    Caffeine exposure induces browning features in adipose tissue in vitro and in vivo

    Brown adipose tissue (BAT) is able to rapidly generate heat and metabolise macronutrients, such as glucose and lipids, through activation of mitochondrial uncoupling protein 1 (UCP1). Diet can modulate UCP1 function but the capacity of individual nutrients to promote the abundance and activity of UCP1 is not well established. Caffeine consumption has been associated with loss of body weight and increased energy expenditure, but whether it can activate UCP1 is unknown. This study examined the effect of caffeine on BAT thermogenesis in vitro and in vivo. Stem cell-derived adipocytes exposed to caffeine (1 mM) showed increased UCP1 protein abundance and cell metabolism with enhanced oxygen consumption and proton leak. These functional responses were associated with browning-like structural changes in mitochondrial and lipid droplet content. Caffeine also increased peroxisome proliferator-activated receptor gamma coactivator 1-alpha expression and mitochondrial biogenesis, together with a number of BAT selective and beige gene markers. In vivo, drinking coffee (but not water) stimulated the temperature of the supraclavicular region, which co-locates to the main region of BAT in adult humans, and is indicative of thermogenesis. Taken together, these results demonstrate that caffeine can promote BAT function at thermoneutrality and may have the potential to be used therapeutically in adult humans

    Communicative competence and institutional affiliation: interactional processes of identity construction by immigrant students in Catalonia

    The growing presence of children of immigrant families in the public school system in the bilingual region of Catalonia provides us with an opportunity to study how young multilingual and multicultural speakers construct their social competencies and their identity within the specific context of a gate-keeping social institution such as the school. The study reported in this paper approaches language learning as a process of socialisation that involves not only learning how to make sense of linguistic signs but also learning how to enact different social roles in particular institutions. The analysis focuses on the interactional profiles of two immigrant students in two types of communicative activities that are representative of the school context: responding to questions from an adult and cooperating with a peer in the resolution of a learning task. By shifting the focus of analysis from a decontextualised notion of communicative competence to the notion of 'institutionally affiliated communicative competence' and concentrating on issues such as the (1) the relationship between knowledge and participation, (2) language choice inside and outside school and (3) definitions of correctness in language use, the study reveals how the two students construct a highly 'affiliated' identit

It is shown that the nondecaying mode of perturbation has a finite value even in this case and that its amplitude is given by the standard formula with the time derivation of the scalar field replaced by the potential gradient using the slow-roll equation

    Occurrence of an isolate of maize stripe virus on sorghum in India

    A disease characterised by chlorotic stripes and bands, named sorghum stripe disease (SStD), was observed on sorghum in India with an incidence of less than 0.5% to nearly 10%. The affected plants were dwarfed and had poor or no panicle formation. This disease could be transmitted by the delphacid planthopper Peregrinus maidis to sorghum but not to Brachiaria eruciformis; Cenchrus ciliaris; Chloris barbata; Dichantium annulatum; Dichantium aristatum; Digitaria ciliaris: Dinebra retroflexa; Echinocloa colona; Eleusine coracana; Pennisetum glaucum; Pennisetum violaceum; Setaria pallida Fusca; Triticum aestivum and Zea mays. Sorghum stripe disease was shown to be caused by a tenuivirus serologically related to maize stripe virus (MStV). Virus particles were filamentous, less than 10 nm in width. The purified virus preparation contained only one polypeptide of 34 500 D. Eight species of nucleic acids, four ssRNA of 1.21, 0.87, 0.73, 0.47 ± 106D and four dsRNA of 2.43, 1.69, 1.40, 0.71 ± 106D, were extracted from purified virus preparations. When the four dsRNA were denatured, they migrated along with the four ssRNA species indicating that dsRNA contained duplex RNA of same molecular weight as the four ssRN A. In enzyme-linked immunosorbent assay and in electro-blot immunoassay it was evident that MStV-Sorg was serologically more closely related to the MStV isolates from Florida, Reunion and Venezuela than to a RStV isolate from Japan. The virus was named MStV-Sorg to distinguish it from MStV which readily infects maize. This is the first report of occurrence of a tenuivirus in the Indian subcontinent

    Factors Influencing Dietetic Interns\u27 Dietary Habits During Supervised Practice

    Supervised practice is a prerequisite to becoming a registered dietitian. Research suggests that environmental and social factors may affect dietary choices. This focus group research aimed to gather opinions from dietetic interns to understand what factors related to supervised practice, if any, affected their dietary habits. Qualitative data were collected via seven recorded virtual focus groups in which trained moderators facilitated a discussion using a series of controlled questions. Participants, dietetic interns (n = 42) who were currently completing or had completed their supervised practice within the previous six months, attended one of seven virtual focus groups. Each focus group had five to eight participants. Transcripts were separately coded by two trained researchers using a grounded theory approach to identify themes and subthemes. Researchers discussed any disagreements in coding and established a consensus. Elements related to the dietetic internship were observed to influence participants’ dietary choices. Main themes included time, finances, food access and availability, physical and mental effects, non-supervised practice factors, and social factors. Dietetic programs and preceptors should explore ways to raise interns’ awareness and minimize the potential negative impacts of these factors on interns’ dietary habits to improve their overall internship experience

    Immersed boundary-finite element model of fluid-structure interaction in the aortic root

    It has long been recognized that aortic root elasticity helps to ensure efficient aortic valve closure, but our understanding of the functional importance of the elasticity and geometry of the aortic root continues to evolve as increasingly detailed in vivo imaging data become available. Herein, we describe fluid-structure interaction models of the aortic root, including the aortic valve leaflets, the sinuses of Valsalva, the aortic annulus, and the sinotubular junction, that employ a version of Peskin's immersed boundary (IB) method with a finite element (FE) description of the structural elasticity. We develop both an idealized model of the root with three-fold symmetry of the aortic sinuses and valve leaflets, and a more realistic model that accounts for the differences in the sizes of the left, right, and noncoronary sinuses and corresponding valve cusps. As in earlier work, we use fiber-based models of the valve leaflets, but this study extends earlier IB models of the aortic root by employing incompressible hyperelastic models of the mechanics of the sinuses and ascending aorta using a constitutive law fit to experimental data from human aortic root tissue. In vivo pressure loading is accounted for by a backwards displacement method that determines the unloaded configurations of the root models. Our models yield realistic cardiac output at physiological pressures, with low transvalvular pressure differences during forward flow, minimal regurgitation during valve closure, and realistic pressure loads when the valve is closed during diastole. Further, results from high-resolution computations demonstrate that IB models of the aortic valve are able to produce essentially grid-converged dynamics at practical grid spacings for the high-Reynolds number flows of the aortic root

    Sequential design of computer experiments for the estimation of a probability of failure

    This paper deals with the problem of estimating the volume of the excursion set of a function f:Rd→Rf:\mathbb{R}^d \to \mathbb{R} above a given threshold, under a probability measure on Rd\mathbb{R}^d that is assumed to be known. In the industrial world, this corresponds to the problem of estimating a probability of failure of a system. When only an expensive-to-simulate model of the system is available, the budget for simulations is usually severely limited and therefore classical Monte Carlo methods ought to be avoided. One of the main contributions of this article is to derive SUR (stepwise uncertainty reduction) strategies from a Bayesian-theoretic formulation of the problem of estimating a probability of failure. These sequential strategies use a Gaussian process model of ff and aim at performing evaluations of ff as efficiently as possible to infer the value of the probability of failure. We compare these strategies to other strategies also based on a Gaussian process model for estimating a probability of failure.Comment: This is an author-generated postprint version.
