Insights into the Roles of Gut Microbes in Obesity
Obesity is a major public health issue as it enhances the risk of suffering several chronic diseases of increasing prevalence. Obesity results from an imbalance between energy intake and expenditure, associated with a chronic low-grade inflammation. Gut microbes are considered to contribute to body weight regulation and related disorders by influencing metabolic and immune host functions. The gut microbiota as a whole improves the host's ability to extract and store energy from the diet leading to body weight gain, while specific commensal microbes seem to exert beneficial effects on bile salt, lipoprotein, and cholesterol metabolism. The gut microbiota and some probiotics also regulate immune functions, protecting the host form infections and chronic inflammation. In contrast, dysbiosis and endotoxaemia may be inflammatory factors responsible for developing insulin resistance and body weight gain. In the light of the link between the gut microbiota, metabolism, and immunity, the use of dietary strategies to modulate microbiota composition is likely to be effective in controlling metabolic disorders. Although so far only a few preclinical and clinical trials have demonstrated the effects of specific gut microbes and prebiotics on biological markers of these disorders, the findings indicate that advances in this field could be of value in the struggle against obesity and its associated-metabolic disorders
Bacteroides uniformis CECT 7771 ameliorates metabolic and immunological dysfunction in mice with high-fat-diet induced obesity
Background: Associations have been made between obesity and reduced intestinal numbers of members of the phylum Bacteroidetes, but there is no direct evidence of the role these bacteria play in obesity. Herein, the effects of Bacteroides uniformis CECT 7771 on obesity-related metabolic and immune alterations have been evaluated. Methods and Findings: Adult (6–8 week) male wild-type C57BL-6 mice were fed a standard diet or a high-fat-diet HFD to induce obesity, supplemented or not with B. uniformis CECT 7771 for seven weeks. Animal weight was monitored and histologic, biochemical, immunocompetent cell functions, and features of the faecal microbiota were analysed after intervention. The oral administration of B. uniformis CECT 7771 reduced body weight gain, liver steatosis and liver cholesterol and triglyceride concentrations and increased small adipocyte numbers in HFD-fed mice. The strain also reduced serum cholesterol, triglyceride, glucose, insulin and leptin levels, and improved oral tolerance to glucose in HFD fed mice. The bacterial strain also reduced dietary fat absorption, as indicated by the reduced number of fat micelles detected in enterocytes. Moreover, B. uniformis CECT 7771 improved immune defence mechanisms, impaired in obesity. HFD-induced obesity led to a decrease in TNF-a production by peritoneal macrophages stimulated with LPS, conversely, the administration of B. uniformis CECT 7771 increased TNF-a production and phagocytosis. Administering this strain also increased TNF-a production by dendritic cells (DCs) in response to LPS stimulation, which was significantly reduced by HFD. B. uniformis CECT 7771 also restored the capacity of DCs to induce a T-cell proliferation response, which was impaired in obese mice. HFD induced marked changes in gut microbiota composition, which were partially restored by the intervention. Conclusions: Altogether, the findings indicate that administration of B. uniformis CECT 7771 ameliorates HFD-induced metabolic and immune dysfunction associated with intestinal dysbiosis in obese mice.Fil: Gauffin Cano, María Paola. Microbiota and host determinants of behavioural phenotype in maternally separated mice
Early-life stress is a determinant of vulnerability to a variety of disorders that include dysfunction of the brain and gut. Here we exploit a model of early-life stress, maternal separation (MS) in mice, to investigate the role of the intestinal microbiota in the development of impaired gut function and altered behaviour later in life. Using germ-free and specific pathogen-free mice, we demonstrate that MS alters the hypothalamic-pituitary-adrenal axis and colonic cholinergic neural regulation in a microbiota-independent fashion. However, microbiota is required for the induction of anxiety-like behaviour and behavioural despair. Colonization of adult germ-free MS and control mice with the same microbiota produces distinct microbial profiles, which are associated with altered behaviour in MS, but not in control mice. These results indicate that MS-induced changes in host physiology lead to intestinal dysbiosis, which is a critical determinant of the abnormal behaviour that characterizes this model of early-life stress
The management of bronchus intermedius complications after lung transplantation: A retrospective study
<p>Abstract</p> <p>Background</p> <p>Airway complications following lung transplantation remain a significant cause of morbidity and mortality. The management of bronchial complications in Bronchus Intermedius (BI) is challenging due to the location of right upper bronchus. The aim of this study was to analyze the results of BI Montgomery T-tube stent in a consecutive patients with lung transplantations.</p> <p>Methods</p> <p>Between January 2007 and December 2010, 132 lung transplantations were performed at Foch Hospital, Suresnes, France. All the patients who had BI Montgomery T-tube after lung transplantation were included in this retrospective study. The demographic and interventional data and also complications were recorded.</p> <p>Results</p> <p>Out of 132 lung transplant recipients, 12 patients (9 male and 3 female) were entered into this study. The indications for lung transplantation were: cystic fibrosis 8 (67%), emphysema 3 (25%), and idiopathic pulmonary fibrosis 1 (8%). Most of the patients (83%) had bilateral lung transplantation. The mean interval between lung transplantation and interventional bronchoscopy was 11.5 ± 9.8 (SD) months. There was bronchial stenosis at the level of BI in 7 patients (58.3%). The Montgomery T-tube number 10 was used in 9 patients (75%). There was statistically significant difference in Forced Expiratory Volume in one second (FEV1) before and after stent placement (p = 0.01). The most common complication after stent placement was migration (33%).</p> <p>Conclusion</p> <p>BI complications after lung transplantation are still a significant problem. Stenosis or malacia following lung transplantation could be well managed with modified Montgomery T-tube.</p
Gut Microbiota Dysbiosis Is Associated with Inflammation and Bacterial Translocation in Mice with CCl4-Induced Fibrosis
BACKGROUND: Gut is the major source of endogenous bacteria causing infections in advanced cirrhosis. Intestinal barrier dysfunction has been described in cirrhosis and account for an increased bacterial translocation rate. HYPOTHESIS AND AIMS: We hypothesize that microbiota composition may be affected and change along with the induction of experimental cirrhosis, affecting the inflammatory response. ANIMALS AND METHODS: Progressive liver damage was induced in Balb/c mice by weight-controlled oral administration of carbon tetrachloride. Laparotomies were performed at weeks 6, 10, 13 and 16 in a subgroup of treated mice (n = 6/week) and control animals (n = 4/week). Liver tissue specimens, mesenteric lymph nodes, intestinal content and blood were collected at laparotomies. Fibrosis grade, pro-fibrogenic genes expression, gut bacterial composition, bacterial translocation, host's specific butyrate-receptor GPR-43 and serum cytokine levels were measured. RESULTS: Expression of pro-fibrogenic markers was significantly increased compared with control animals and correlated with the accumulated dose of carbon tetrachloride. Bacterial translocation episodes were less frequent in control mice than in treated animals. Gram-positive anaerobic Clostridia spp count was decreased in treated mice compared with control animals and with other gut common bacterial species, altering the aerobic/anaerobic ratio. This fact was associated with a decreased gene expression of GPR43 in neutrophils of treated mice and inversely correlated with TNF-alpha and IL-6 up-regulation in serum of treated mice along the study protocol. This pro-inflammatory scenario favoured blood bacterial translocation in treated animals, showing the highest bacterial translocation rate and aerobic/anaerobic ratio at the same weeks. CONCLUSIONS: Gut microbiota alterations are associated with the development of an inflammatory environment, fibrosis progression and bacterial translocation in carbon tetrachloride-treated mice
Influencia de la microbiota intestinal en la obesidad
La prevalencia de la obesidad es cada vez mayor y especialmente en niños y adolescentes.
En el desarrollo de la obesidad interviene la dieta, factores genéticos, endócrinos, psicológicos y ambientales. El primer objetivo específico fue determinar la influencia de una intervención para el tratamiento de la obesidad sobre la microbiota intestinal de adolescentes obesos y con sobrepeso. Mediante las técnicas de la reacción en cadena de la polimerasa a tiempo real (q-PCR) y de hibridación con fluorescencia in situ (FISH) se cuantificaron los principales grupos bacterianos del tracto intestinal. Los adolescentes que experimentaron mayor perdida de peso, presentaron mayor cantidad de bacterias de los grupos Bacteroides fragilis, Clostridium leptum, Bifidobacterium catenulatum, Lactobacillus, y menor de los grupos y especies de Bifidobacterium, Lactobacillus, Clostridium coccoides, B. breve y B.bifidum, antes y después de la intervención con respecto al grupo de menor pérdida de peso.
El segundo objetivo fue evaluar la microbiota intestinal de mujeres embarazadas con sobrepeso debido a que su microbiota influye en la microbiota del hijo. Con el análisis de la microbiota por qPCR y de parámetros bioquímicos se obtuvo que las mujeres con sobrepeso presentaron menor cantidad de Bifidobacterias y Bacteroides, y mayor cantidad de Staphylococcus, Enterobacterias y E.coli. que en el grupo normopeso. El número de Staphylococcus presentó una correlación positiva con el colesterol total, Bacteroides positiva con el colesterol HDL y con el ácido fólico, y negativa con los triglicéridos, el número de Bifidobacteriums presentó una correlación positiva con el ácido fólico, y los de Enterobacteriaceae y E.coli presentaron una correlación positiva con ferritina y negativa con transferrina. Por todo ello se puede decir, que la microbiota intestinal esta relacionada con el peso corporal, con la ganancia de peso y los parámetros metabólicos durante el embarazo, lo cual puede serSantacruz López, YA. Impregnación de lactobacillus en productos de manzana
Al iniciarse el nuevo milenio en Japón surge una nueva área
de la ciencia de los alimentos y la nutrición, la de los llamados,
"alimentos funcionales", los cuales han sido modificados en su composición
o su biodisponibilidad esta enfocada a una función benéfica
para el organismo. Los componentes de los alimentos enriquecidos
(probióticos, prebióticos y simbiontes) se hallan también
en los alimentos convencionales. La propiedad funcional está relacionada
con el papel metabólico, estructural o fisiológico del organismo
y no con la capacidad de tratar una patología. Los alimentos funcionales
pueden prevenir pero no curar enfermedades.
Los componentes probióticos de un alimento se definen como "la mezcla
o el microorganismo(s) aplicado al hombre o al animal que afecta
benéficamente al huésped mejorando las propiedades de la flora
intestinal". La mayoría de las especies comúnmente usadas para
la preparación de los probióticos son: Bifidobacterium
Lactobacillus Enterococcus y Propionibacterium, en su mayoría son
cepas intestinales. Se ha observado que los organismos probióticos
pueden penetrar y permanecer viables en sustancias porosas, como las frutas.
En el presente trabajo se realizó la impregnación al vacío
de Lactobacillus en trozos de manzana fresca, para combinar los efectos
benéficos de los probióticos con los ofrecidos por las frutas.
Después de analizar 4 formas geométricas diferentes, se trabajo
con la forma de moneda debido a que presentó una mejor porosidad efectiva.
Para la impregnación al vacío se utilizó como medio
de impregnación jugo de manzana comercial y leche fermentada Yakult
con una población estimada de 107 o 108 ufc/mL de Lactobacillus casei.
Las manzanas después de la impregnación presentó un
recuento alrededor de 106 ufc/g. Para asegurar la estabilidad del producto,
los cilindros impregnados se sometieron a dos diferentes tipos de secado
para su preservación. El primero fue el secado por liofilización
o sublimación el cual se realizó por 24 hrs. a 25° C y
10 m de Hg. El segundo fue con aire caliente a 35 y 40° C durante 20
horas.
Se observó que utilizando el primer método de secado se obtiene
un recuento del orden de 106 ufc/g y por el segundo método se obtiene
107 ufc/g de lactobacilos. Se realizaron pruebas sensoriales con escala
hedónica para conocer el agrado de los productos. Los resultados
demuestran que los productos secados por liofilización son preferidos
por su color. Las demás características sensoriales no mostraron
una diferencia evidente
