    High aspect reactor vessel and method of use

    An improved bio-reactor vessel and system useful for carrying out mammalian cell growth in suspension in a culture media are presented. The main goal of the invention is to grow and maintain cells under a homogeneous distribution under acceptable biochemical environment of gas partial pressures and nutrient levels without introducing direct agitation mechanisms or associated disruptive mechanical forces. The culture chamber rotates to maintain an even distribution of cells in suspension and minimizes the length of a gas diffusion path. The culture chamber design is presented and discussed

    Regulatory Fit and Consumer Brand Preferences

    Research has demonstrated that consumer perceptions of products are affected by the fit between their regulatory focus or goal orientation and their conception of what products can offer in terms of satisfying the goals activated by that orientation. This research has focused on product features and the way product messages are framed for consumers. However, research has not focused on fit in terms of brand names and the types of regulatory orientations (promotion vs. prevention) that can be associated with them. This issue has potential implications for consumers and how products can be more effectively marketed to them. Given that research has demonstrated the consumers make product choices based on fit, the following was hypothesized in this study: H1: The more promotion-focused one is, the more positive product ratings will be if the brand name is associated with promotion concerns and strategies and H2: The more prevention-focused one is, the more positive product ratings will be if the brand name is associated with prevention concerns and strategies. While not the primary foci of this paper, effects based on the following two hypotheses were also examined: H3: As independent self-construal scores increase, the more likely it is that product ratings will be higher for brand names associated with promotion concerns and strategies and H4: As interdependent self-construal scores increase, the more likely it is that product ratings will be higher for brand names associated with prevention concerns and strategies. To test the hypotheses, participants were given individual difference measures, then asked to rate cars and sports teams with brand names created for the study. Sports teams were included given the expectation that consumers identify strongly with them (in addition to car brands). Brand names appeared in two types: promotion and prevention.     Cooperative Effects of Corticosteroids and Catecholamines upon Immune Deviation of the Type-1/Type-2 Cytokine Balance in Favor of Type-2 Expression in Human Peripheral Blood Mononuclear Cells

    A growing number of studies show strong associations between stress and altered immune function. In vivo studies of chronic and acute stress have demonstrated that cognitive stressors are strongly correlated with high levels of catecholamines (CT) and corticosteroids (CS). Although both CS and CT individually can inhibit the production of T-helper 1 (TH1, type-1 like) cytokines and simultaneously promote the production of T-helper 2 (TH2, type-2 like) cytokines in antigen-specific and mitogen stimulated human leukocyte cultures in vitro, little attention has been focused on the effects of combination CT and CS in immune responses that may be more physiologically relevant. We therefore investigated the combined effects of in vitro CT and CS upon the type-1/type-2 cytokine balance of human peripheral blood mononuclear cells (PBMC) as a model to study the immunomodulatory effects of superimposed acute and chronic stress. Results demonstrated a significant decrease in type-1 cytokine production (IFN-gamma) and a significant increase in type-2 cytokine production (IL-4, IL-10) in our CS+CT incubated cultures when compared to either CT or CS agents alone. Furthermore, variable enhancement of type-1/type-2 immune deviation occurred depending upon when the CT was added. The data suggest that CS can increase the sensitivity of PBMC to the immunomodulatory effects of CT and establishes an in vitro model to study the combined effects of in vivo type-1/type-2 cytokine alterations observed in acute and chronic stress

    The Importance of Data Visualization: Incorporating Storytelling into the Scientific Presentation

    From its inception in 2000, one of the primary tasks of the Biomedical Data Reduction Analysis (BDRA) group has been translation of large amounts of data into information that is relevant to the audience receiving it. BDRA helps translate data into an integrated model that supports both operational and research activities. This data integrated model and subsequent visual data presentations have contributed to BDRA's success in delivering the message (i.e., the story) that its customers have needed to communicate. This success has led to additional collaborations among groups that had previously not felt they had much in common until they worked together to develop solutions in an integrated fashion. As more emphasis is placed on working with "big data" and on showing how NASA's efforts contribute to the greater good of the American people and of the world, it becomes imperative to visualize the story of our data to communicate the greater message we need to share. METHODS To create and expand its data integrated model, BDRA has incorporated data from many different collaborating partner labs and other sources. Data are compiled from the repositories of the Lifetime Surveillance of Astronaut Health and the Life Sciences Data Archive, and from the individual laboratories at Johnson Space Center that support collection of data from medical testing, environmental monitoring, and countermeasures, as designated in the Medical Requirements Integration Documents. Ongoing communication with the participating collaborators is maintained to ensure that the message and story of the data are retained as data are translated into information and visual data presentations are delivered in different venues and to different audiences. RESULTS We will describe the importance of storytelling through an integrated model and of subsequent data visualizations in today's scientific presentations and discuss the collaborative methods used. We will illustrate the discussion with examples of graphs from BDRA's past work supporting operations and/or research efforts

    Biochemical and Structural Effects of Rigor Mortis-Accelerating Treatments in Broiler \u3ci\u3ePectoralis\u3c/i\u3e

    This study was conducted to elucidate the mechanism of action of two selected rigor mortis-accelerating treatment systems employed in the prevention of the toughness associated with early-harvested (1 h post-mortem) broiler Pectoralis muscle. The treatments included 14 min of low voltage electrical stimulation (110 V, 1 A, pulsing 1 s on and 1 s off) combined with high temperature conditioning (39 C) and muscle tensioning (LV + HTC + MT); a 15-s high voltage stimulation (440 V, 1 A pulsing 2 s on and 1 s off) combined with muscle tensioning (HV + MT); and a control simulating commercial broiler processing practices. The rigor-accelerating treatments reduced pH and increased R-value (inosine:adenosine ratio) at 1 h post-mortem, but only the LV + HTC + MT treatment reduced sarcomere shortening. Both rigor treatments reduced the amount of measurable myofibrillar fragmentation. Cathepsin B and B + L activities were not affected by the rigor treatments. Calpain I activity was not detectable in any 24-h post-mortem sample. Calpain II activity at 24 h post-mortem was greater in muscles receiving HV + MT than from the LV + HTC + MT or control carcasses, but was reduced in all muscles by 24 h postmortem. An SDS-PAGE indicated a 30-kDa polypeptide that was absent at death and appeared in control and LV + HTC + MT muscles but to a lesser extent in HV + MT muscles. These results suggested that the LV + HTC + MT treatment has a greater tenderizing effect than the HV + MT treatment because the former achieves a better balance between reduced sarcomere shortening and myofibrillar fragmentation
