    Optical Identification of Cepheids in 19 Host Galaxies of Type Ia Supernovae and NGC 4258 with the Hubble Space Telescope

    We present results of an optical search for Cepheid variable stars using the Hubble Space Telescope (HST) in 19 hosts of Type Ia supernovae (SNe Ia) and the maser-host galaxy NGC 4258, conducted as part of the SH0ES project (Supernovae and H0 for the Equation of State of dark energy). The targets include 9 newly imaged SN Ia hosts using a novel strategy based on a long-pass filter that minimizes the number of HST orbits required to detect and accurately determine Cepheid properties. We carried out a homogeneous reduction and analysis of all observations, including new universal variability searches in all SN Ia hosts, that yielded a total of 2200 variables with well-defined selection criteria -- the largest such sample identified outside the Local Group.     A mixed-method investigation of patient monitoring and enhanced feedback in routine practice: Barriers and facilitators

    Objective: To investigate the barriers and facilitators of an effective implementation of an outcome monitoring and feedback system in a UK National Health Service psychological therapy service. Method: An outcome monitoring system was introduced in two services. Enhanced feedback was given to therapists after session 4. Qualitative and quantitative methods were used, including questionnaires for therapists and patients. Thematic analysis was carried out on written and verbal feedback from therapists. Analysis of patient outcomes for 202 episodes of therapy was compared with benchmark data of 136 episodes of therapy for which feedback was not given to therapists. Results: Themes influencing the feasibility and acceptability of the feedback system were the extent to which therapists integrated the measures and feedback into the therapy, availability of administrative support, information technology, and complexity of the service. There were low levels of therapist actions resulting from the feedback, including discussing the feedback in supervision and with patients. Conclusions: The findings support the feasibility and acceptability of setting up a routine system in a complex service, but a number of challenges and barriers have to be overcome and therapist differences are apparent. More research on implementation and effectiveness is needed in diverse clinical settings

    Impact of changing climate on bryophyte contributions to terrestrial water, carbon, and nitrogen cycles

    Bryophytes, including the lineages of mosses, liverworts, and hornworts, are the second-largest photoautotroph group on Earth. Recent work across terrestrial ecosystems has highlighted how bryophytes retain and control water, fix substantial amounts of carbon (C), and contribute to nitrogen (N) cycles in forests (boreal, temperate, and tropical), tundra, peatlands, grasslands, and deserts. Understanding how changing climate affects bryophyte contributions to global cycles in different ecosystems is of primary importance. However, because of their small physical size, bryophytes have been largely ignored in research on water, C, and N cycles at global scales. Here, we review the literature on how bryophytes influence global biogeochemical cycles, and we highlight that while some aspects of global change represent critical tipping points for survival, bryophytes may also buffer many ecosystems from change due to their capacity for water, C, and N uptake and storage. However, as the thresholds of resistance of bryophytes to temperature and precipitation regime changes are mostly unknown, it is challenging to predict how long this buffering capacity will remain functional. Furthermore, as ecosystems shift their global distribution in response to changing climate, the size of different bryophyte-influenced biomes will change, resulting in shifts in the magnitude of bryophyte impacts on global ecosystem functions

    Development of the Low Frequency Telescope Focal Plane Detector Modules for LiteBIRD

    LiteBIRD is a JAXA-led strategic large-class satellite mission designed to measure the polarization of the cosmic microwave background and Galactic foregrounds from 34 to 448 GHz across the entire sky from L2 in the late 2020s. The scientific payload includes three telescopes which are called the low-, mid-, and high-frequency telescopes each with their own receiver that covers a portion of the mission's frequency range. The low frequency telescope will map synchrotron radiation from the Galactic foreground and the cosmic microwave background. We discuss the design, fabrication, and characterization of the low-frequency focal plane modules for low-frequency telescope, which has a total bandwidth ranging from 34 to 161 GHz. There will be a total of 4 different pixel types with 8 overlapping bands to cover the full frequency range. These modules are housed in a single low-frequency focal plane unit which provides thermal isolation, mechanical support, and radiative baffling for the detectors. The module design implements multi-chroic lenslet-coupled sinuous antenna arrays coupled to transition edge sensor bolometers read out with frequency-domain mulitplexing. While this technology has strong heritage in ground-based cosmic microwave background experiments, the broad frequency coverage, low optical loading conditions, and the high cosmic ray background of the space environment require further development of this technology to be suitable for LiteBIRD.     dATF4 regulation of mitochondrial folate-mediated one-carbon metabolism is neuroprotective.

    Neurons rely on mitochondria as their preferred source of energy. Mutations in PINK1 and PARKIN cause neuronal death in early-onset Parkinson's disease (PD), thought to be due to mitochondrial dysfunction. In Drosophila pink1 and parkin mutants, mitochondrial defects lead to the compensatory upregulation of the mitochondrial one-carbon cycle metabolism genes by an unknown mechanism. Here we uncover that this branch is triggered by the activating transcription factor 4 (ATF4). We show that ATF4 regulates the expression of one-carbon metabolism genes SHMT2 and NMDMC as a protective response to mitochondrial toxicity. Suppressing Shmt2 or Nmdmc caused motor impairment and mitochondrial defects in flies. Epistatic analyses showed that suppressing the upregulation of Shmt2 or Nmdmc deteriorates the phenotype of pink1 or parkin mutants. Conversely, the genetic enhancement of these one-carbon metabolism genes in pink1 or parkin mutants was neuroprotective. We conclude that mitochondrial dysfunction caused by mutations in the Pink1/Parkin pathway engages ATF4-dependent activation of one-carbon metabolism as a protective response. Our findings show a central contribution of ATF4 signalling to PD that may represent a new therapeutic strategy. A video abstract for this article is available at https://youtu.be/cFJJm2YZKKM

    Evaluation of the recombinant proteins RlpB and VacJ as a vaccine for protection against Glaesserella parasuis in pigs

Prevention of Glӓsser’s disease in pigs has been plagued with an inability to design broadly protective vaccines, as many bacterin based platforms generate serovar or strain specific immunity. Subunit vaccines are of interest to provide protective immunity to multiple strains of G. parasuis. Selected proteins for subunit vaccination should be widespread, highly conserved, and surface exposed. Results: Two candidate proteins for subunit vaccination (RlpB and VacJ) against G. parasuis were identified using random mutagenesis and an in vitro organ culture system. Pigs were vaccinated with recombinant RlpB and VacJ, outer membrane proteins with important contributions to cellular function and viability. Though high antibody titers to the recombinant proteins and increased interferon-γ producing cells were found in subunit vaccinated animals, the pigs were not protected from developing systemic disease. Conclusions: It appears there may be insufficient RlpB and VacJ exposed on the bacterial surface for antibody to bind, preventing high RlpB and VacJ specific antibody titers from protecting animals from G. parasuis. Additionally, this work confirms the importance of utilizing the natural host species when assessing the efficacy of vaccine candidates
