    A dual sensor device to estimate fluid flow velocity at diffuse hydrothermal vents

In this study, we combine two methods to accurately estimate fluid flow velocities at diffuse flow areas. While the first method uses a hot film anemometer that performs high frequency measurements, the second allows a relatively rapid assessment of fluid flow velocity through video imagery and provides in situ data to calibrate the sensor. Measurements of flow velocities on hydrothermal diffuse flow areas were obtained on the Mid-Atlantic Ridge (MAR). They range from 1.1 to 4.9 mm/sec., at the substratum level, in low temperature (4.5 to 16.4°C) diffuse flow areas from the Tour Eiffel sulfide edifice. A strong correlation was observed between fluid flow velocities and temperature, supporting the possible use of temperature as a proxy to estimate flow rates in diffuse flow areas where such a simple linear flow/temperature relation is shown to dominate.The first part of this research was sponsored by a NOAA/NURP grant award #NA96RU0221 and NSF grant OCE-9901563 to MKT and JS.     Relationship between the occurrence of filamentous bacteria on Bathymodiolus azoricus shell and the physiological and toxicological status of the vent mussel

    En libre-accĂšs sur Archimer : http://archimer.ifremer.fr/doc/2009/publication-6821.pdfInternational audienceThe edifice walls of the Eiffel Tower hydrothermal vent site (Mid-Atlantic Ridge, Lucky Strike vent field) are populated with dense communities of dual symbioses harboring vent mussel Bathymodiolus azoricus, some of which are covered by white filamentous mats belonging to sulfur-oxidizing bacteria. Mussels were collected in both the presence and absence of the filamentous bacteria. A sample of the filamentous bacteria was collected and water measurements of temperature, CH4 and H2S were recorded at the collection area. The whole soft tissues were analyzed for total lipid, carbohydrate and total protein. Metallothioneins and metals (Cu, Fe and Zn) levels were determined in the major organs. The results showed no significant physiological and toxicological evidence that emphasizes the influence of associated sulfur-oxidizing filamentous bacteria on B. azoricus mussel shells. However, B. azoricus mussel seems to be well adapted to the assorted physico-chemical characteristics from the surrounding environment since it is able to manage the constant fluctuation of physico-chemical compounds

    Le texte intĂ©gral est accessible via Archimer: http://archimer.ifremer.fr/doc/2008/publication-3830.pdfInternational audienceThe distribution of Fe, Cu, Zn, Pb, Cd between the dissolved ( 2 ÎŒm) fractions was measured after in-situ filtration in two hydrothermal habitats. The total metal concentration ranges exhibit a clear enrichment compared with the seawater concentration, accounting for the hydrothermal input for all the metals considered. Iron is the predominant metal (5-50 ÎŒM) followed by Zn and Cu. Cd and Pb are present at the nM level. At the scale studied, the behavior of temperature, pH and dissolved iron is semi-conservative whereas the other dissolved and particulate metals are characterized by non-conservative patterns. The metal enrichment of the > 2 ÎŒm fraction results from the settlement and accumulation of particulate matter close to the organisms, acting as a secondary metal source. The enrichment observed in the dissolved fraction can be related to the dissolution or oxidation of particles (mainly polymetallic sulfide) or to the presence of small particles and large colloids not retained on the 2 ÎŒm frit. SEM observations indicate that the bulk particulate observed is characteristic of crystalline particles settling rapidly from the high temperature smoker (sphalerite, wurtzite and pyrite), amorphous structures and eroded particles formed in the external zone of the chimney. Precipitation of Zn, Cu, Cd and Pb with Fe as wurtzite, sphalerite and pyrite is the main process taking place within the area studied and is semi-quantitative. The distribution of the dominant observed fauna has been related to the gradient resulting from the dilution process, with the alvinellids worms colonizing the hotter and more variable part of the mixing zone, but also to the metallic load of the mixing zone. Dissolved and particulate metal concentrations are therefore necessary abiotic factors to be studied in a multiparametric approach to understand the faunal distribution in hydrothermal ecosystems

    Hydrothermal activity along the slow-spreading Lucky Strike ridge segment (Mid-Atlantic Ridge) : distribution, heatflux, and geological controls

The definitive version was published in Earth and Planetary Science Letters 431 (2015): 1730185, doi:10.1016/j.epsl.2015.09.025.We have reviewed available visual information from the seafloor, and recently acquired microbathymetry for several traverses across the Lucky Strike segment, to evaluate the distribution of hydrothermal activity. We have identified a new on-axis site with diffuse flow, Ewan, and anactive vent structure ~1.2 km from the axis, Capelinhos. These sites are minor relative to the Main field, and our total heatflux estimate for all active sites (200-1200 MW) is only slightly higher than previously published estimates. We also identify fossil sites W of the main Lucky Strike field. A circular feature ~200 m in diameter located on the flanks of a rifted off-axis central volcano, is likely a large and inactive hydrothermal edifice, named Grunnus. We find no indicator of focused hydrothermal activity elsewhere along the segment, suggesting that the enhanced melt supply and the associated melt lenses, required to form central volcanoes, also sustain hydrothermal circulation to form and maintain large and long-lived hydrothermal fields. Hydrothermal discharge to the seafloor occurs along fault traces, suggesting focusing of hydrothermal circulation in the shallow crust along permeable fault zones.This work has been partly financed by ANR (France) Mothseim Project NT05-3 42213 toJE, and by EU-RTN-MOMARNET to MC.     peer reviewedCattle are commonly subclinically infected following natural or experimental infection with bluetongue virus (BTV). The introduction of BTV serotype 8 (BTV-8) in Europe has been characterized by the manifestation of clinical signs in infected cattle. In order to study the pathogenesis of BTV-8 in this host, an animal model able to reproduce the clinical manifestations of the disease is required. In this work, two calves were subcutaneously and intravenously injected with a low passage cell-adapted strain of BTV-8. Both calves showed typical bluetongue clinical signs, including pyrexia, ocular discharge, conjunctivitis, oral mucosal congestion, development of ulcers and necrotic lesions on the lips and tongue, submandibular oedema, coronitis and oedema of the coronet and pastern region. A score was assigned depending on the severity of the lesions and a total clinical score was calculated for each animal daily and at the end of the experiment. Both calves became viraemic 24h post-infection and seroconversion occurred between 7 and 11 days P.I. In this study we present the development of a protocol of infection in calves able to reproduce the severity of the lesions observed with BTV-8 in field conditions

    Experimental infection of cattle with Mycobacterium tuberculosis isolates shows the attenuation of the human tubercle bacillus for cattle

    The Mycobacterium tuberculosis complex (MTBC) is the collective term given to the group of bacteria that cause tuberculosis (TB) in mammals. It has been reported that M. tuberculosis H37Rv, a standard reference MTBC strain, is attenuated in cattle compared to Mycobacterium bovis. However, as M. tuberculosis H37Rv was isolated in the early 1930s, and genetic variants are known to exist, we sought to revisit this question of attenuation of M. tuberculosis for cattle by performing a bovine experimental infection with a recent M. tuberculosis isolate. Here we report infection of cattle using M. bovis AF2122/97, M. tuberculosis H37Rv, and M. tuberculosis BTB1558, the latter isolated in 2008 during a TB surveillance project in Ethiopian cattle. We show that both M. tuberculosis strains caused reduced gross and histopathology in cattle compared to M. bovis. Using M. tuberculosis H37Rv and M. bovis AF2122/97 as the extremes in terms of infection outcome, we used RNA-Seq analysis to explore differences in the peripheral response to infection as a route to identify biomarkers of progressive disease in contrast to a more quiescent, latent infection. Our work shows the attenuation of M. tuberculosis strains for cattle, and emphasizes the potential of the bovine model as a ‘One Health’ approach to inform human TB biomarker development and post-exposure vaccine development
