    Escala de perceção dos alunos sobre o feedback dos professores: construção e validação

    O presente estudo procura testar as propriedades psicométricas de um questionário que avalia (a) perceção do aluno sobre o feedback do professor; a identificação escolar do aluno; as trajetórias escolares (factos e expectativas) e; a perceção do aluno sobre o seu envolvimento escolar. O questionário foi aplicado a 1089 alunos dos 6º, 7º, 9º e 10º anos de escolaridade (M=13.4, DP=1.7), sendo que 52% são do sexo feminino. A amostra é composta por alunos essencialmente de nacionalidade portuguesa (95.9%). A partir dos resultados da análise factorial e seguindo o racional teórico, chegou-se a uma estrutura composta por oito dimensões principais. O QFITE apresenta bons índices de consistência interna, com sete das oito principais dimensões a obterem valores entre .77 e .89. Assim, as análises psicométricas realizadas revelam valores satisfatórios, concluindo-se que o QFITE é um instrumento útil e adequado para avaliar a identificação escolar dos alunos, o envolvimento comportamental escolar, e as perceções dos alunos sobre o feedback do professor

    Scale of student perceptions regarding their Behavioral Engagement in school: construction and validation

    Este estudo descreve a construção e validação de uma escala que avalia a perceção dos alunos sobre o seu envolvimento comportamental escolar. Os questionários foram aplicados a 1089 alunos do 6 º, 7 º, 9 º e 10º anos de escolaridade (idade mediana = 13) em Portugal Continental, a partir dos quais foram extraídas duas subamostras aleatórias. Uma subamostra foi submetida a análise fatorial exploratória e a segunda, a uma análise fatorial confirmatória. Emergiram duas dimensões: envolvimento no trabalho acadêmico e participação na aula. A primeira escala revelou uma consistência interna aceitável, revelando-se útil e adequada para avaliar as percepções dos alunos sobre o seu envolvimento comportamental escolar.     Engineering the Controlled Assembly of Filamentous Injectisomes in E. coli K-12 for Protein Translocation into Mammalian Cells.

    Bacterial pathogens containing type III protein secretion systems (T3SS) assemble large needle-like protein complexes in the bacterial envelope, called injectisomes, for translocation of protein effectors into host cells. The application of these molecular syringes for the injection of proteins into mammalian cells is hindered by their structural and genomic complexity, requiring multiple polypeptides encoded along with effectors in various transcriptional units (TUs) with intricate regulation. In this work, we have rationally designed the controlled expression of the filamentous injectisomes found in enteropathogenic Escherichia coli (EPEC) in the nonpathogenic strain E. coli K-12. All structural components of EPEC injectisomes, encoded in a genomic island called the locus of enterocyte effacement (LEE), were engineered in five TUs (eLEEs) excluding effectors, promoters and transcriptional regulators. These eLEEs were placed under the control of the IPTG-inducible promoter Ptac and integrated into specific chromosomal sites of E. coli K-12 using a marker-less strategy. The resulting strain, named synthetic injector E. coli (SIEC), assembles filamentous injectisomes similar to those in EPEC. SIEC injectisomes form pores in the host plasma membrane and are able to translocate T3-substrate proteins (e.g., translocated intimin receptor, Tir) into the cytoplasm of HeLa cells reproducing the phenotypes of intimate attachment and polymerization of actin-pedestals elicited by EPEC bacteria. Hence, SIEC strain allows the controlled expression of functional filamentous injectisomes for efficient translocation of proteins with T3S-signals into mammalian cells

    Effect of different restoration techniques and cavity designs on cuspal deflection of posterior teeth restored with resin composite inlays

    Objectives: The aim of this study was to evaluate the influence of different restoration techniques (immediate dentin sealing (IDS) restoration techniques) and cavity designs on the cuspal deflection of posterior teeth that were restored with resin composite inlays. Methods: Sixty caries-free extracted maxillary premolars were selected and equally divided into two experimental groups. Group D1: MOD cavities were prepared. Group D2: MOD cavity with gingival steps. Each group was subdivided to three subgroups (n=10) and restored as follows: inlay without IDS; inlay with IDS and adhesive system; inlay with IDS and adhesive system and low-viscosity resin. Cuspal deflection was measured with a micrometer. One-Way Analysis of Variance ANOVA was used to analyze the results. Results: Cavity design D1 showed the lowest cuspal deflection compared to cavity design D2. In group D1; Inaly without IDS showed the highest significant cuspal deflection (9.85 μm) followed by Inaly+IDS+Low viscosity resin (7.16 μm). The lowest value was obtained for Inaly + IDS group (4.76 μm) with significant difference between all tested restoration techniques. In group D2; Inaly without IDS showed the highest significant cuspal deflection (14.7 μm) followed by Inaly + IDS + Low viscosity resin (11.69 μm). The lowest value was obtained for Inaly + IDS (9.59 μm) with a significant difference between all tested restoration techniques. Conclusion: IDS and Protect Liner F allowed less cuspal deflection comparable with traditional technique. However, IDS restoration techniques did not decrease the cuspal deflection in case of more extensive loss of dental structure in the premolar teeth

    Immunotoxic effect of thiamethoxam in immunized mice with Brucella abortus cultural filtrate antigen

    Aim: This study was planned for determination the toxic effect of thiamethoxam (TMX) in immunized mice with Brucella abortus culture filtrate antigen (CFBAgs) (as a vaccine) and its role of TMX on decrease activity of B. abortus antigen on eliciting of humoral and cellular immunity. Materials and Methods: To achieve these goals 60 female mice were used, 7-8 weeks age, they were divided equally into three groups (20 in each group) and treated as follows: 1st group: Mice were immunized with CFBAgs intraperitoneally in two doses, 2 weeks intervals with (protein concentration 2 mg\ml), 2nd group: Mice immunized as in the 1st group and was administrated orally with 1/10 lethal dose 50% of TMX (83.7 mg/kg B.W.) for 4 weeks daily, 3rd group was administrated orally with 0.3 ml normal saline served as a control group. At day 28 post immunization (PI) delayed type hypersensitivity (skin test) was done, and serum samples were collected at day 30 (PI) for detection of passive hemagglutination test (PHA); interferon gamma (IFN-γ) which was done by enzyme-linked immunosorbent assay test in addition to phagocytes assay. Results: The results of skin test post injection with soluble antigen of B. abortus intradermally showed a high significantly mean values at p≤0.05 of footpad skin thickness in the 1st group of mice which recorded (0.51±0.002 mm) as compared with the 2nd group of mice which showed (0.08±0.002 mm) after 24 h; the mean values of skin thickness were declined in the 1st mice (0.46±0.002) and 2nd mice (0.070±0.001) at 48 h; control group showed a negative results. These results were agreed with results of serum levels of IFN-γ (pg/ml) that showed that a significant increase the vaccinated 1st group (406.36±1.52), than those values in the 2nd group (151.61±0.89) and negative result in 3rd group (46.47±0.60), in addition to results of PHA test which showed a significant increase in antibody titer in the 1st group (139±12.16) with low level of serum antibody in the 2nd group (7.66±0.33). Phagocytic ratio results in the 1st group showed an increase to reach (18.55±0.44) than a ratio in the 2nd group (13.24±0.32) and the control group (5.46±0.25). Conclusion: It was concluded that TMX induced suppression of humoral and cellular immune responses in immunized mice with CFBAgs
