Chemokines: key players in innate and adaptive immunity.

    Get PDF
Healthy individuals initiate an immediate immune response to microbes by using a set of germline-encoded receptors that recognize common molecular patterns found on the surface of pathogens that are distinct from self-antigens. This innate immune response is the first line of defense against microorganisms in vertebrates, and constitutes the only immune response in plants and invertebrates. The innate immune system includes cellular components, as well as a host of soluble products (antimicrobial peptides, complement fragments, cytokines, and chemokines). The adaptive immune response, which provides long-lasting protection, takes days to develop and requires somatic mutations leading to the development of antigen-specific T cell receptors (cell-mediated immunity) and immunoglobulins (humoral immunity). Members of the chemokine superfamily are crucially involved in both innate and adaptive responses. We review the biological actions of the chemokine superfamily, focusing on several functions that are relevant for both immune responses, such as cell recruitment, microbicidal activity, cell activation, polarization of CD4+ T cells, and effects on structural cells. In particular, we will illustrate the central role that chemokines play in host defense, best demonstrated by the tremendous number of chemokine and chemokine receptor homologs found in microbial genomes, which deflect the immune response of the host

Design and simulation of a solar assisted desiccant-based air handling unit

Paper presented at the 8th International Conference on Heat Transfer, Fluid Mechanics and Thermodynamics, Mauritius, 11-13 July, 2011.Desiccant-based Air Handling Units (AHU) can guarantee significant technical and energy/environmental advantages related to the use of traditional ones (with dehumidification by cooling). For these reasons, a test facility has been located in Benevento (Southern Italy), in which a silica-gel desiccant wheel is inserted in an AHU which treats outside air only. For this wheel, the regeneration temperature can be as low as 65 °C, therefore energy savings and emissions reductions are more consistent when the regeneration of the desiccant material is obtained by means of available low grade thermal energy, such as that from solar collectors or cogenerators. In the actual configuration, regeneration is obtained by means of thermal energy recovered from a micro-cogenerator (MCHP, Micro Combined Heat and Power) based on a natural gas-fired reciprocating internal combustion engine, eventually integrated through a natural gas-fired boiler. Future activity aims to reduce the regeneration fossil energy requirements by introducing a solar collector system that substitutes or integrates thermal energy supplied by the MHCP. To this aim, a commercial software has been used to design the solar collector system (collectors type, absorber area, water flow rate…) considering the thermal power and temperature requirements of the regeneration process. The existing AHU and the designed solar collector system have been successively simulated by means of TRNSYS software, in order to evaluate operational data and performance parameters of the system in a typical week of operation, e.g. thermal-hygrometric conditions of air in the mean sections of the AHU, solar collectors efficiency and solar fraction.mp201

39 hints to facilitate the use of semantics for data on agriculture and nutrition

    Get PDF
In this paper, we report on the outputs and adoption of the Agrisemantics Working Group of the Research Data Alliance (RDA), consisting of a set of recommendations to facilitate the adoption of semantic technologies and methods for the purpose of data interoperability in the field of agriculture and nutrition. From 2016 to 2019, the group gathered researchers and practitioners at the crossing point between information technology and agricultural science, to study all aspects in the life cycle of semantic resources: Conceptualization, edition, sharing, standardization, services, alignment, long term support. First, the working group realized a landscape study, a study of the uses of semantics in agrifood, then collected use cases for the exploitation of semantics resources – a generic term to encompass vocabularies, terminologies, thesauri, ontologies. The resulting requirements were synthesized into 39 “hints” for users and developers of semantic resources, and providers of semantic resource services. We believe adopting these recommendations will engage agrifood sciences in a necessary transition to leverage data production, sharing and reuse and the adoption of the FAIR data principles. The paper includes examples of adoption of those requirements, and a discussion of their contribution to the field of data science

Health-related quality of life and mental health problems after a disaster: Are chronically ill survivors more vulnerable to health problems?

    Get PDF
Studies have shown that the chronically ill are at higher risk for reduced health-related quality of life (HRQL) and for mental health problems. A combination with traumatic events might increase this risk. This longitudinal study among 1216 survivors of a disaster examines whether chronically ill survivors had a different course of HRQL and mental health problems compared to survivors without chronic diseases. HRQL and mental health problems were measured 3 weeks, 18 months and 4 years post-disaster. Data on pre-disaster chronic diseases was obtained from the electronic medical records of general practitioners. Random coefficient analyses showed significant interaction effects for social functioning, bodily pain and emotional role limitations at T2 only. Chronically ill survivors did not consistently have a different course of general health, physical role limitations, and mental health problems. In conclusion, chronic diseases were not an important risk factor for impaired HRQL and mental health problems among survivors

Towards VocBench 3: Pushing collaborative development of thesauri and ontologies further beyond

    Get PDF
More than three years have passed since the release of the second edition of VocBench, an open source collaborative web platform for the development of thesauri complying with Semantic Web standards. In these years, a vibrant user community has gathered around the system, consisting of public organizations, companies and independent users looking for open source solutions for maintaining their thesauri, code lists and authority resources. The focus on collaboration, the differentiation of user roles and the workflow management for content validation and publication have been the strengths of the platform, especially for those organizations requiring a centralized and controlled publication environment. Now the time has come to widen the scope of the platform: funded by the ISA2programme of the European Commission, VocBench 3 will offer a general-purpose collaborative environment for development of any kind of RDF dataset, improving the editing capabilities of its predecessor, while still maintaining the peculiar aspects that determined its success. In this paper, we review the requirements and the new objectives set for version 3, and then introduce the new characteristics that were implemented for this next iteration of the platform

SIRT1 activity in peripheral blood mononuclear cells correlates with altered lung function in patients with chronic obstructive pulmonary disease

    Get PDF
Background. Oxidative stress is a recognized pathogenic mechanism in chronic obstructive pulmonary disease (COPD). Expression of the NAD+-dependent deacetylase Sirtuin 1 (SIRT1), an antiaging molecule with a key role in oxidative stress response, has been described as decreased in the lung of COPD patients. No studies so far investigated whether systemic SIRT1 activity was associated to decreased lung function in this disease. Methods. We measured SIRT1 protein expression and activity in peripheral blood mononuclear cells (PBMCs) and total oxidative status (TOS), total antioxidant capacity (TEAC), and oxidative stress index (TOS/TEAC) in the plasma of 25 COPD patients, 20 healthy nonsmokers (HnS), and 20 healthy smokers (HS). Results. The activity of SIRT1 was significantly lower in COPD patients compared to both control groups while protein expression decreased progressively (HnS > HS > COPD). TOS levels were significantly lower in HnS than in smoke-associated subjects (COPD and HS), while TEAC levels were progressively lower according (HnS > HS > COPD). In COPD patients, SIRT1 activity, but not protein levels, correlated significantly with both lung function parameters (FEV1/FVC and FEV1) and TEAC. Conclusions. These findings suggest loss of SIRT1-driven antioxidant activity as relevant in COPD pathogenesis and identify SIRT1 activity as a potential convenient biomarker for identification of mild/moderate, stable COPD

Anti-EGFR Therapy in Metastatic Small Bowel Adenocarcinoma: Myth or Reality?

    Get PDF
Background: Due to the relative rarity of small bowel adenocarcinoma (SBA), prospective trials, helping to guide therapeutic decisions, are lacking and the optimal therapy for advanced SBA is unknown. The role of targeted agents, such as anti\u2013epidermal growth factor receptor (EGFR) and anti\u2013vascular endothelial growth factor (VEGF), is unknown. Patients and Methods: This is a retrospective multicenter observational study that included patients with metastatic SBA treated with anti-EGFR antibodies (cetuximab or panitumumab) \ub1 chemotherapy in the first (I) or second (II) line. Results: Thirteen patients with metastatic SBA, recruited from 5 Italian referral institutions, were included in the present retrospective analysis. All patients received anti-EGFR inhibitors as a single agent or in association with chemotherapy. More common G2 treatment\u2013related side effects were skin reaction (8 patients, 53.8%), hypomagnesemia (6 patients, 46.2%), and diarrhea (8 patients, 61.5%). Grade 3 diarrhea was observed in only 1 patient. Conjunctivitis was not reported in any patients. Grade 4 toxicity was not reported. In the overall population, median progression-free survival was 5.526 months (95% confidence interval [CI]: 3.684-12.467). Median overall survival was 15.86 months (95% CI: 14.43-24.30). Complete response was observed in 15% of patients, partial response in 39% of patients, stable disease in 23% of patients, and progression disease in 15% of patients. Conclusions: In this retrospective analysis, anti-EGFR inhibitors showed to be a suitable addendum to chemotherapy in the I and II line, with an excellent tolerance and safety profile both in I and II line
