Nanosheet thickness-modulated MoS2 dielectric property evidenced by field-effect transistor performance
We report on the nanosheet-thickness effects on the performance of top-gate MoS2 field-effect transistors (FETs), which is directly related to the MoS2 dielectric constant. Our top-gate nanosheet FETs with 40 nm thin Al2O3 displayed at least an order of magnitude higher mobility than those of bottom-gate nanosheet FETs with 285 nm thick SiO2, benefiting from the dielectric screening by high-k Al2O3. Among the top-gate devices, the single-layered FET demonstrated the highest mobility of similar to 170 cm(2) V-1 s(-1) with 90 mV dec(-1) as the smallest subthreshold swing (SS) but the double-and triple-layered FETs showed only similar to 25 and similar to 15 cm(2) V-1 s(-1) respectively with the large SS of 0.5 and 1.1 V dec(-1). Such property degradation with MoS2 thickness is attributed to its dielectric constant increase, which could rather reduce the benefits from the top-gate high-k dielectric.open115353Nsciescopu

Predictive impact of rare genomic copy number variations in siblings of individuals with autism spectrum disorders.
Identification of genetic biomarkers associated with autism spectrum disorders (ASDs) could improve recurrence prediction for families with a child with ASD. Here, we describe clinical microarray findings for 253 longitudinally phenotyped ASD families from the Baby Siblings Research Consortium (BSRC), encompassing 288 infant siblings. By age 3, 103 siblings (35.8%) were diagnosed with ASD and 54 (18.8%) were developing atypically. Thirteen siblings have copy number variants (CNVs) involving ASD-relevant genes: 6 with ASD, 5 atypically developing, and 2 typically developing. Within these families, an ASD-related CNV in a sibling has a positive predictive value (PPV) for ASD or atypical development of 0.83; the Simons Simplex Collection of ASD families shows similar PPVs. Polygenic risk analyses suggest that common genetic variants may also contribute to ASD. CNV findings would have been pre-symptomatically predictive of ASD or atypical development in 11 (7%) of the 157 BSRC siblings who were eventually diagnosed clinically
Genotoxicity evaluation of the insecticide ethion in root of Allium cepa L.
In this study, the genotoxic effects of ethion were investigated in the mitotic cell division of Allium cepa. Primary roots of A. cepa were treated with various concentrations (25, 50, 75, and 100%) of ethion solutions for different duration of time. The result revealed that increase in the concentration and duration of treatment decreases the mitotic indices. 24 h treatment at 100% concentration of ethion induced lowest mitotic index (20.08%) than that of the control (36.37%). The percentage of chromosomal abnormalities in different mitotic stages was significantly generally higher than that of the control in all the treatment period and concentrations. These abnormalities appeared in various degrees depending on the treatment duration and concentrations of ethion. The abnormalities in dividing cell reached a maximum value of 11.30% after 12 h of treatment at 75% concentration. The type of abnormalitiesproduced were scattered prophase, non-synchronized condensation of chromosome, disturbed prophase, equatorial plate shifting, sticky chromosomes, C-metaphase and sticky metaphase. Overall, it can be concluded that ethion has a potential genotoxic effects on mitotic divisions in A. cepa root tip cells. So, it will be necessary to test the mutagenic potential of ethion on a more intensive and extensive basis especially on non-target systems before it is recommended for wider use in agriculturalfield
The Evolution of X-ray Clusters of Galaxies
Considerable progress has been made over the last decade in the study of the
evolutionary trends of the population of galaxy clusters in the Universe. In
this review we focus on observations in the X-ray band. X-ray surveys with the
ROSAT satellite, supplemented by follow-up studies with ASCA and Beppo-SAX,
have allowed an assessment of the evolution of the space density of clusters
out to z~1, and the evolution of the physical properties of the intra-cluster
medium out to z~0.5. With the advent of Chandra and Newton-XMM, and their
unprecedented sensitivity and angular resolution, these studies have been
extended beyond redshift unity and have revealed the complexity of the
thermodynamical structure of clusters. The properties of the intra-cluster gas
are significantly affected by non-gravitational processes including star
formation and Active Galactic Nucleus (AGN) activity. Convincing evidence has
emerged for modest evolution of both the bulk of the X-ray cluster population
and their thermodynamical properties since redshift unity. Such an
observational scenario is consistent with hierarchical models of structure
formation in a flat low density universe with Omega_m=0.3 and sigma_8=0.7-0.8
for the normalization of the power spectrum. Basic methodologies for
construction of X-ray-selected cluster samples are reviewed and implications of

Rapid generation of endogenously driven transcriptional reporters in cells through CRISPR/Cas9
CRISPR/Cas9 technologies have been employed for genome editing to achieve gene knockouts and knock-ins in somatic cells. Similarly, certain endogenous genes have been tagged with fluorescent proteins. Often, the detection of tagged proteins requires high expression and sophisticated tools such as confocal microscopy and flow cytometry. Therefore, a simple, sensitive and robust transcriptional reporter system driven by endogenous promoter for studies into transcriptional regulation is desirable. We report a CRISPR/Cas9-based methodology for rapidly integrating a firefly luciferase gene in somatic cells under the control of endogenous promoter, using the TGFΞ²-responsive gene PAI-1. Our strategy employed a polycistronic cassette containing a non-fused GFP protein to ensure the detection of transgene delivery and rapid isolation of positive clones. We demonstrate that firefly luciferase cDNA can be efficiently delivered downstream of the promoter of the TGFΞ²-responsive gene PAI-1. Using chemical and genetic regulators of TGFΞ² signalling, we show that it mimics the transcriptional regulation of endogenous PAI-1 expression. Our unique approach has the potential to expedite studies on transcription of any gene in the context of its native chromatin landscape in somatic cells, allowing for robust high-throughput chemical and genetic screens
Dose-dependent effects of Allopurinol on human foreskin fibroblast cell and human umbilical vein endothelial cell under hypoxia
Allopurinol, an inhibitor of xanthine oxidase, has been used in clinical trials of patients with cardiovascular and chronic kidney disease. These are two pathologies with extensive links to hypoxia and activation of the transcription factor hypoxia inducible factor (HIF) family. Here we analysed the effects of allopurinol treatment in two different cellular models, and their response to hypoxia. We explored the dose-dependent effect of allopurinol on Human Foreskin Fibroblasts (HFF) and Human Umbilical Vein Endothelial Cells (HUVEC) under hypoxia and normoxia. Under normoxia and hypoxia, high dose allopurinol reduced the accumulation of HIF-1Ξ± protein in HFF and HUVEC cells. Allopurinol had only marginal effects on HIF-1Ξ± mRNA level in both cellular systems. Interestingly, allopurinol effects over the HIF system were independent of prolyl-hydroxylase activity. Finally, allopurinol treatment reduced angiogenesis traits in HUVEC cells in an in vitro model. Taken together these results indicate that high doses of allopurinol inhibits the HIF system and pro-angiogenic traits in cells
Characterisation of mechanical and thermal properties in flax fabric reinforced geopolymer composites
This paper presents the mechanical and thermal properties of flax fabric reinforced fly ash based geopolymer composites. Geopolymer composites reinforced with 2.4, 3.0 and 4.1 wt% woven flax fabric in various layers were fabricated using a hand lay-up technique and tested for mechanical properties such as flexural strength, flexural modulus, compressive strength, hardness, and fracture toughness. All mechanical properties were improved by increasing the flax fibre contents, and showed superior mechanical properties over a pure geopolymer matrix. Fourier transform infrared spectroscopy (FTIR) and scanning electron microscopy (SEM) studies were carried out to evaluate the composition and fracture surfaces of geopolymer and geopolymer/flax composites. The thermal behaviour of composites was studied by thermogravimetric analysis (TGA) and the results showed significant degradation of flax fibres at 300 Β°C

Sex-Specific Differences in Shoaling Affect Parasite Transmission in Guppies
Background: Individuals have to trade-off the costs and benefits of group membership during shoaling behaviour. Shoaling can increase the risk of parasite transmission, but this cost has rarely been quantified experimentally. Guppies (Poecilia reticulata) are a model system for behavioural studies, and they are commonly infected by gyrodactylid parasites, notorious fish pathogens that are directly transmitted between guppy hosts.
Methodology/Principal Findings:Parasite transmission in single sex shoals of male and female guppies were observed using an experimental infection of Gyrodactylus turnbulli. Parasite transmission was affected by sex-specific differences in host behaviour, and significantly more parasites were transmitted when fish had more frequent and more prolonged contact with each other. Females shoaled significantly more than males and had a four times higher risk to contract an infection.
Conclusions/Significance: Intersexual differences in host behaviours such as shoaling are driven by differences in natural and sexual selection experienced by both sexes. Here we show that the potential benefits of an increased shoaling tendency are traded off against increased risks of contracting an infectious parasite in a group-living species
Spondin-2 (SPON2), a More Prostate-Cancer-Specific Diagnostic Biomarker
BACKGROUND: Prostate-specific antigen (PSA) screening, although common, has recently been called into question. To find prostate cancer (PCa) diagnostic biomarkers that can make up for the defects of PSA, we compared the secretomes of several benign and PCa cell lines, selected candidate molecules, and then confirmed their clinical value. METHODOLOGY/PRINCIPAL FINDINGS: We first identified extracellular proteins by two-dimensional gel electrophoresis (2-DE) coupled with liquid chromatography-tandem mass spectrometry (LC-MS/MS) identification. We then validated the secreted proteins on a cellular level, and finally determined whether they could be used as PCa diagnostic biomarkers using prostate tissue and serum specimens of Chinese volunteers by immunohistostaining and sandwich ELISA. We obtained credible extracellular protein 2-DE graphs of prostate cell lines. The 5 spots that showed superior repeatability were selected for LC-MS/MS analysis, which identified seven candidate molecules. One of the candidate molecules, spondin-2 (SPON2), was only expressed in the conditioned media (CM) of androgen receptor (AR) positive PCa cell lines. Using tissue microarray by immunohistostaining, we found SPON2 to be over-expressed in PCa. SPON2 staining was more intense in Gleason score sum 7-8 and in PCa patients with metastasis. By receiver operator characteristic (ROC) curve analysis, we found that the serum SPON2 level was elevated in PCa patients, showing sensitivity and specificity suitable for diagnostic use. We also found that SPON2 could be used to identify PCa patients with serum PSA levels no higher than 10 ng/ml from healthy elderly men. CONCLUSION/SIGNIFICANCE: SPON2 is a new serum and histological diagnostic biomarker for PCa. It can avoid some of the problems of PSA testing and was here found to offer relatively high sensitivity and specificity relative to PSA
