    "Do I really want to do this?" Longitudinal cohort study participants' perspectives on postal survey design: a qualitative study

    <p>Abstract</p> <p>Background</p> <p>Randomised controlled trials have investigated aspects of postal survey design yet cannot elaborate on reasons behind participants' decision making and survey behaviour. This paper reports participants' perspectives of the design of, and participation in, a longitudinal postal cohort survey. It describes strengths and weaknesses in study design from the perspectives of study participants and aims to contribute to the: 1) design of future cohort surveys and questionnaires generally and, 2) design of cohort surveys for people with musculoskeletal disorders (MSDs) specifically.</p> <p>Methods</p> <p>In-depth interviews explored the design of postal surveys previously completed by participants. Interviews used open ended questioning with a topic guide for prompts if areas of interest were not covered spontaneously. Thematic data analysis was undertaken based on the framework method. A second researcher verified all coding.</p> <p>Results</p> <p>Data from fourteen interviews were analysed within three main themes; participation, survey design and survey content. One of the main findings was the importance of clear communication aimed at the correct audience both when inviting potential participants to take part and within the survey itself. Providing enough information about the study, having a topic of interest and an explanation of likely benefits of the study were important when inviting people to participate. The neutrality of the survey and origination from a reputable source were both important; as was an explanation about why information was being collected within the survey itself. Study findings included participants' impressions when invited to take part, why they participated, the acceptability of follow-up of non-responders and why participants completed the follow-up postal survey. Also discussed were participants' first impression of the survey, its length, presentation and participants' views about specific questions within the survey.</p> <p>Conclusions</p> <p>Ideas generated in this study provide an insight into participants' decision making and survey behaviour and may enhance the acceptability of future surveys to potential participants. As well as clear communication, participants valued incentives and survey questions that were relevant to them. However, opinions varied as to the preferred format for responses with some advising more opportunity for open-ended feedback. We also found that some standard format questions can raise quandaries for individual participants.</p

    Horizontally acquired glycosyltransferase operons drive salmonellae lipopolysaccharide diversity.

    The immunodominant lipopolysaccharide is a key antigenic factor for Gram-negative pathogens such as salmonellae where it plays key roles in host adaptation, virulence, immune evasion, and persistence. Variation in the lipopolysaccharide is also the major differentiating factor that is used to classify Salmonella into over 2600 serovars as part of the Kaufmann-White scheme. While lipopolysaccharide diversity is generally associated with sequence variation in the lipopolysaccharide biosynthesis operon, extraneous genetic factors such as those encoded by the glucosyltransferase (gtr) operons provide further structural heterogeneity by adding additional sugars onto the O-antigen component of the lipopolysaccharide. Here we identify and examine the O-antigen modifying glucosyltransferase genes from the genomes of Salmonella enterica and Salmonella bongori serovars. We show that Salmonella generally carries between 1 and 4 gtr operons that we have classified into 10 families on the basis of gtrC sequence with apparent O-antigen modification detected for five of these families. The gtr operons localize to bacteriophage-associated genomic regions and exhibit a dynamic evolutionary history driven by recombination and gene shuffling events leading to new gene combinations. Furthermore, evidence of Dam- and OxyR-dependent phase variation of gtr gene expression was identified within eight gtr families. Thus, as O-antigen modification generates significant intra- and inter-strain phenotypic diversity, gtr-mediated modification is fundamental in assessing Salmonella strain variability. This will inform appropriate vaccine and diagnostic approaches, in addition to contributing to our understanding of host-pathogen interactions

    A crossmodal crossover: opposite effects of visual and auditory perceptual load on steady-state evoked potentials to irrelevant visual stimuli

    Mechanisms of attention are required to prioritise goal-relevant sensory events under conditions of stimulus competition. According to the perceptual load model of attention, the extent to which task-irrelevant inputs are processed is determined by the relative demands of discriminating the target: the more perceptually demanding the target task, the less unattended stimuli will be processed. Although much evidence supports the perceptual load model for competing stimuli within a single sensory modality, the effects of perceptual load in one modality on distractor processing in another is less clear. Here we used steady-state evoked potentials (SSEPs) to measure neural responses to irrelevant visual checkerboard stimuli while participants performed either a visual or auditory task that varied in perceptual load. Consistent with perceptual load theory, increasing visual task load suppressed SSEPs to the ignored visual checkerboards. In contrast, increasing auditory task load enhanced SSEPs to the ignored visual checkerboards. This enhanced neural response to irrelevant visual stimuli under auditory load suggests that exhausting capacity within one modality selectively compromises inhibitory processes required for filtering stimuli in another

    YhdJ, a Nonessential CcrM-Like DNA Methyltransferase of Escherichia coli and Salmonella entericaā–æ

    The Caulobacter crescentus DNA adenine methyltransferase CcrM and its homologs in the Ī±-Proteobacteria are essential for viability. CcrM is 34% identical to the yhdJ gene products of Escherichia coli and Salmonella enterica. This study provides evidence that the E. coli yhdJ gene encodes a DNA adenine methyltransferase. In contrast to an earlier report, however, we show that yhdJ is not an essential gene in either E. coli or S. enterica

    West Florida Shelf Pipeline Serves as Sea Turtle Benthic Habitat Based on in Situ Towed Camera Observations

    The use of marine offshore benthic habitats by sea turtles is poorly characterized due to the difficulty of obtaining in situ data. Understanding benthic habitat use that is important to the speciesā€™ reproduction, foraging, and migrations is critical for guiding management decisions. A towed camera-based assessment survey system (C-BASS) equipped with environmental sensors was used to characterize and assess benthic habitats on the West Florida Shelf (WFS) from 2014 to 2018. During these cruises, sea turtles were opportunistically observed during the surveys, and critical in situ data such as spatiotemporal information, species identification, habitat use, behavior, and environmental data were collected and evaluated. In total, 79 sea turtles were observed during 97 transects of approximately 2700 km of seafloor, which was recorded on 380 h of video. Several sea turtle species were spotted within the WFS, including loggerhead Caretta caretta, Kempā€™s ridley Lepidochelys kempii, and green turtles Chelonia mydas. These opportunistic sightings revealed an area of high use on the WFS, an anthropogenic structure known as the Gulfstream natural gas pipeline (GSPL). C-BASS survey results suggest that 2 sea turtle species (C. caretta and L. kempii) utilize this artificial structure primarily as a resting area. We emphasize the importance of combining habitat mapping techniques (towed underwater video and multibeam bathymetry/backscatter) with tracking technology to better understand the fine-scale habitat use of sea turtles

    Heterogeneity of Gtr families localized within locus 2 of<i>Salmonella</i> serovars.

    <p>Represented is an alignment and sequence comparison of the <i>cysS</i> to <i>pheP</i> genomic region encompassing locus 2 (SPI-16, <a href="http://www.plosgenetics.org/article/info:doi/10.1371/journal.pgen.1003568#pgen-1003568-g003" target="_blank">Figure 3</a>) from nine <i>gtr</i> positive <i>Salmonella</i> serovars. The phage-associated locus 2 is defined by a pair of direct repeats (pink) and a 5ā€² Arg-tRNA <a href="http://www.plosgenetics.org/article/info:doi/10.1371/journal.pgen.1003568#pgen.1003568-Vernikos1" target="_blank">[25]</a>. Black coding sequences refer to those located within locus 2 boundaries, while white coding sequences are located outside locus 2. The <i>gtr</i> operon located within locus 2 is colored on the basis of GtrC family clustering (<a href="http://www.plosgenetics.org/article/info:doi/10.1371/journal.pgen.1003568#pgen-1003568-g001" target="_blank">Figure 1</a>). A different colored GtrC family likely reflects a functionally different <i>gtr</i> operon. <i>S.</i> Schwarzengrund contains a <i>gtr</i> positive lambda-like prophage and <i>S.</i> Dublin harbors a large remnant P22-like phage <a href="http://www.plosgenetics.org/article/info:doi/10.1371/journal.pgen.1003568#pgen.1003568-Villafane1" target="_blank">[11]</a> within locus 2. Regions of genetic similarity between genome sequences are shaded in grey and were determined by BLASTn analysis using Easyfig <a href="http://www.plosgenetics.org/article/info:doi/10.1371/journal.pgen.1003568#pgen.1003568-Sullivan1" target="_blank">[52]</a>.</p
