    Direct and indirect air particle cytotoxicity in human alveolar epithelial cells

    Air particulate matter has been associated with adverse impact on the respiratory system leading to cytotoxic and proinflammatory effects. The biological mechanisms behind these associations may be initiated by inhaled small size particles, particle components (soluble fraction) and/or mediators released by particle-exposed cells (conditioned media). The effect of Urban Air Particles from Buenos Aires (UAP-BA) and Residual Oil Fly Ash (ROFA) a surrogate of ambient air pollution, their soluble fractions (SF) and conditioned media (CM) on A549 lung epithelial cells was examined. After 24h exposure to TP (10 and 100 μg/ml), SF or CM, several biological parameters were assayed on cultured A549 cells. We tested cell viability by MTT, superoxide anion (O2-) generation by NBT and proinflammatory cytokine (TNF α, IL-6 and IL-8) production by ELISA. UAP-BA particles or its SF (direct effect) did not modify cell viability and generation of O2- for any of the doses tested. On the contrary, UAP-BA CM (indirect effect) reduced cell viability and increased both generation of O2- and IL-8 production. Exposure to ROFA particles, SF or ROFA CM reduced proliferation and O2- but, stimulated IL-8. It is worth to note that UAP-BA and ROFA depicted distinct effects on particle-exposed A549 cells implicating morphochemical dependence.     Heterogeneous impacts of mobility restrictions on air quality in the State of Sao Paulo during the COVID-19 pandemic

    Air quality in the State of Sao Paulo was evaluated during the first general State plan of mobility restrictions due to the COVID-19 pandemic (24th March to May 31, 2020). Nitrogen dioxide (NO2), ozone (O-3), particulate matter PM10 and PM2.5 and sulphur dioxide (SO2) concentrations were assessed in cities of the Sao Paulo State with a monitoring station and compared to historical data. Linear regression models were built to investigate the relationship between the isolation of the population - determined using mobile phone monitoring data - and the concentration of each pollutant during the studied period. Although the reduction of pollutants such as NO2, SO2 and PM2.5 is very clear, the economic and climatic characteristics of each region were decisive in the general behaviour of O-3 and PM10. It was not possible to establish a correlation between the pollutants and the isolation index, partly due to the lack of data, partly due to the compliance of the population to those measurements, which was variable over time. Another important limitation factor was the absence of data related to the pollutants of interest in many of the stations. However, the isolation measures carried out in the State opened the opportunity to individually assess the air quality measurements in each of the stations, enabling an understanding that will allow in the future the design of air quality policies together with local sanitary policies

    The impact of sugar cane-burning emissions on the respiratory system of children and the elderly

    We analyzed the influence of emissions front burning sugar cane on the respiratory system during almost I year in the city of Piracicaba in southeast Brazil. From April 1997 through March 1998, samples of inhalable particles were collected, separated into fine and coarse particulate mode, and analyzed for black carbon and tracer elements. At the same time, we examined daily records of children ( 64 years of age) admitted to the hospital because of respiratory diseases. Generalized linear models were adopted with natural cubic splines to control for season and linear terms to control for weather. Analyses were carried out for the entire period, as well as for burning and rionburning periods. Additional models were built using three factors obtained from factor analysis instead of particles or tracer elements. Increases of 10.2 mu g/m(3) in particles <= 2.5 mu m/m(3) aerodynamic diameter (PM2.5) and 42.9 mu g/m(3) in PM10 were associated with increases of 21.4% [95% confidence interval (0), 4.3-38.5] and 31.03% (95% Cl, 1.25-60.21) in child and elderly respiratory hospital admissions, respectively. When we compared periods, the effects during the burning period were much higher than the effects during nonburning period. Elements generated from sugar cane burning (factor 1) were those most associated with both child and elderly respiratory admissions.     Micronucleus formation induced by biomass burning particles derived from biomass burning induce high micronucleus frequency in Tradescantia pallida assay (TRAD-MN)

    Manual harvesting is usually done after to sugar cane burning which is responsible for seasonal emission of air pollutants in Brazil and it is believed to be responsible for deleterious health effects in exposed populations. The mutagenic potential of sugar cane burning harvesting particulate and particle surrogates of residual oil fly ash (ROFA) were evaluated in assays measuring micronuclei (MN) in the pollen mother cells of Tradescantia pallida (TRAD-MN). Micronuclei frequencies in TRAD-MN to sugar cane burning residues (SCBR) at doses 0.3 and 0.03 mg/mL were respectively 2.18 ± 0.35 and 5.53 ± 1.04, whereas to ROFA from incinerator and ROFA from an electrostatic precipitator installed in one of the chimneys of a steel plant, MN frequencies were, respectively, 3.43 ± 0.7 and 4.90 ± 1.07. Significant differences were detected among the groups (p < 0.001), demonstrating that SCBR was at least as genotoxic as the fossil fuel derived particles. The results suggest that the burning process to harvest sugar cane should be better controlled. Keywords: pollution, burning particles, micronuclei, Tradescantia pallida . Partículas derivadas da queima de biomassa induzem alta frequência de micronúcleos no ensaio de Tradescantia pallida (TRAD-MN) Resumo A coleta de cana-de-açúcar manual é geralmente realizada após a queima, a qual é responsável pela emissão sazonal de poluentes atmosféricos no Brasil e acredita-se que este seja responsável pelos efeitos deletérios a saúde da população exposta. O potencial mutagênico da queima de cana-de-açúcar e partículas oriundas de resíduos em cinza de óleo queimado (ROFA) foi avaliado em ensaio que mede a freqüência de micronúcleos (MN) em células mãe de grão de pólen de Tradescantia pallida (TRAD-MN). A freqüência de TRAD-MN em resíduos de queima de cana de açúcar (SCBR) nas doses de 0.3 e 0,03mg/mL foi, respectivamente de 2,18 ± 0,35 e 5,53 ± 1,04, enquanto para ROFA de incinerador e ROFA de um precipitador eletrostático instalado em uma chaminé de uma indústria siderúrgica, a freqüência de MN foi, respectivamente, 3,43 ± 0,7 e 4,90 ± 1,07. Diferença significante foi observada entre os grupos (p<0,001), demonstrando que SCBR foi ao menos tão genotóxico quanto às partículas derivadas de combustíveis fósseis. Os resultados sugerem que o processo de queima de cana-de-açúcar no período da safra deveria ser mais bem controlado.     The Tradescantia micronucleus test is a sensitive bioassay for mutagenesis that may be employed both under field and laboratory conditions. This test has been standardized mostly on the basis of the results obtained with clone 4430. However, this clone is not well adapted to tropical weather, frequently showing problems with growth and flowering. In addition, it is attacked by parasites and insects, a fact that limits its use in field studies aiming at the biomonitoring of air pollution. In the city of São Paulo, Tradescantia pallida (Rose) Hunt. var. purpurea Boom is widely distributed as an ornamental plant in gardens and along roadsides and streets, mostly because of its natural resistance and its easy propagation. In this report, we present dose-response curves indicating that the sensitivity of T. pallida and clone 4430 to X-radiation (1, 10, 25 and 50 cGy) is similar. The results confirm our previous suggestion that T. pallida represents a good alternative for in situ mutagenesis testing in tropical regions, especially biomonitoring studies in which the exposure conditions may not be fully controllable

    Vehicle emissions and PM2.5 mass concentrations in six Brazilian cities

    In Brazil, the principal source of air pollution is the combustion of fuels (ethanol, gasohol, and diesel). In this study, we quantify the contributions that vehicle emissions make to the urban fine particulate matter (PM2.5) mass in six state capitals in Brazil, collecting data for use in a larger project evaluating the impact of air pollution on human health. From winter 2007 to winter 2008, we collected 24-h PM2.5 samples, employing gravimetry to determine PM2.5 mass concentrations; reflectance to quantify black carbon concentrations; X-ray fluorescence to characterize elemental composition; and ion chromatography to determine the composition and concentrations of anions and cations. Mean PM2.5 concentrations in the cities of São Paulo, Rio de Janeiro, Belo Horizonte, Curitiba, Porto Alegre, and Recife were 28, 17.2, 14.7, 14.4, 13.4, and 7.3 μg/m3, respectively. In São Paulo and Rio de Janeiro, black carbon explained approximately 30% of the PM2.5 mass. We used receptor models to identify distinct source-related PM2.5 fractions and correlate those fractions with daily mortality rates. Using specific rotation factor analysis, we identified the following principal contributing factors: soil and crustal material; vehicle emissions and biomass burning (black carbon factor); and fuel oil combustion in industries (sulfur factor). In all six cities, vehicle emissions explained at least 40% of the PM2.5 mass. Elemental composition determination with receptor modeling proved an adequate strategy to identify air pollution sources and to evaluate their short- and long-term effects on human health.     Efeitos da poluição do ar nas doenças cardiovasculares: estruturas de defasagem

    OBJETIVO: Investigar a estrutura de defasagem entre exposição à poluição do ar e internações hospitalares por doenças cardiovasculares em idosos, separada por gênero. MÉTODOS: Os dados de saúde de pessoas com mais de 64 anos de idade foram estratificados por gênero, na cidade de São Paulo, entre 1996 e 2001. Os níveis diários de poluentes do ar (CO, PM10, O3, NO2, SO2) e os dados de temperatura mínima e umidade relativa do ar foram também foram analisados. Foram utilizados modelos restritos de distribuição polinomial em modelos aditivos generalizados de regressão de Poisson para estimar os efeitos dos poluentes no dia da exposição e até 20 dias após, controlando-se para sazonalidades de longa e curta durações, feriados e fatores meteorológicos. RESULTADOS: Variações interquartis de PM10 (26,21 mig/m³) e SO2 (10,73 mig/m³) foram associados com aumentos de 3,17% (IC 95%: 2,09-4,25) nas admissões por insuficiência cardíaca congestiva e de 0,89% (IC 95%: 0,18-1,61) para admissões por todas as doenças cardiovasculares no dia da exposição, respectivamente. Os efeitos foram predominantemente agudos e maiores para o gênero feminino. Além disso, foi observado efeito colheita. CONCLUSÕES: Os achados mostraram que as doenças cardiovasculares em São Paulo são fortemente afetadas pela poluição do ar.OBJECTIVE: To assess the lag structure between air pollution exposure and elderly cardiovascular diseases hospital admissions, by gender. METHODS: Health data of people aged 64 years or older was stratified by gender in São Paulo city, Southeastern Brazil, from 1996 to 2001. Daily levels of air pollutants (CO, PM10, O3, NO2, and SO2) , minimum temperature, and relative humidity were also analyzed. It were fitted generalized additive Poisson regressions and used constrained distributed lag models adjusted for long time trend, weekdays, weather and holidays to assess the lagged effects of air pollutants on hospital admissions up to 20 days after exposure. RESULTS: Interquartile range increases in PM10 (26.21 mug/m³) and SO2 (10.73 mug/m³) were associated with 3.17% (95% CI: 2.09-4.25) increase in congestive heart failure and 0.89% (95% CI: 0.18-1.61) increase in total cardiovascular diseases at lag 0, respectively. Effects were higher among female group for most of the analyzed outcomes. Effects of air pollutants for different outcomes and gender groups were predominately acute and some "harvesting" were found. CONLUSIONS: The results show that cardiovascular diseases in São Paulo are strongly affected by air pollution
