    Nutrition-Based Modulation of Poly-ADP-Ribosylation and its possible role in Alzheimer’s disease

    Alzheimer’s Disease (AD) is the most common neurodegenerative disease and the main reason of dementia in the elderly. On the pathological point of view, it is characterized by extracellular aggregates of amyloid peptides and intracellular deposits of tau protein. These deposits affect neuron viability and functions by inducing (among other pathological pathways) oxidative stress and triggering mitochondrial dysfunction. It is now evident that free radicalinduced oxidative damage is strongly involved in the pathogenesis of AD. Oxidative damage occurs early in disease pathogenesis and can exacerbate its progression. Post-mortem brain of individuals affected by AD, evidenced an extensive state of oxidative stress compared to healthy controls; markers of increased oxidation include, among others, DNA damage. DNA damage can induce the activity of the enzyme poly (ADP-ribose) polymerase 1 (PARP-1) that catalyze the reaction of poly (ADP-ribosylation). This post-translational modification modulates the functions of proteins involved in many physiological processes such as gene expression, maintenance of genomic stability and cell death. Therefore, inhibiting PARP-1 activity can represent a possible new strategy to reduce the impact of the oxidative stress in AD as well as in other neurodegenerative diseases.     Effect of ambient temperature on sleep breathing phenotype in mice: the role of orexins

    The loss of orexinergic neurons, releasing orexins, results in narcolepsy. Orexins participate in the regulation of many physiological functions, and their role as wake-promoting molecules has been widely described. Less is known about the involvement of orexins in body temperature and respiratory regulation. The aim of this study was to investigate whether orexin peptides modulate respiratory regulation as a function of ambient temperature (T°a) during different sleep stages. Respiratory phenotype of male orexin knockout (KO-ORX, n=9) and wild-type (WT, n=8) mice was studied at thermoneutrality (T°a=30°C) or during mild cold exposure (T°a=20°C) inside a whole-body plethysmography chamber. The states of wakefulness (W), non-rapid-eye-movement sleep (NREMS) and rapid-eye-movement sleep (REMS) were scored non-invasively, using a previously validated technique. Both in WT and KO-ORX mice T°a strongly and significantly affected ventilatory period and minute ventilation values during NREMS and REMS; moreover, the occurrence rate of sleep apneas in NREMS was significantly reduced at T°a=20°C compared to T°a=30°C. Overall, there were no differences in respiratory regulation during sleep between WT and KO-ORX mice, except for sigh occurrence rate, which was significantly increased at T°a=20°C with respect to T°a =30°C in WT mice, but not in KO-ORX mice. These results do not support a main role for orexin peptides in the temperature-dependent modulation of respiratory regulation during sleep. However, we showed that the occurrence rate of sleep apneas critically depends on T°a, without any significant effect of orexin peptides

    Effect of boat noise on the behaviour of bluefin tuna Thunnus thynnus in the Mediterranean Sea

    The effect of boat noise on the behaviour of bluefin tuna Thunnus thynnus was investigated in the Egadi Islands, Sicily, during spring 2005 using a fixed tuna trap set near shipping routes. Tuna behaviour was observed when exposed to both natural ambient sound and sound generated by hydrofoil passenger ferries, small boats and large car ferries. Acoustical and behavioural analyses were conducted with and without extraneous sound to define a list of behavioural categories. Each vessel produced different engine sounds with regard to their composition and bandwidth, and all were distinctly different from ambient sound levels. In the absence of boat noise, tuna assumed a concentrated coordinated school structure with unidirectional swimming and without a precise shape. When a car ferry approached, tuna changed swimming direction and increased their vertical movement toward surface or bottom; the school exhibited an unconcentrated structure and uncoordinated swimming behaviour. Hydrofoils appeared to elicit a similar response, but for shorter periods. Agonistic behaviour was more evident when exposed to sounds from outboard motors of small boats. This study showed that local noise pollution generated by boats produced behavioural deviations in tuna schools. Schooling enhances tuna homing accuracy during their spawning migration, and an alteration in schooling behaviour can affect the accuracy of their migration to spawning and feeding grounds

    Bioactivity of gallic acid–conjugated silica nanoparticles against Paenibacillus larvae and their host, Apis mellifera honeybee

    The aim of this work was to evaluate antimicrobial activity against Paenibacillus larvae and oral toxicity against workers and larvae of Apis mellifera of gallic acid (GA) and two nanohybrids of GA and silica. Also, the physicochemical, structural, and energetic properties of GA and the nanohybrids were determined through structure–activity relationship (SAR). The minimum inhibitory concentration (MIC) against P. larvae was determined. GA showed MIC values between 62.5 and 125 μg/ml, whereas the nanoparticle functionalized through the GA carboxylic moiety (NP2) showed the best antimicrobial activity with a MIC value of 23 μg GA/ml for four of the five isolates used. SAR analysis showed that electronegativity, chemical hardness, and dipolar moment are reliable estimators of the antimicrobial activity. NP2 showed the lowest toxicity against workers and was innocuous for bee larvae. Therefore, the nanohybrid NP2 was the best antibacterial and resulted in non-toxic against workers and larvae of honeybees, becoming a potentially effective and safe agent for the treatment of American Foulbrood disease.Fil: Dominguez, Enzo.     The paradox of an unpolluted coastal site facing a chronically contaminated industrial area

    Present and past industrial activities in coastal areas have left us a legacy of contamination and habitat degradation with potential implications for human health. Here, we investigated a coastal marine area enclosed in a Site of National Interest (SNI) of the central-western Adriatic (Mediterranean Sea), where priority actions of environmental remediation are required by governmental laws due the high environmental and human risk, and that is off-limits to any human activity since 2002. In particular, our investigation was focused on an area located in front of a chemical industry dismissed more than 3 decades ago. We report that the concentrations of heavy-metal and organic contaminants in the investigated sediments were generally lower than those expected to induce detrimental biological effects. Meiofaunal abundance, biomass and community structure changed among stations, but regardless of the distance from the abandoned industrial plant. Taxa richness within the SNI did not change significantly compared to the controls and the lack of some taxa in the SNI transects was not due to the contamination of the SNI area. The results of this study suggest a natural recovery of the marine area over 2 decades of restrictions on human activities, including fishing and shipping bans. If the hypothesis of the natural recovery of this SNI will be further confirmed by other studies, the plans forthe identification and monitoring of the most polluted areas in Italy should necessarily be redefined also in the light of the Water Framework, the Marine Strategy Framework and the Environmental Quality Standard Directives

    Sleep and Tibialis Anterior Muscle Activity in Mice With Mild Hypoxia and Iron Deficiency: Implications for the Restless Legs Syndrome

    Restless legs syndrome (RLS) is a neurological disorder that entails an urge to move with a circadian pattern during the evening/night. RLS may be accompanied by decreased sleep time and increased occurrence of periodic leg movements during sleep (PLMS), which involve bursts of tibialis anterior (TA) muscle electromyogram (EMG). Mild hypoxia and non-anemic iron deficiency, a highly prevalent nutritional deficiency, are relatively unexplored factors in RLS pathophysiology. We tested whether mice exposed to mild hypoxia, alone or in combination with non-anemic iron deficiency, show decreased sleep time particularly in the light (rest) period and increased occurrence of TA EMG phasic events similar to human PLMS. Female C57BL/6J mice were fed diets with low or normal iron for 6 months from weaning and instrumented with electrodes to record the electroencephalogram and the EMG of both TA muscles. Mice were recorded in a whole-body plethysmograph while breathing a normoxic or mildly hypoxic (15% O2) gas mixture for 48 h. Hypoxia increased minute ventilation during sleep. The low-iron diet decreased liver and serum iron, leaving blood hemoglobin and brainstem iron levels unaffected. Hypoxia, either alone or in combination with non-anemic iron deficiency, decreased non-rapid-eye-movement (non-REM) sleep time, but this occurred irrespective of the light/dark period and was not associated with increased occurrence of TA EMG events during non-REM sleep. These results do not support the hypothesis that mild hypoxia is sufficient to cause signs of RLS, either alone or in combination with non-anemic iron deficiency, pointing to the necessity of further susceptibility factors

    A Systematic Review of Social Factors and Suicidal Behavior in Older Adulthood

    Suicide in later life is a global public health problem. The aim of this review was to conduct a systematic analysis of studies with comparison groups that examined the associations between social factors and suicidal behavior (including ideation, non-fatal suicidal behavior, or deaths) among individuals aged 65 and older. Our search identified only 16 articles (across 14 independent samples) that met inclusion criteria. The limited number of studies points to the need for further research. Included studies were conducted in Canada (n = 2), Germany (n = 1), Hong Kong (n = 1), Japan (n = 1), Singapore (n = 1), Sweden (n = 2), Taiwan (n = 1), the U.K. (n = 2), and the U.S. (n = 3). The majority of the social factors examined in this review can be conceptualized as indices of positive social connectedness—the degree of positive involvement with family, friends, and social groups. Findings indicated that at least in industrialized countries, limited social connectedness is associated with suicidal ideation, non-fatal suicidal behavior, and suicide in later life. Primary prevention programs designed to enhance social connections as well as a sense of community could potentially decrease suicide risk, especially among men

    L'elaborato ha ad oggetto la disciplina della legge n. 40 del 2004, in materia di procreazione medicalmente assistita. A seguito di un'analisi della normativa originaria, sopratutto con riferimento ai principi costituzionali coinvolti in materia, si prosegue esaminando le pronunce della Corte costituzionale sul tema, le quali hanno portato ad una riscrittura della legge che tenesse conto dei valori fondamentali della Costituzione italiana. Segue una riflessione su alcune tematiche eticamente più complesse, quali l'omogenitorialità e la surrogazione di maternità. L'obiettivo dell'elaborato, volgendo uno sguardo anche al diritto straniero, è quello di valutare se il bilanciamento effettuato dal legislatore del 2004 sia quello più ragionevole, così come se i plurimi interventi della Corte costituzionale abbiano, davvero, adeguato la normativa all'odierna coscienza sociale
