    A versatile interface model for thermal conduction phenomena and its numerical implementation by XFEM

    A general interface model is presented for thermal conduction and characterized by two jump relations. The first one expresses that the temperature jump across an interface is proportional to the interfacial average of the normal heat flux while the second one states that the normal heat flux jump is proportional to the surface Laplacian of the interfacial average of the temperature. By varying the two scalar proportionality parameters, not only the Kapitza resistance and highly conducting interface models can be retrieved but also all the intermediate cases can be covered. The general interface model is numerically implemented by constructing its weak form and by using the level-set method and XFEM. The resulting numerical procedure, whose accuracy and robustness are thoroughly tested and discussed with the help of a benchmark problem, is shown to be efficient for solving the problem of thermal conduction in particulate composites with various imperfect interfaces

    A mutation in the LAMC2 gene causes the Herlitz junctional epidermolysis bullosa (H-JEB) in two French draft horse breeds

    Epidermolysis bullosa (EB) is a heterogeneous group of inherited diseases characterised by skin blistering and fragility. In humans, one of the most severe forms of EB known as Herlitz-junctional EB (H-JEB), is caused by mutations in the laminin 5 genes. EB has been described in several species, like cattle, sheep, dogs, cats and horses where the mutation, a cytosine insertion in exon 10 of the LAMC2 gene, was very recently identified in Belgian horses as the mutation responsible for JEB. In this study, the same mutation was found to be totally associated with the JEB phenotype in two French draft horse breeds, Trait Breton and Trait Comtois. This result provides breeders a molecular test to better manage their breeding strategies by genetic counselling

    Holocene range collapse of giant muntjacs and pseudo-endemism in the Annamite large mammal fauna

    Aim: To clarify the postglacial biogeography of the Annamite and eastern Chinese ungulate faunas, and determine whether current understanding of Asian mammalian biogeography is biased by pseudo-extinctions and pseudo-endemism associated with a historical extinction filter. Location: Modern-day specimens of giant muntjac (Muntiacus vuquangensis) from the Annamite Mountains of Laos and Vietnam were compared with zooarchaeological specimens of extinct giant muntjac (M. gigas) from eastern China, and with a reference sample of northern red muntjac (M. vaginalis) from China, Southeast Asia and South Asia. Methods: We analyzed a dataset of antler measurements using MANOVAs, PCAs and scaling relationships, to quantify morphometric variation between extinct and living giant muntjacs in relation to variation shown by a different sympatric large-bodied muntjac species. We also attempted ancient biomolecule analysis of Holocene samples from China. Results: Whereas the combined giant muntjac sample can be differentiated from the reference red muntjac sample in all of our multivariate morphometric analyses, no significant differences are shown between extinct and living giant muntjacs using any analyses, matching the pattern seen when comparing conspecific red muntjac samples from across the same geographic region. Main conclusions: We find no support for recognizing extinct and living giant muntjacs as distinct taxa, and postglacial populations from China and the Annamites should probably all be referred to M. gigas. The likely conspecificity of giant muntjacs across Eastern and Southeast Asia demonstrates that current-day Asian mammalian biogeography has been shaped by an extinction filter and challenges the idea that the Annamite region represents a cradle of evolution; instead, it may represent a refuge of diversity for some taxa, preserving remnant pseudo-endemic populations of species that have been extirpated across other parts of their former ranges

    Aspirin and extended-release dipyridamole versus clopidogrel for recurrent stroke

    Background Recurrent stroke is a frequent, disabling event after ischemic stroke. This study compared the efficacy and safety of two antiplatelet regimens — aspirin plus extendedrelease dipyridamole (ASA–ERDP) versus clopidogrel. Methods In this double-blind, 2-by-2 factorial trial, we randomly assigned patients to receive 25 mg of aspirin plus 200 mg of extended-release dipyridamole twice daily or to receive 75 mg of clopidogrel daily. The primary outcome was first recurrence of stroke. The secondary outcome was a composite of stroke, myocardial infarction, or death from vascular causes. Sequential statistical testing of noninferiority (margin of 1.075), followed by superiority testing, was planned. Results A total of 20,332 patients were followed for a mean of 2.5 years. Recurrent stroke occurred in 916 patients (9.0%) receiving ASA–ERDP and in 898 patients (8.8%) receiving clopidogrel (hazard ratio, 1.01; 95% confidence interval [CI], 0.92 to 1.11). The secondary outcome occurred in 1333 patients (13.1%) in each group (hazard ratio for ASA–ERDP, 0.99; 95% CI, 0.92 to 1.07). There were more major hemorrhagic events among ASA–ERDP recipients (419 [4.1%]) than among clopidogrel recipients (365 [3.6%]) (hazard ratio, 1.15; 95% CI, 1.00 to 1.32), including intracranial hemorrhage (hazard ratio, 1.42; 95% CI, 1.11 to 1.83). The net risk of recurrent stroke or major hemorrhagic event was similar in the two groups (1194 ASA–ERDP recipients [11.7%], vs. 1156 clopidogrel recipients [11.4%]; hazard ratio, 1.03; 95% CI, 0.95 to 1.11). Conclusions The trial did not meet the predefined criteria for noninferiority but showed similar rates of recurrent stroke with ASA–ERDP and with clopidogrel. There is no evidence that either of the two treatments was superior to the other in the prevention of recurrent stroke. (ClinicalTrials.gov number, NCT00153062.

    Screening of Bioactive Compounds from Moutan Cortex and Their Anti-Inflammatory Activities in Rat Synoviocytes

    Moutan Cortex, a widely used traditional Chinese medicine for the treatment of various diseases, is the root bark of Paeonia suffruticosa Andrews (Paeoniaceae). Most of the pharmacological investigations of Moutan Cortex have been addressed to its central nervous system activities, anti-oxidative and sedative actions. Otherwise, there are few reports about the active compounds with anti-inflammatory activity of Moutan Cortex. The aim of the present study was to screen and identify bioactive compounds with anti-inflammatory effect from Moutan Cortex. With the aid of preparative high performance liquid chromatography (HPLC) technique, ethyl acetate and ethanol extract of Moutan Cortex were isolated into twenty-two fractions. Bioactivities of these fractions were evaluated by measuring expression of tumor necrosis factor-α (TNF-α) in rat synoviocytes subjected to interleukin-1β (IL-1β). Eight compounds were isolated from six active fractions and identified by HPLC/MSn. Purified compounds, paeoniflorin, paeonol and pentagalloylglucose resulted in dose-dependent inhibition of TNF-α synthesis and IL-6 production in synoviocytes treated with proinflammatory mediator. These results suggested that paeonol, paeoniflorin, glycosides and pentagalloylglucose contribute to the anti-inflammatory effect of Moutan Cortex

    Complement dependent cytotoxicity activity of therapeutic antibody fragments is acquired by immunogenic glycan coupling

    Abstract Oligosaccharides are implicated in the development of the immune response notably in complement activation. Anti-tumoural immunotherapy by monoclonal antibodies (mAbs) offers some advantages to chemotherapy including cell targeting but some of them are inefficient to generate cytotoxicity dependent complement (CDC) known to be important in the antibody\u2019s efficacy. The aim of this study is to give a CDC activity of mAb by linkage of a complement activating oligosaccharide to this antibody via a hetero-bifunctional linker allowing control of the conjugation reaction. We worked on non Hodgkin Burkitt\u2019s lymphoma as cancer source, Fab fragments of rituximab devoid of complement activity as mAb and the trisaccharide Gal\u3b1(1\u21923)Gal\u3b2(1\u21924)GlcNAc as immunogenic glycan. The bioconjugate Fab-Gal was characterized by biochemical methods and we demonstrated that the \u3b1-Gal epitope was recognized by seric immunoglobulins. After checking the recognition capacity of the Fab- Gal conjugate for the CD20 epitope, in vitro assays were performed to evaluate the activation of the complement cascade by the Fab-Gal conjugate. The effect of this bioconjugate was confirmed by the evaluation of the proliferation response of Burkitt\u2019s cell line. The relative facility realization of this strategy represents new approaches to increase activities of mAbs
