High spatial resolution and high contrast optical speckle imaging with FASTCAM at the ORM
In this paper, we present an original observational approach, which combines,
for the first time, traditional speckle imaging with image post-processing to
obtain in the optical domain diffraction-limited images with high contrast
(1e-5) within 0.5 to 2 arcseconds around a bright star. The post-processing
step is based on wavelet filtering an has analogy with edge enhancement and
high-pass filtering. Our I-band on-sky results with the 2.5-m Nordic Telescope
(NOT) and the lucky imaging instrument FASTCAM show that we are able to detect
L-type brown dwarf companions around a solar-type star with a contrast DI~12 at
2" and with no use of any coronographic capability, which greatly simplifies
the instrumental and hardware approach. This object has been detected from the
ground in J and H bands so far only with AO-assisted 8-10 m class telescopes
(Gemini, Keck), although more recently detected with small-class telescopes in
the K band. Discussing the advantage and disadvantage of the optical regime for
the detection of faint intrinsic fluxes close to bright stars, we develop some
perspectives for other fields, including the study of dense cores in globular
clusters. To the best of our knowledge this is the first time that high

Correlational Study of Aminopeptidase Activities between Left or Right Frontal Cortex versus the Hypothalamus, Pituitary, Adrenal Axis of Spontaneously Hypertensive Rats Treated with Hypotensive or Hypertensive Agents.
It has been suggested that the neuro-visceral integration works asymmetrically and that this asymmetry is dynamic and modifiable by physio-pathological influences. Aminopeptidases of the renin-angiotensin system (angiotensinases) have been shown to be modifiable under such conditions. This article analyzes the interactions of these angiotensinases between the left or right frontal cortex (FC) and the same enzymes in the hypothalamus (HT), pituitary (PT), adrenal (AD) axis (HPA) in control spontaneously hypertensive rats (SHR), in SHR treated with a hypotensive agent in the form of captopril (an angiotensin-converting enzyme inhibitor), and in SHR treated with a hypertensive agent in the form of the L-Arginine hypertensive analogue L-NG-Nitroarginine Methyl Ester (L-NAME). In the control SHR, there were significant negative correlations between the right FC with HPA and positive correlations between the left FC and HPA. In the captopril group, the predominance of negative correlations between the right FC and HPA and positive correlations between the HPA and left FC was maintained. In the L-NAME group, a radical change in all types of interactions was observed; particularly, there was an inversion in the predominance of negative correlations between the HPA and left FC. These results indicated a better balance of neuro-visceral interactions after captopril treatment and an increase in these interactions in the hypertensive animals, especially in those treated with L-NAME
CHK1 expression in gastric cancer is modulated by p53 and RB1/E2F1: Implications in chemo/radiotherapy response
Radiation has a limited but relevant role in the adjuvant therapy of gastric cancer (GC) patients. Since Chk1 plays a critical function in cellular response to genotoxic agents, we aimed to analyze the role of Chk1 in GC as a biomarker for radiotherapy resistance. We analyzed Chk1 expression in AGS and MKN45 human GC cell lines by RT-QPCR and WB and in a small cohort of human patient’s samples. We demonstrated that Chk1 overexpression specifically increases resistance to radiation in GC cells. Accordingly, abrogation of Chk1 activity with UCN-01 and its expression with shChk1 increased sensitivity to bleomycin and radiation. Furthermore, when we assessed Chk1 expression in human samples, we found a correlation between nuclear Chk1 accumulation and a decrease in progression free survival. Moreover, using a luciferase assay we found that Chk1’s expression is controlled by p53 and RB/E2F1 at the transcriptional level. Additionally, we present preliminary data suggesting a posttranscriptional regulation mechanism, involving miR-195 and miR-503, which are inversely correlated with expression of Chk1 in radioresistant cells. The renin-angiotensin system: new insight into old therapies.
Although the renin-angiotensin system (RAS) is already an old acquaintance, there are often exciting discoveries that improve our knowledge of it and open new therapeutic possibilities. Moreover, well-established drugs, such as angiotensin-converting enzyme inhibitors (ACEI), angiotensin receptor blockers (ARB), or beta-blockers, show that their mechanism of action may be the result of parallel pathways other than the ones initially established. A detailed analysis of the RAS can be carried out in part through the study of the enzymes, named angiotensinases, involved in its cascade, whose activity is a reflection of the functionality of their peptide substrates. The study of these enzymes offers the possibility of controlling the effects of angiotensins through various pharmacological manipulations. For example, angiotensinase inhibitors or activators are being used or have been proposed as antihypertensive agents. They have also been suggested as analgesic and antidepressant drugs or targets for drug development against different pathologies such as Alzheimer's disease, epilepsy or ischemia. On the other hand, the analysis of brain asymmetry has revealed surprising results about the laterality of central and peripheral components of the RAS. Such studies indicate that the neurovisceral integration, already proposed by Claude Bernard (1867) should also be analyzed from a bilateral perspective. In this review, the RAS and the role of various angiotensinases implicated in the cascade are revisited. Therapeutic strategies involving some components of the RAS with an unusual vision resulting from a bilateral perspective added to their study are discussed.SAF-2008 04685 C0
Potentiation of radiation therapy by the oncolytic adenovirus dl1520 (ONYX-015) in human malignant glioma xenografts
In spite of aggressive surgery, irradiation and/or chemotherapy, treatment of malignant gliomas remains a major challenge in adults and children due to high treatment failure. We have demonstrated significant cell lysis and antitumour activity of the E1B-55 kDa-gene-deleted adenovirus ONYX-015 (dl1520, CI-1042; ONYX Pharmaceuticals) in subcutaneous human malignant glioma xenografts deriving from primary tumours. Here, we show the combined efficacy of this oncolytic therapy with radiation therapy. Total body irradiation (5 Gy) of athymic nude mice prior to intratumoral injections of ONYX-015 1 x 10(8) PFU daily for 5 consecutive days yielded additive tumour growth delays in the p53 mutant xenograft IGRG88. Radiation therapy was potentiated in the p53 functional tumour IGRG121 with a 'subtherapeutic' dose of 1 x 10(7) PFU daily for 5 consecutive days, inducing significant tumour growth delay, 90% tumour regression and 50% tumour-free survivors 4 months after treatment. These potentiating effects were not due to increased adenoviral infectivity or replication. Furthermore, cell lysis and induction of apoptosis, the major mechanisms for adenoviral antitumour activity, did not play a major role in the combined treatment strategy. Interestingly, the oncolytic adenovirus seemed to accelerate radiation-induced tumour fibrosis. Potentiating antitumour activity suggests the development of this combined treatment for these highly malignant tumours
Brain, Heart and Kidney Correlate for the Control of Blood Pressure and Water Balance: Role of Angiotensinases.
The renin-angiotensin system (RAS) plays a major role in the control of blood pressure (BP) and water balance by coordinating brain, heart and kidney functions, connected with each other by hormonal and neural mechanisms through the autonomic nervous system (ANS). RAS function may be monitored by the study of the enzymes (angiotensinases) involved in the metabolism of its active peptides. In order to study the relationship between the brain-heart-kidney axis and the control of BP and water balance, we analyzed the correlation of angiotensinase activities, assayed as arylamidase activities, between hypothalamus, left ventricle, renal cortex and renal medulla, collected from Wistar-Kyoto and spontaneously hypertensive rats, treated or not treated with L-NAME [N(G)-nitro-L-arginine methyl ester]. This compound not only inhibits the formation of nitric oxide but also disrupts the normal function of the ANS activating the sympathetic nervous system (SNS) to increase BP. In addition, to assess the influence of the SNS, we studied the effect of its blockade by treatment of both strains with propranolol. The present results support the notion that RAS function of the brain-heart-kidney axis, as reflected by the activities of angiotensinases, is reciprocally connected by afferent and efferent mechanisms between these locations, presumably through the ANS. High contrast optical imaging of companions: the case of the brown dwarf binary HD-130948BC
High contrast imaging at optical wavelengths is limited by the modest
correction of conventional near-IR optimized AO systems.We take advantage of
new fast and low-readout-noise detectors to explore the potential of fast
imaging coupled to post-processing techniques to detect faint companions to
stars at small separations. We have focused on I-band direct imaging of the
previously detected brown dwarf binary HD130948BC,attempting to spatially
resolve the L2+L2 benchmark system. We used the Lucky-Imaging instrument
FastCam at the 2.5-m Nordic Telescope to obtain quasi diffraction-limited
images of HD130948 with ~0.1" resolution.In order to improve the detectability
of the faint binary in the vicinity of a bright (I=5.19 \pm 0.03) solar-type
star,we implemented a post-processing technique based on wavelet transform
filtering of the image which allows us to strongly enhance the presence of
point-like sources in regions where the primary halo dominates. We detect for
the first time the BD binary HD130948BC in the optical band I with a SNR~9 at
2.561"\pm 0.007" (46.5 AU) from HD130948A and confirm in two independent
dataset that the object is real,as opposed to time-varying residual speckles.We
do not resolve the binary, which can be explained by astrometric results
posterior to our observations that predict a separation below the NOT
resolution.We reach at this distance a contrast of dI = 11.30 \pm 0.11, and
estimate a combined magnitude for this binary to I = 16.49 \pm 0.11 and a I-J
colour 3.29 \pm 0.13. At 1", we reach a detectability 10.5 mag fainter than the
primary after image post-processing. We obtain on-sky validation of a technique
based on speckle imaging and wavelet-transform processing,which improves the
high contrast capabilities of speckle imaging.The I-J colour measured for the
BD companion is slightly bluer, but still consistent with what typically found
for L2 dwarfs(~3.4-3.6).Comment: accepted in A\&
Cold exposure modulates potential brown adipokines in humans, but only FGF21 is associated with brown adipose tissue volume
Objective: The study objective was to investigate the effect of cold exposure on the plasma levels of five potential human brown adipokines (chemokine ligand 14 [CXCL14], growth differentiation factor 15 [GDF15], fibroblast growth factor 21 [FGF21], interleukin 6 [IL6], and bone morphogenic protein 8b [BMP8b]) and to study whether such cold-induced effects are related to brown adipose tissue (BAT) volume, activity, or radiodensity in young humans.Methods: Plasma levels of brown adipokines were measured before and 1 h and 2 h after starting an individualized cold exposure in 30 young adults (60% women, 21.9 +/- 2.3 y; 24.9 +/- 5.1 kg/m(2)). BAT volume, F-18-fluorodeoxyglucose uptake, and radiodensity were assessed by a static positron emission tomography-computerized tomography scan after cold exposure.Results: Cold exposure increased the concentration of CXCL14 (Delta 2h = 0.58 +/- 0.98 ng/mL; p = 0.007), GDF15 (Delta 2h = 19.63 +/- 46.2 pg/mL; p = 0.013), FGF21 (Delta 2h = 33.72 +/- 55.13 pg/mL; p = 0.003), and IL6 (Delta 1h = 1.98 +/- 3.56 pg/mL; p = 0.048) and reduced BMP8b (Delta 2h = -37.12 +/- 83.53 pg/mL; p = 0.022). The cold-induced increase in plasma FGF21 was positively associated with BAT volume (Delta 2h: beta = 0.456; R-2 = 0.307; p = 0.001), but not with F-18-fluorodeoxyglucose uptake or radiodensity. None of the changes in the other studied brown adipokines was related to BAT volume, activity, or radiodensity.Conclusions: Cold exposure modulates plasma levels of several potential brown adipokines in humans, whereas only cold-induced changes in FGF21 levels are associated with BAT volume.
