    Used food oils: physical-chemical indicators of quality degradation

Jelgava, LLUUsed food oil (UFO), designated as frying oil, is a residue. Degradation by reuse or during storage, may occur by contacting, chemical, enzymatic and microbiological pathways, but oxidation is a major concern of the industry, as it affects sensory and nutritional quality of edible oils, with potentially toxic compounds formation. In Portugal, UFO's main destination still is the sewerage system, an environmental problem and waste of raw material, which can be re-qualified for non-food uses. However, quality control applied to UFO's, often results into expensive analysis inappropriate for small laboratories and catering industry. This project, developed with the Musketeers Group Portugal co-promotion (2012-2016), aimed to identify low-cost physicochemical parameters for further implementation as UFO's Quality Degradation Indicators (QDI) indicating defects quickly and accurately. UFO's analysis was tested on the use, for industrial frying, and by degradation induced in the laboratory (frying and heat stability tests) by applying following parameters: moisture, water activity (a(w)), total acidity, peroxide index, iodine index, colour (CIE, CIE Lab), UV absorbency, total polar compounds and microbiological indicators. Internal procedures (ESAS) were validated, redefining working ranges and test conditions, as standards procedures did not provide reliable results for the entire life cycle of oils, whose profile changes with time and reuse. Results demonstrate significant differences with quick response parameters as Total Acidity, Peroxide Index and CIE Lab colour, outlined as QDI's. Moisture, aw and CIE Lab colour proved to be inadequate for this purpose.     A Genetic Code Alteration Is a Phenotype Diversity Generator in the Human Pathogen Candida albicans

    BACKGROUND: The discovery of genetic code alterations and expansions in both prokaryotes and eukaryotes abolished the hypothesis of a frozen and universal genetic code and exposed unanticipated flexibility in codon and amino acid assignments. It is now clear that codon identity alterations involve sense and non-sense codons and can occur in organisms with complex genomes and proteomes. However, the biological functions, the molecular mechanisms of evolution and the diversity of genetic code alterations remain largely unknown. In various species of the genus Candida, the leucine CUG codon is decoded as serine by a unique serine tRNA that contains a leucine 5'-CAG-3'anticodon (tRNA(CAG)(Ser)). We are using this codon identity redefinition as a model system to elucidate the evolution of genetic code alterations. METHODOLOGY/PRINCIPAL FINDINGS: We have reconstructed the early stages of the Candida genetic code alteration by engineering tRNAs that partially reverted the identity of serine CUG codons back to their standard leucine meaning. Such genetic code manipulation had profound cellular consequences as it exposed important morphological variation, altered gene expression, re-arranged the karyotype, increased cell-cell adhesion and secretion of hydrolytic enzymes. CONCLUSION/SIGNIFICANCE: Our study provides the first experimental evidence for an important role of genetic code alterations as generators of phenotypic diversity of high selective potential and supports the hypothesis that they speed up evolution of new phenotypes

    Distribuição dos genótipos do vírus GB-C (HGV) em indivíduos da cidade de São Paulo, Brasil

    Há na literatura vários estudos filogenéticos e de distribuição de genótipos do chamado "Vírus GB-C" ou da "Hepatite G", mais conhecido pela dupla sigla "GBV-C/HGV". Ocorre que, em sua grande maioria, estas pesquisas foram realizadas com amostras de grupos ligados epidemiologicamente e não com indivíduos representativos da população geral. O presente estudo é uma continuação do primeiro trabalho no Brasil feito com este tipo de amostragem. Trata-se de análise filogenética e distribuição genotípica do GBV-C/HGV a partir de amostras isoladas dentre mais de 1.000 indivíduos da cidade de São Paulo. Para tanto, um fragmento de 728 pares de base da região 5' não-codificadora (5´NCR) do genoma viral, de 24 amostras, foi sequenciado e submetido à analise filogenética. Foram identificados os genótipos 1, 2a e 2b nas respectivas freqüências: 8,3% (2/24), 50% (12/24) e 41,7% (10/24). Concluindo, São Paulo apresenta uma distribuição de genótipos semelhante à publicada para outros estados e regiões do Brasil, endossando a idéia de que os tipos 1 e 2 teriam vindo com os africanos e europeus, respectivamente, e portanto estariam na população do país desde a sua formação.There has been several studies worldwide on phylogenetics and genotype distribution of the GB-virus C / Hepatitis G virus (GBV-C/HGV). However, in their great majority, those investigations were based on some epidemiologically linked group, rather than on a representative sampling of the general population. The present is a continuation of the first study in Brazil with such a population; it addresses the GBV-C/HGV phylogenetics and genotype distribution based on samples identified among more than 1,000 individuals of the city of São Paulo. For this purpose, a 728 bp fragment of the 5´non-coding region (5´NCR) of the viral genome, from 24 isolates, was sequenced and subjected to phylogenetic analysis. Genotypes 1, 2a and 2b were found at 8.3% (2/24), 50% (12/24) and 41.7% (10/24), respectively. In conclusion São Paulo displays a genotype distribution similar to the published data for other States and Regions of Brazil, endorsing the notion that types 1 and 2 would have entered the country with African and European people, respectively, since its earliest formation

    The evaluation of laser weldability of the third-generation advanced high strength steel

    To meet the demands of vehicular safety and greenhouse gas emission reduction, the automotive industry is increasingly using advanced high strength steels (AHSS) in the production of the components. With the development of the new generation of AHSS, it is essential to study their behavior towards manufacturing processes used in the automotive industry. For this purpose, the welding capability of newly developed third-generation Gen3 980T steel was investigated using the Nd:YAG (Neodymium:Yittrium Aluminum Garnet) laser-welding with different parameter conditions. The analysis was made by uniaxial tensile tests, micro-hardness, Scanning Electron Microscopy (SEM) and X-Ray Diffraction (XRD). The criteria used to evaluate the quality of the weld were the distance between the fracture and the weld bead and the surface finish. A relationship between the quality of the weld and the energy density was observed, expressed by a partial penetration for values below the optimal, and by irregularities in the weld bead and a high number of spatters for the values above the optimal.publishe

    Development of a device compatible with universal testing machine to perform hole expansion and Erichsen cupping tests

    The material characterization regarding sheet metal formability is usually assessed by the forming limit curve (FLC). The FLC requires specialized and expensive equipment, several samples with different geometries, and can be a very time-consuming procedure for data treatment. Alternatively, the hole expansion test (HET) and the Erichsen Cupping Test (ECT) can be used for routine evaluations of sheet metal mechanical behavior. These formability tests require fewer quantities of material and easy specimen preparation with a fast analysis of results. The HET and ECT procedures provide a proper evaluation of sheet material stretch-flangeability, formability, strength, and ductility. In this work, we developed a low-cost mechanical device capable of performing the HET and ECT tests using a universal testing machine. The equipment is designed to meet the test parameters set by ISO 16630 (HET) and ISO 20482 (ECT) standards. In order to verify its functionality, tests were carried out with the dual-phase steels DP600 and DP780. The corresponding values determined for the hole expansion ratio and the Erichsen index provided reliable results in terms of the accuracy and repeatability of the proposed testing device.publishe

    The importance of Brazilian Conservation Units for the diversity of gall-inducing insects: a study on gall-inducing insect richness in the Chapada Diamantina National Park, state of Bahia, Brazil

    Abstract Conservation Units (CUs) tend to have a high richness of herbivorous insects, including gall-inducing insects. Despite this, gall surveys carried out in these environments are punctual and some units have never had their galls investigated, such as the Chapada Diamantina National Park, Bahia (Chapada Diamantina Parna). Aiming to reduce this gap and contribute to future studies in CUs, this study aimed to survey the galls of the Chapada Diamantina Parna, Lençóis, as well as to investigate trends in research on galls in CUs in Brazil. For that, collections were carried out on monthly trips for one year. Published gall surveys were compiled. A total of 107 morphotypes induced in 88 host species were recorded. Most galls are formed in leaves, globoid in shape, green in color, and induced by Cecidomyiidae. This park has a relatively high richness of galls compared to other CUs, demonstrating its importance in the conservation of gall-inducing insects. The results also revealed that the number of surveys has been increasing over the years and that the Southeast concentrates the largest number of studies, a region that also gathers the largest number of specialists, demonstrating a geographic bias in the data

    Dietary whey protein lessens several risk factors for metabolic diseases: a review

    Obesity and type 2 diabetes mellitus (DM) have grown in prevalence around the world, and recently, related diseases have been considered epidemic. Given the high cost of treatment of obesity/DM-associated diseases, strategies such as dietary manipulation have been widely studied; among them, the whey protein diet has reached popularity because it has been suggested as a strategy for the prevention and treatment of obesity and DM in both humans and animals. Among its main actions, the following activities stand out: reduction of serum glucose in healthy individuals, impaired glucose tolerance in DM and obese patients; reduction in body weight; maintenance of muscle mass; increases in the release of anorectic hormones such as cholecystokinin, leptin, and glucagon like-peptide 1 (GLP-1); and a decrease in the orexigenic hormone ghrelin.
