    TINDAK TUTUR DIREKTIF DALAM TRANSAKSI JUAL BELI DI PASAR SENTRAL WATAMPONEAbstractThe purpose of this research is to describe the directive act of transaction in central market of Watampone. This research is focused on directive speech act used by the speakers in the tansaction. This research uses qualitative research method. Techniques of collecting data are: 1) recording each respondent and 2) collecting additional data by interviewing. Validity of data was verified by using credibility, transferability, dependability, and confirmability. Data were analyzed based on two sources of information: 1) during data collection that is: data reduction stage, encodes, data selection, memo, make summaries and temporary hypotesis and 2) after collecting the data: such as conclusion results, either recording results, observation, or interview. Type of directive speech act found in the transaction at central market Watampone in this research are making question, order, advice, request, and recommendation.Keyword: direcitive speech act, transactionAbstrakPenelitian ini bertujuan untuk mendeskripsikan tindak tutur direktif pada transaksi jual beli di Pasar Sentral Watampone. Fokus penelitian pada tindak tutur direktif dalam transaksi jual beli. Penelitian ini menggunakan metode kualitatif. Teknik pengumpulan data meliputi: 1) melakukan perekaman kepada setiap responden dan 2) pengumpulan data tambahan dilakukan wawancara. Untuk menverifikasi keabsahan data dilakukan dengan kredibilitas, transferbilitas, dependabilitas, dan komfirmabilitas. Data dianalisis dengan mengacu kepada dua sumber informasi, yaitu 1) selama pengumpulan data berlangsung, yakni: tahap reduksi data, mengkode, memilih data, memo, membuat ringkasan, dan membuat kesimpulan sementara dan 2) setelah pengumpulan data meliputi: penarikan kesimpulan hasil temuan, baik berupa hasil rekaman, observasi, atau wawancara.     Penelitian ini dilakukan dalam rangka pengembangan prospek geowisata dan agrowisata dari potensi sumber daya alam dan lingkungan secara lebih terperinci, baik yang sudah digali tapi belum optimal maupun yang belum digali dan diusahakan akan ditemukan potensi baru untuk produk wisata. Penelitian ini berlokasi di kabupaten Konawe Selatan dengan pengembangan objek dan daya tarik wisata alam. Desain penelitian yang digunakan dalam penelitian ini adalah desain dengan metode survei. Jenis data yang digunakan dalam penelitian ini adalah data kualitatif dan data kuantitatif. Jenis data ini diambil dari sumber data primer dan data sekunder. Pengumpulan data penelitian ini, melalui tiga teknik. yakni wawancara, pengamatan dan dokumen. Analisis data dalam penelitian ini adalah deskriptif kualitatif. Hasil penelitian menunjukkan bahwa Kelompok geowisata yang merupakan bagian dari objek kepariwisataan yang memanfaatkan alam atau kebumian sebagai objek utama yang ditemukan adalah geowisata pantai Pulau Hari dan Keindahan Alam Bawah Air, Pantai Polewali, Torobulu, Lapuko dan Trobulu, pemandian boro-boro, bendungan Aopu, Laeya dan Pudohoa, air terjun Moramo, Lasidala, dan Sandei, gua Anggase dan Kumapo dan air panas Kaendi dan Amohoa. Kelompok Agrowisata yang memanfaatkan USAha pertanian (agro) sebagai objek utama. ditemukan adalah agrowisata kebun tebu dan Rawa Aopa. Kesimpulan penelitian ini adalah obyek wisata yang tersedia di Kabupaten Konawe Selatan sangatlah bervariasi, meliputi obyek wisata alam yang dikelompokkan menjadi geowisata dan agrowisata dan juga obyek wisata budaya. Obyek wisata yang berpotensi tinggi untuk pengembangan antara lain keindahan dan keunikan bawah laut Pulau Hari, air terjun Moramo dan Rawa Aopa, bahkan dapat dijadikan sebagai salah satu daerah tujuan wisata utama di Provinsi Sulawesi Tenggara

    Theory of a Continuous Hc2_{c2} Normal-to-Superconducting Transition

    I study the Hc2H_{c2} transition within the Ginzburg-Landau model, with mm-component order parameter ψi\psi_i. I find a renormalized fixed point free energy, exact in m→∞m\rightarrow\infty limit, suggestive of a 22nd-order transition in contrast to a general belief of a 11st-order transition. The thermal fluctuations for H≠0H\neq 0 force one to consider an infinite set of marginally relevant operators for d<duc=6d<d_{uc}=6. I find dlc=4d_{lc}=4, predicting that the ODLRO does not survive thermal fluctuations in d=2,3d=2,3. The result is a solution to a critical fixed point that was found to be inaccessible within ϵ=6−d\epsilon=6-d-expansion, previously considered in E.Brezin, D.R.Nelson, A.Thiaville, Phys.Rev.B {\bf 31}, 7124 (1985), and was interpreted as a 11st-order transition.Comment: 4 pages, self-unpacking uuencoded compressed postscript file with a figure already inside text; to appear in Phys.     Ultra-performance liquid chromatography–tandem mass spectrometric determination of ramipril in human plasma

    Purpose: To develop a sensitive and accurate ultra-performance liquid chromatography–tandem mass spectrometric (UPLC-MS) method for quantification of ramipril in human plasma.Methods: Ramipril was extracted from biological fluid using equal volumes of n-hexane and propanol (1:1, v/v), and then chromatographed in a suitable C18 column with methanol: 0.1 % HCOOH (4: 1, v/v) as mobile phase. Atorvastatin was used as an internal standard for the&nbsp; chromatographic separation and quantification. The method was validated according to the United States Food and Drug Administration guidelines for standard indices.Results: Ramipril was determined in the concentration range 0.05 and 1000 ng/mL the validation procedure exhibited a correlation coefficient of 0.9979 + 0.002 (p = 0.05). The studied drug was quantified with lower ceiling of 0.05 ng/mL, and showed an accuracy of 105.00 %.Conclusion: A sensitive UPLC-MS analytical method has been successfully developed for the quantification of ramipril in human plasma. This method can be applied efficiently for the quantification of ramipril in bioavailability and pharmacokinetic studies.     THE IMPLEMENTATION OF AUTHENTIC ASSESSMENT IN THE 2013 CURRICULUM AT MAN 2 MODEL MAKASSAR

    This research focuses on the implementation and the challenges of authentic assessment in 2013 Curriculum. Employing case study design, the study draws insights of the implementation embracing the types of assessment, the competences to assess, and the challenges encountered by two English teachers and two classes of the students from a senior high school in Makassar. The obtained data from six nonparticipant observations, two sessions of a semi-structured interview with teachers and documentation toward teachers’ documents were all analyzed using thematic analysis model in relations to the concerns of this study. he findings demonstrated that the teachers implemented the authentic assessment in three cyclical stages covering planning, implementing, and evaluating. Three assessment types covering observation, written test, and performance assessment were employed to assess three competencies consisting of attitude, knowledge, and skills. Unfortunately, this practice still remained challenges for teacher covering pertains to scoring, time and effort consuming, and validity issue. It, therefore, could be concluded that the challenges will adhere to the implementation of authentic assessment. These findings inform teachers, policy maker, and future researcher to take possible actions on what and how to do next in order to enhance the practice of assessment in Indonesia.     Influence of hybridization on tensile behaviors of non-absorbable braided polymeric sutures

    This paper aims to investigate the effects of fiber hybridization technique on the mechanical behaviors of non-absorbable braided composite sutures. Fifteen types of hybrid braided sutures (HBSs) made of polyester (PET), polypropylene (PP), and polyamide 6 (PA6) are produced and tested to measure ultimate tensile strength (UTS), maximum strain, elastic modulus, and breaking toughness. Based on the results, it is observed that the suture material plays a significant role in the tensile and mechanical performance of HBSs, and they can be tailored through the different combinations of yarns according to the required mechanical properties. Experiments exhibit occurrence positive hybrid effect in both maximum strain and elastic modulus, and negative hybrid effect in UTS. The optimal tensile performance is associated with the hybrid structure comprising 75% PA6-12.5% PET-12.5% PP. This means the ternary structure with higher PA6 content along with PP and PET, demonstrates a synergistic effect. Thus, such a ternary composite structure is very promising for the design of novel non-absorbable sutures. Due to the absence of similar results in the specialized literature, this paper is likely to advance the state-of-the-art composite non-absorbable sutures and contribute to a better understanding of the hybridization concept for optimizing composite material systems

    Evidence that human P2Y1 and P2Y12 receptors form heterodimers

    Introduction: P2Y1 and P2Y12 receptors belong to the class A family of transmembrane GPCRs that are activated by endogenous nucleotides1. There is growing evidence that many GPCRs, including P2Y receptors, can exist as dimers or higher-order oligomers2. For example, P2Y12 and PAR4 receptors were recently reported to dimerise3. Our previous studies indicated that hP2Y1 and hP2Y12 receptors may form a functional heterodimer with novel pharmacological and signalling properties4. The aim of this project was, therefore, to characterise the physical interaction between hP2Y1 and hP2Y12 receptors. Method:tSA201 cells were transfected or co-transfected with hP2Y1 and hP2Y12 receptors, tagged with HA or a fluorescent protein. Cellular localisation and co-localisation of the receptors were determined using confocal microscopy. Transfected cells were cultured in the absence or presence of the N-glycosylation inhibitor tunicamycin (2.0 μg/ml) for 16 hours to determine the role of N-glycosylation in receptors expression. Receptor cell surface expression was quantified using ELISA. To investigate physical interaction between the two P2Y subtypes, co-immunoprecipitation was performed using anti-HA-agarose beads followed by immunoblotting with anti-GFP, anti-HA then alpha-Tubulin antibodies. Result: Following transfection on their own or together, both receptors were localised mainly at the cell membrane, and this was unaffected by tunicamycin. Co-immunoprecipitation confirmed that P2Y1 and P2Y12 receptors associate physically. Each subtype enhanced the other's surface expressions. In particular, expression of the P2Y12 receptor more than doubled that of P2Y1 receptors at the cell surface. Conclusion: These results show that P2Y1 and P2Y12 receptors are physically associated at the cell membrane and that they enhance each other's cell surface expressions.     Acaricidal efficiency of solar 50 % new emulsifiable concentrate formulation against the two-spotted spider mite (TSSM) Tetranychus urticae Koch (Acari: Tetranychidae) under laboratory and greenhouse conditions

    Oils are some of the most efficient and secure alternatives to synthetic fungicides, acaricides and insecticides used as pesticides for decades. Around the world, mineral oils are a potential pesticide against many pests. To provide novel active ingredients and new pesticide formulations to the pesticide industry, the major goal of this research was to formulate one of the petroleum fractions and test its acaricidal efficiency against two-spotted spider mite (TSSM), Tetranychus urticae Koch (Acari: Tetranychidae). Solar's physical features were put to the test. Then, it was prepared as an emulsifiable concentrate following the guidelines provided by specialized pesticide organizations for this kind of formulation. The novel formulation was subsequently biologically tested against T. urticae adults in the lab, and it demonstrated good acaricidal activity with an LC50 of 4548 ppm. Under greenhouse conditions, it was also tested against T. urticae immature, adults (males and females) and number of deposited eggs. There was a direct correlation for all stages between concentration, the percentage of immature and adult mortality, and the percentage of egg-hatching inhibition. In the case of the immature, 100% mortality was shown after 7 days of treatment. However, in the case of adult males and females, 100% mortality was shown after 3 days of treatment. Additionally, after 14 days from treatment, it entirely stopped egg depositing. The new formulation might be applied to manage the TSSM.

    Nonlocal Conductivity in the Vortex-Liquid Regime of a Two-Dimensional Superconductor

    We have simulated the time-dependent Ginzburg-Landau equation with thermal fluctuations, to study the nonlocal dc conductivity of a superconducting film. Having examined points in the phase diagram at a wide range of temperatures and fields below the mean-field upper critical field, we find a portion of the vortex-liquid regime in which the nonlocal ohmic conductivity in real space is negative over a distance several times the spacing between vortices. The effect is suppressed when driven beyond linear response. Earlier work had predicted the existence of such a regime, due to the high viscosity of a strongly-correlated vortex liquid. This behavior is clearly distinguishable from the monotonic spatial fall-off of the conductivity in the higher temperature or field regimes approaching the normal state. The possibilities for experimental study of the nonlocal transport properties are discussed.Comment: 18 pages, revtex, 6 postscript figure
