    Co-Production of Cellulase and Xylanase Enzymes By Thermophilic Bacillus subtilis 276NS

    Co-production of thermostable extracellular cellulase and xylanase was investigated using bacterial soil isolate. To evaluate the effect of culture conditions on the c-oproduction of both enzymes by Bacillus subtilis 276NS (GenBank accession number JF801740), a Plackett-Burman fractional factorial design was applied. Among the tested variables, yeast extract, sucrose and incubation time were the most significant variables increased cellulase and xylanase productions. Both of xylan and CMC (Carboxymethyl Cellulose) induced the xylanse enzyme production. A near-optimum medium formulation was obtained which increased the cellulase and xylanase enzymes 5.7- and 1.08 –fold higher than the yield obtained with the basal medium, respectively. Thereafter, the response surface methodology was adopted to acquire the best process conditions among the selected variables (xylan, CMC and Yeast Extract (YE)) required for improving xylanase yield. The optimal combinations of the major medium constituents for xylanase production evaluated using non-linear optimization algorithm of EXCEL-solver, was as follows (g/L): D-sucrose, 10; xylan, 10.367, CMC, 10.535; (NH4)2SO4, 1.0; YE, 1.71; Tween-80, 0.4 and FeSO4, 0.25 mg/L, at pH 8.0, temperature 35◦C and incubation time 24h under shaking. The predicted optimum thermostable xylanase activity was 360 U/ml, which was around 4-times the activity with the basal medium

    Integration of Adolescent Health Needs Into Primary Health Care Services: Designing Care Model

    Integration of the health needs of all adolescent into primary health services will package all services together which leads to better health overall. Aims: toconduct a situational analysis of adolescent health needs in Assiut governorate, assess the primary health care facilities anddevelop a design model to integrate adolescent health needs into primary health care settings and assess its effectiveness. Methodology: cross-sectional and quasi-experimental design was used.Sample size was 350 adolescents (175 study and 175 control group) attending two health facilities (urban and rural)which were selected randomly. Three questionnaires were used. Firstly, pre and posttest interview structured questionnaire was used which developed according to evidence –based questionnaires. Secondly, Health care providers’ questionnaire asked question in the same modules of adolescent’s questionnaire. Last one, questionnaireto assess PHC services’ quality as perceived by adolescents. Results: One third of adolescents had moderate level of anxiety. Half of them were suffering from various level of depression. About two thirds of them hadn’t stress.Violence attitude is lower in studied group. The knowledge scores of all modules (mental, reproductive, Nutritional health, smoking / addiction, accidents and violence)  were significantly better among adolescents in studied group than control group. The majority of health providers had high level of knowledge.There are high levels of satisfaction among adolescents of most PHC services. Conclusion: Adolescents' knowledge, habits and beliefs were mostly inadequate as regards all modules. After implementation of the program remarkable improvement were achieved in adolescents’ knowledge and practice. Keywords: Adolescent Health , Primary Services, Designing Care Mode

    Impact of Nutrition Education Program Based on Nutritional Assessment of Adolescent in Assiut -Egypt

    Background: Adolescence is a stage of rapid changing, adequate nutrition is vital for achieving full growth probable, and any disturbance will occur in this stage may lead to various health problems. Aims: to assess the nutritional status and food habits among adolescents and evaluate the impact of the educational program in primary health care sittings. Methodology: quasi-experimental research design was used. Sample size was 350 adolescents (175 intervention group and 175 control group) attending two health facilities (urban and rural) which were selected randomly. Pre and posttest (immediate and late) interview structured questionnaire was used which included socio-demographic data, anthropometric measurements, hemoglobin level, questions regarding knowledge about nutrition and food habits. Results: more than two fifths (44.4%) of the sample was anemic. The mean body mass index was 21.09±4.6. Obesity was significantly more prevalent in female (13.4%) than male (3.4%).The knowledge scores of nutritional health was significantly better among adolescents in studied group than control group (p value= 0.00). Also their eating and consumption habits had a slightly improvement. Conclusion: After implementation of the program the Adolescents' knowledge and habits had a remarkable improvement.     اربع معقدات قاعدة شف ثنائية النواة جديدة [(MCl2)2L] = M} Fe (1), (2) Co, Cu (3), Sn (4) { ليكاند = 1,4-ثنائي فنيلين (ميثان دايين) ثنائي(ايثان ثنائي امين). حضرت بطريقة التفاعل المباشر بين اليكاند وعدد من هاليدات الفلزات. الصيغ التركيبية للمركبات اثبتت من خلال عدد من التقنيات الطيفية مكيف الاشعة تحت الحمراء وطيف الرنين النووي المغناطيسي وطيف الكتلة. حددت الفعالية الحيوية للمركبات تجاه عدد من العزلات البكتيرية السالبة والموجبة الغرام مثل العصية القولونية, الزوائف, العصيات, المكورات العنقودية, المسبحات العنقودية.Four new binuclear Schiff base metal complexes [(MCl2)2L] {M = Fe 1, Co 2, Cu 3, Sn 4, L = N,N’-1,4-Phenylenebis (methanylylidene) bis (ethane-1,2-diamine)} have been synthesized using direct reaction between proligand (L) and the corresponding metal chloride (FeCl2, CoCl2, CuCl2 and SnCl2). The structures of the complexes have been conclusively determined by a set of spectroscopic techniques (FT-IR, 1H-NMR, and mass spectra). Finally, the biological properties of the complexes have been investigated with a comparative approach against different species of bacteria (E. coli G-, Pseudomonas G-, Bacillus G+, Staphylococcus G+, and Streptococcus G+)

    Effects of increasing levels of pea hulls in the diet on productive performance, development of the gastrointestinal tract, and nutrient retention of broilers from one to eighteeen days of age

    The effects of inclusion of pea hulls (PH) in the diet on growth performance, development of the gastrointestinal tract and nutrient retention were studied in broilers from 1 to 18d of age. There were a control diet based on low fibre ingredients (69.3 total dietary fibre (16.1g crude fibre/kg)) and three additional diets that resulted from the dilution of the basal diet with 25, 50 and 75g PH/kg (81.2, 93.2, and 105.1g total dietary fibre/kg diet, respectively). Each treatment was replicated six times and the experimental unit was a cage with 12 chicks. Growth performance, development of the gastrointestinal tract and the coefficients of total tract apparent retention (CTTAR) of nutrients were recorded at 6, 12 and 18d of age. In addition, jejunal morphology was measured at 12 and 18d and the coefficients of apparent ileal digestibility (CAID) of nutrients at 18d of age. Pea hulls inclusion affected all the parameters studied. The inclusion of 25 and 50g PH/kg diet improved growth performance as compared to the control diet. The relative weight (g/kg body weight) of proventriculus (P≤0.01), gizzard (P≤0.001) and ceca (P≤0.05) increased linearly as the level of PH in the diet increased. The inclusion of PH affected quadratically (P≤0.01) villus height:crypt depth ratio with the highest value shown at 25g PH/kg. In general, the CTTAR and CAID of nutrients increased linearly and quadratically (P≤0.05) with increasing levels of PH, showing maximum values with PH level between 25 and 50g/kg diet. We conclude that the size of the digestive organs increases with increasing levels of PH in the diet. In general, the best performance and nutrient digestibility values were observed with levels of PH within the range of 25 and 50g/kg. Therefore, young broilers have a requirement for a minimum amount of dietary fibre. When pea hulls are used as a source of fibre, the level of total dietary fibre required for optimal performance is within the range of 81.2–93.2g/kg diet (25.6–35.0g crude fibre/kg diet). An excess of total dietary fibre (above 93.2g/kg diet) might reduce nutrient digestibility and growth performance to values similar to those observed with the control diet

    EXPERIMENTAL EVIDENCES FOR THE PROMISING HERAPEUTIC ROLE OF VITIS VINIFERA SEED EXTRACT AGAINST NONALCOHOLIC STEATOHEPATITIS

    Objective: The present study was planned to investigate the possible therapeutic effect of methanolic extract of Vitis vinifera seeds on high fat diet-induced nonalcoholic steatohepatitis (NASH) in forty adult female Wistar rats.Methods: The animals were divided into four groups, (G1)was served as healthy control group and the other three groups received high fat diet for 32 weeks for induction of NASH were assigned as follow: (G2) in which the animals bearing NASH were left untreated, (G3) in which the animals bearing NASH were treated with Vitis vinifera seed extract in a dose of 0.28g/kg b. wt (GSH) and (G4) in which the animals bearing NASH were treated with Vitis vinifera seed extract in a dose of 0.14g/kg b. wt (GSL).Results: The results revealed significant increase in serum ALT activity, plasma glucose, insulin levels, serum resist in, NF-κB, TNF-α, HGF levels, hepatic TNF-α and HGF gene expression levels. While, serum albumin, adiponectin levels and hepatic adiponectin gene expression level were decreased significantly in NASH group. Conversely, treatment of NASH groups with GSH or GSL resulted in significant decrease in serum ALT activity, plasma glucose, insulin levels, serum resist in, NF-κB, TNF-α, HGF levels, hepatic TNF-α and HGF gene expression levels. However, serum albumin, adiponectin levels and hepatic adiponectin gene expression level were increased significantly as a consequence of treatment with GSH or GSL.Conclusions: The efficacy of Vitis vinifera extract against NASH might be attributed to its strong hepatoprotective potential and powerful anti-inflammatory activity in addition to its potent role in ameliorating insulin resistance indices.Â

    This paper presents an experimental study on the effect of shot peening onmechanical properties and residual stresses of 2024-T351 Aluminum alloy. Under theeffects of shot peening time SPT the results show that the existence of SPT canimprove the mechanical properties and fatigue life up to a limit value of SPT. The 15minutes SPT gave the highest value of (σu, σy) which is about 6.7 % for (σu) and 11.7% for (σy). Empirical equations were proposed to evaluate the SPT with theendurance limit stress and the residual stresses

    Nanoparticle Catalyzed Hydrodesulfurization of Diesel Fuel in a Trickle Bed Reactor: Experimental and Optimization Study

    This work focuses on the preparation, simulation, and optimization of the hydrodesulfurization (HDS) of dibenzothiophene (DBT) using a nanocatalyst. A homemade nanocatalyst (3 percent Co, 10 percent Mo/γ-Al2O3 nanoparticles) was used in a trickle bed reactor (TBR). The HDS kinetic model was estimated based on experimental observations over ranges of operating conditions to evaluate kinetic parameters of the HDS process and apply the key parameters. Based on these parameters, the performance of the TBR catalyzed by the nanocatalyst was evaluated and scaled up to a commercial scale. Also, the selectivity of HDS reactions was also modeled to achieve the highest yield of the desired hydrogenation product based on the desirable route of HDS. A comprehensive modeling and simulation of the HDS process in a TBR was developed and the output results were compared with experimental results. The comparison showed that the simulated and experimental data of the HDS process match well with a standard error of up to 5%. The best reaction kinetic variables obtained from the HDS pilot-plant (specific reaction rate expression, rate law, and selectivity) TBR have been utilized to develop an industrial scale HDS of DBT.     Production of Cold-Active Lipase by Free and Immobilized Marine Bacillus cereus HSS: Application in Wastewater Treatment

    Lipases are enzymes that have the potential to hydrolyze triacylglycerol to free fatty acids and glycerol and have various applications. The aim of the present study was to isolate and screen marine bacteria for lipase production, optimize the production, and treat wastewater. A total of 20 marine bacterial isolates were obtained from the Mediterranean Sea and were screened for lipase production. All isolates were found to have lipolytic ability. The differences between the isolates were studied using RAPD-PCR. The most promising lipase producer (isolate 3) that exhibited the highest lipolytic hydrolysis (20 mm) was identified as Bacillus cereus HSS using 16S rDNA analysis and had the accession number MF581790. Optimization of lipase production was carried out using the Plackett–Burman experimental design with cotton seed oil as the inducer under shaking conditions at 10°C. The most significant factors that affected lipase production were FeSO4, KCl, and oil concentrations. By using the optimized culture conditions, the lipase activity increased by 1.8-fold compared with basal conditions. Immobilization by adsorption of cells on sponge and recycling raised lipase activity by 2.8-fold compared with free cells. The repeated reuse of the immobilized B. cereus HSS maintained reasonable lipase activity. A trial for the economic treatment of oily wastewater was carried out. Removal efficiencies of biological oxygen demand, total suspended solids, and oil and grease were 87.63, 90, and 94.7%, respectively, which is promising for future applications

    ANTIVIRAL AND IMMUNE STIMULANT ACTIVITIES OF GLYCYRRHIZIN AGAINST DUCK HEPATITIS VIRUS

    This study was conducted to investigate the effect of glycyrrhizin as an immune stimulant against duck hepatitis virus (DHV). In vitro study was carried out to determine cytotoxic and antiviral effects of glycyrrhizin in VERO cells. In vivo study was performed on 40 one-day-old White Pekin ducklings. –and the birds weres divided into 4 groups: control, glycyrrhizin treated, vaccinated with live attenuated DHV vaccine and glycyrrhizin treated and vaccinated; to investigate the changes in immunity and challenge test. Blood samples were collected from each duckling for evaluation of cellular and humeral immunity. The in vitro results revealed that glycyrrhizin had antiviral and no toxic effects till 106 dilutions. Higher antibody titer was observed from the 5th week till the end of experiment in glycyrrhizin and vaccinated group. Treatment with glycyrrhizin alone or with DHV vaccine demonstrated a pronounced lymphocytic proliferation response after 4 days post-inoculation till the end of experiment, while vaccinated group revealed a pronounced proliferation response after 24 days post-inoculation. Treatment with glycyrrhizin alone or combination with DHV vaccine revealed good immune stimulant and antiviral effect against DHV
